Pulsation of electrified jet in capillary microfluidics.

ABSTRACT: In this work, we investigate the pulsation of an electrically charged jet surrounded by an immiscible dielectric liquid in flow-focusing capillary microfluidics. We have characterized a low-frequency large-amplitude pulsation and a high-frequency small-amplitude pulsation, respectively. The former, due to the unbalanced charge and fluid transportation is responsible for generating droplets with a broad size distribution. The latter is intrinsic and produces droplets with a relatively narrow size distribution. Moreover, the average size of the final droplets can be tuned via the intrinsic pulsating frequency through changing the diameter of the emitted liquid jet. Our results provide degree of control over the emulsion droplets with submicron sizes generated in microfluidic-electrospray platform.

Coupling Capillary-Driven Microfluidics with Lateral Flow Immunoassay for Signal Enhancement.

Project description:Microfluidics has emerged as a versatile technology that is applied to enhance the performance of analytical techniques, among others. Pursuing this, we present a capillary-driven microfluidic device that improves the sensitivity of lateral flow immunoassay rapid tests thanks to offering an automated washing step. A novel multilevel microfluidic chip was 3D-printed with a photocurable black resin, sealed by an optically clear pressure-sensitive adhesive, and linked to the lateral flow strip. To depict the efficacy of microfluidics and the washing step, cortisol was measured quantitatively within the proposed device. Measuring cortisol levels is a way to capture physiological stress responses. Among biofluids, saliva is less infectious and easier to sample than others. However, higher sensitivity is demanded because the salivary cortisol concentrations are much lower than in blood. We carried out a competitive lateral flow immunoassay protocol with the difference that the microfluidic device applies an automated washing step after the sample is drained downstream. It washes the trapped quantum-dot-labeled antibodies out from nitrocellulose, diminishing background noise as these are bonded to cortisols and not to the immobilized receptors. Fluorescence spectroscopy, as a high-precision analysis, was successfully applied to determine clinically relevant salivary cortisol concentrations within a buffer quantitatively. The microfluidic design relied on a 3D valve that avoids reagent cross-contamination. This cross-contamination could make the washing buffer impure and undesirably dilute the sample. The proposed device is cost-effective, self-powered, robust, and ideal for non-expert users.

Flow characterization and patch clamp dose responses using jet microfluidics in a tubeless microfluidic device.

Project description:BackgroundSurface tension passive pumping is a way to actuate flow without the need for pumps, tubing or valves by using the pressure inside small drop to move liquid via a microfluidic channel. These types of tubeless devices have typically been used in cell biology. Herein we present the use of tubeless devices as a fluid exchange platform for patch clamp electrophysiology.New methodInertia from high-speed droplets and jets is used to create flow and perform on-the-fly mixing of solutions. These are then flowed over GABA transfected HEK cells under patch in order to perform a dose response analysis.ResultsTIRF imaging and electrical recordings are used to study the fluid exchange properties of the microfluidic device, resulting in 0-90% fluid exchange times of hundreds of milliseconds. COMSOL is used to model flow and fluid exchange within the device. Patch-clamping experiments show the ability to use high-speed passive pumping and its derivatives for studying peak dose responses, but not for studying ion channel kinetics.Comparison with existing method(s)Our system results in fluid exchange times slower than when using a standard 12-barrel application system and is not as stable as traditional methods, but it offers a new platform with added functionality.ConclusionsSurface tension passive pumping and tubeless devices can be used in a limited fashion for electrophysiology. Users may obtain peak dose responses but the system, in its current form, is not capable of fluid exchange fast enough to study the kinetics of most ion channels.

Electro-actuated valves and self-vented channels enable programmable flow control and monitoring in capillary-driven microfluidics.

Project description:Microfluidics are essential for many lab-on-a-chip applications, but it is still challenging to implement a portable and programmable device that can perform an assay protocol autonomously when used by a person with minimal training. Here, we present a versatile concept toward this goal by realizing programmable liquid circuits where liquids in capillary-driven microfluidic channels can be controlled and monitored from a smartphone to perform various advanced tasks of liquid manipulation. We achieve this by combining electro-actuated valves (e-gates) with passive capillary valves and self-vented channels. We demonstrate the concept by implementing a 5-mm-diameter microfluidic clock, a chip to control four liquids using 100 e-gates with electronic feedback, and designs to deliver and merge multiple liquids sequentially or in parallel in any order and combination. This concept is scalable, compatible with high-throughput manufacturing, and can be adopted in many microfluidics-based assays that would benefit from precise and easy handling of liquids.

Benefit of pulsation in soft corals.

Project description:Soft corals of the family Xeniidae exhibit a unique, rhythmic pulsation of their tentacles (Movie S1), first noted by Lamarck nearly 200 y ago. However, the adaptive benefit of this perpetual, energetically costly motion is poorly understood. Using in situ underwater particle image velocimetry, we found that the pulsation motions thrust water upward and enhance mixing across the coral-water boundary layer. The induced upward motion effectively prevents refiltration of water by neighboring polyps, while the intensification of mixing, together with the upward flow, greatly enhances the coral's photosynthesis. A series of controlled laboratory experiments with the common xeniid coral Heteroxenia fuscescens showed that the net photosynthesis rate during pulsation was up to an order of magnitude higher than during the coral's resting, nonpulsating state. This enhancement diminished when the concentration of oxygen in the ambient water was artificially raised, indicating that the enhancement of photosynthesis was due to a greater efflux of oxygen from the coral tissues. By lowering the internal oxygen concentration, pulsation alleviates the problem of reduced affinity of ribulose-1,5-bisphosphate carboxylase oxygenase (RuBisCO) to CO2 under conditions of high oxygen concentrations. The photosynthesis-respiration ratio of the pulsating H. fuscescens was markedly higher than the ratios reported for nonpulsating soft and stony corals. Although pulsation is commonly used for locomotion and filtration in marine mobile animals, its occurrence in sessile (bottom-attached) species is limited to members of the ancient phylum Cnidaria, where it is used to accelerate water and enhance physiological processes.

