Project description:Cell lipid membranes are the primary site of irreversible injury during freezing/thawing and cryopreservation of cells, but the underlying causes remain unknown. Here, we probe the effect of cooling from 20 °C to 0 °C on the structure and mechanical properties of 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) bilayers using atomic force microscopy (AFM) imaging and AFM-based nanoindentation in a liquid environment. The Young's modulus of elasticity (E) at each temperature for DPPC was obtained at different ionic strengths. Both at 20 mM and 150 mM NaCl, E of DPPC bilayers increases exponentially -as expected-as the temperature is lowered between 20 °C and 5 °C, but at 0 °C E drops from the values measured at 5 °C. Our results support the hypothesis that mechanical weakening of the bilayer at 0 °C is produced by structural changes at the lipid-fluid interface.

Project description:Silicon carbide is desirable for many nuclear applications, making it necessary to understand how it deforms after irradiation. Ion implantation combined with nanoindentation is commonly used to measure radiation-induced changes to mechanical properties; hardness and modulus can be calculated from load–displacement curves, and fracture toughness can be estimated from surface crack lengths. Further insight into indentation deformation and fracture is required to understand the observed changes to mechanical properties caused by irradiation. This paper investigates indentation deformation using high-resolution electron backscatter diffraction (HR-EBSD) and Raman spectroscopy. Significant differences exist after irradiation: fracture is suppressed by swelling-induced compressive residual stresses, and the plastically deformed region extends further from the indentation. During focused ion beam cross-sectioning, indentation cracks grow, and residual stresses are modified. The results clarify the mechanisms responsible for the modification of apparent hardness and apparent indentation toughness values caused by the compressive residual stresses in ion-implanted specimens. Project description:Nanoscale mechanical property measurements of nanoporous nanosheets face many challenges. Herein we show atomic force microscope (AFM)-based nanoindentation to probe the nanoscale mechanical properties of a 2-D metal-organic framework (MOF) nanosheet material containing atomic-sized pores, termed CuBDC [copper 1,4-benzenedicarboxylate]. The sample thickness ranged from ∼10 nm (tens of monolayers) up to ∼400 nm (a stack of multilayers). In terms of its elastic-plastic properties, the Young's modulus (E ∼ 23 GPa) and yield strength (σ y ∼ 450 MPa) were determined in the through-thickness direction. Moreover, we characterized the failure mechanisms of the CuBDC nanosheets, where three failure mechanisms were identified: interfacial slippage, fracture of the framework, and delamination of multilayered nanosheets. Threshold forces and indentation depths corresponding to these failure modes were determined. To gain insights into the failure mechanisms, we employ finite-element models with cohesive elements to simulate the interfacial debonding of a stack of 2-D nanosheets during the indentation process. The nanomechanical AFM methodology elucidated here will pave the way for the study of other 2-D hybrid nanosheets and layered van der Waals solids.

Project description:We performed in situ indentation in a transmission electron microscope on Al-TiN multilayers with individual layer thicknesses of 50 nm, 5 nm and 2.7 nm to explore the effect of length scales on the plastic co-deformability of a metal and a ceramic. At 50 nm, plasticity was confined to the Al layers with easy initiation of cracks in the TiN layers. At 5 nm and below, cracking in TiN was suppressed and post mortem measurements indicated a reduction in layer thickness in both layers. The results demonstrate the profound size effect in enhancing plastic co-deformability in nanoscale metal-ceramic multilayers.

Project description:This paper addresses the general problem of deformable linear object manipulation. The main application we consider is in the field of agriculture, for plant grasping, but may have interests in other tasks such as human daily activities and industrial production. We specifically consider an elastic linear object where one of its endpoints is fixed, and another point can be grasped by a robotic arm. To deal with the mentioned problem, we propose a model-free method to control the state of an arbitrary point that can be at any place along the object's length. Our approach allows the robot to manipulate the object without knowing any model parameters or offline information of the object's deformation. An adaptive control strategy is proposed for regulating the state of any point automatically deforming the object into the desired location. A control law is developed to regulate the object's shape thanks to the adaptive estimation of the system parameters and its states. This method can track a desired manipulation trajectory to reach the target point, which leads to a smooth deformation without drastic changes. A Lyapunov-based argument is presented for the asymptotic convergence of the system that shows the process's stability and convergence to desired state values. To validate the controller, numerical simulations involving two different deformation models are conducted, and performances of the proposed algorithm are investigated through full-scale experiments.

Project description:Micropatterned cocultures are a useful experimental tool for the study of cell-cell interactions. Patterning methods often rely on sequential seeding of different cell types or removal of a barrier separating two populations, but it is difficult to pattern sharp interfaces between pure populations with low cross-contamination when using these approaches. Patterning by the use of reconfigurable substrates can overcome these limitations, but such methods can be costly and challenging to employ in a typical biology laboratory. Here, we describe a low-cost and simple-to-use reconfigurable substrate comprised of a transparent elastic material that is partially cut to form a slit that opens when the device is stretched. The slit seals back up when released, allowing two initially separate, adherent cell populations to be brought together to form a contact interface. Fluorescent imaging of patterned cocultures demonstrates the early establishment of a sharp cellular interface. As a proof of principle, we demonstrate the use of this device to study competition at the interface of two stem cell populations.

Project description:A new class of deformation is presented for a planar loop structure made up of slender elastic bodies and joints. In demonstrating the circumferential shortening of the multi-jointed elastic loop, diverse three-dimensional (3D) deformations emerge through piecewise deflections and discrete rotations. These 3D morphologies correspond to conformations of molecular ring systems. Through image processing, the 3D reconstructions of the deformed structures are characterized by number, geometry, and initial imperfections of the body segments. We elucidate from measurements that the conformational deformation without self-stress results from a cyclical assembly of compressive bending of elastic bodies with high shear rigidity. The mechanical insights gained may apply in controlling the polymorphism exhibited by the cyclical structures across scales.

Project description:Although ZnSe has been widely studied due to its attractive electronic and optoelectronic properties, limited data on its plastic deformations are available. Through molecular dynamics simulations, we have investigated the indentations on the (001), (110), and (111) planes of ZnSe nano films. Our results indicate that the elastic modulus, incipient plasticity, elastic recovery ratio, and the structural evolutions during the indenting process of ZnSe nano films show obvious anisotropy. To analyze the correlation of structural evolution and mechanical responses, the atomic displacement vectors, atomic arrangements, and the dislocations of the indented samples are analyzed. Our simulations revealed that the plastic deformations of the indented ZnSe nano films are dominated by the nucleation and propagation of 1/2<110> type dislocations, and the symmetrically distributed prismatic loops emitted during the indenting process are closely related with the mechanical properties. By studying the evolutions of microstructures, the formation process of the dislocations, as well as the formation mechanisms of the emitted prismatic loops under the indented crystalline planes are discussed. The results presented in this work not only provide an answer for the questions about indentation responses of ZnSe nano films, but also offer insight into its plastic deformation mechanisms.

Project description:Cyclic elastoplastic deformation behaviors of austenite phase and ferrite phase in a duplex stainless steel were investigate by load-controlled cyclic nanoindentation with a Berkovich indenter. During the tests, the maximum penetration depth per cycle increased rapidly with cycle number at transient state, and reached stable at quasi-steady state. Plastic dissipated energy was quantitatively proved to be the driving force for the propagation of deformation zones during cyclic nanoindentation tests. Transmission electron microscopy combined with FIB was used to reveal the deformation mechanisms of both phases underneath indents with cycles. After quasi-static single loading, nucleation and concentration of dislocations were observed in both austenite phase and ferrite phase under the indenter. After cyclic loading, dislocations propagated to further regions in both phases. Besides, slip bands were generated within single nanoindentation and propagated during the subsequent cyclic nanoindentation. The sizes of the deformation regions for both phases under the indents after cyclic indentation observed by TEM were consistent with those calculated by the expansion model of spherical cavity.

Project description:The tensile behavior of a Ni60Nb40 metallic glass (MG) has been studied by using ab initio density functional theory (DFT) calculation with a large cell containing 1024 atoms (614 Ni and 410 Nb). We provide insight into how a super elastic limit can be achieved in a MG. Spatially inhomogeneous responses of single atoms and also major polyhedra are found to change greatly with increasing external stress when the strain is over 2%, causing the intrinsically viscoelastic behavior. We uncover the origin of the observed super elastic strain limit under tension (including linear and viscoelastic strains) in small-sized MG samples, mainly caused by inhomogeneous distribution of excess volumes in the form of newly formed subatomic cavities.