Models of cylindrical bubble pulsation.

Project description:Three models are considered for describing the dynamics of a pulsating cylindrical bubble. A linear solution is derived for a cylindrical bubble in an infinite compressible liquid. The solution accounts for losses due to viscosity, heat conduction, and acoustic radiation. It reveals that radiation is the dominant loss mechanism, and that it is 22 times greater than for a spherical bubble of the same radius. The predicted resonance frequency provides a basis of comparison for limiting forms of other models. The second model considered is a commonly used equation in Rayleigh-Plesset form that requires an incompressible liquid to be finite in extent in order for bubble pulsation to occur. The radial extent of the liquid becomes a fitting parameter, and it is found that considerably different values of the parameter are required for modeling inertial motion versus acoustical oscillations. The third model was developed by V. K. Kedrinskii [Hydrodynamics of Explosion (Springer, New York, 2005), pp. 23-26] in the form of the Gilmore equation for compressible liquids of infinite extent. While the correct resonance frequency and loss factor are not recovered from this model in the linear approximation, it provides reasonable agreement with observations of inertial motion.

Pulsation-limited oxygen diffusion in the tumour microenvironment.

Project description:Hypoxia is central to tumour evolution, growth, invasion and metastasis. Mathematical models of hypoxia based on reaction-diffusion equations provide seemingly incomplete descriptions as they fail to predict the measured oxygen concentrations in the tumour microenvironment. In an attempt to explain the discrepancies, we consider both the inhomogeneous distribution of oxygen-consuming cells in solid tumours and the dynamics of blood flow in the tumour microcirculation. We find that the low-frequency oscillations play an important role in the establishment of tumour hypoxia. The oscillations interact with consumption to inhibit oxygen diffusion in the microenvironment. This suggests that alpha-blockers-a class of drugs used to treat hypertension and stress disorders, and known to lower or even abolish low-frequency oscillations of arterial blood flow -may act as adjuvant drugs in the radiotherapy of solid tumours by enhancing the oxygen effect.

Quantification of Intracranial Aneurysm Volume Pulsation with 7T MRI.

Project description:BACKGROUND AND PURPOSE:Aneurysm volume pulsation is a potential predictor of intracranial aneurysm rupture. We evaluated whether 7T MR imaging can quantify aneurysm volume pulsation. MATERIALS AND METHODS:In Stage I of the study, 10 unruptured aneurysms in 9 patients were studied using a high-resolution (0.6-mm, isotropic) 3D gradient-echo sequence with cardiac gating. Semiautomatic segmentation was used to measure aneurysm volume (in cubic millimeters) per cardiac phase. Aneurysm pulsation was defined as the relative increase in volume between the phase with the smallest volume and the phase with the largest volume. The accuracy and precision of the measured volume pulsations were addressed by digital phantom simulations and a repeat image analysis. In Stage II, the imaging protocol was optimized and 9 patients with 9 aneurysms were studied with and without administration of a contrast agent. RESULTS:The mean aneurysm pulsation in Stage I was 8% ± 7% (range, 2%-27%), with a mean volume change of 15 ± 14 mm3 (range, 3-51 mm3). The mean difference in volume change for the repeat image analysis was 2 ± 6 mm3. The artifactual volume pulsations measured with the digital phantom simulations were of the same magnitude as the volume pulsations observed in the patient data, even after protocol optimization in Stage II. CONCLUSIONS:Volume pulsation quantification with the current imaging protocol on 7T MR imaging is not accurate due to multiple imaging artifacts. Future studies should always include aneurysm-specific accuracy analysis.

Pulsation of anastomotic vortex veins in pachychoroid spectrum diseases.

Project description:Accumulating evidence points to pachychoroid possibly being caused by vortex vein congestion which results in remodeling of choroidal drainage routes via intervortex vein anastomosis. This hypothesis prompted us to investigate vortex vein hemodynamics by studying videos of indocyanine green angiography (ICGA) in a retrospective case series of 295 eyes with pachychoroid spectrum diseases. In the early phase of the video-ICGA, pulsatile vortex venous flow was observed in 76 eyes (25.8%) at the vortex veins connected with anastomosis between superior and inferior vortex veins. The patients with pulsatile vortex venous flow were significantly older than those without pulsatile vortex venous flow (67.8 ± 13.2 vs. 63.9 ± 14.5 years, P < 0.05). Pulsatile vortex venous flow was 1.84 times more common in the inferior quadrants than in the superior quadrants. Interestingly, 14 of 76 eyes (18.4%) with pulsatile vortex venous flow showed retrograde pulsatile blood flow in the vortex veins. This retrograde pulsatile blood flow was 2.50 times more common in the inferior than in the superior quadrants. These findings indicate altered vortex vein hemodynamics due to vortex vein congestion in pachychoroid spectrum diseases.

Photopyroelectric microfluidics.

Project description:Precision manipulation of various liquids is essential in many fields. Unlike solid objects, fluids are intrinsically divisible, enriching their fundamental operations with merging, dispensing, and splitting on top of moving. Fluids are sticky as well, calling for their lossless manipulation to prevent mass loss and contamination. We present photopyroelectric microfluidics that meet all the requirements. In response to the irradiation from even one single beam of light, our platform creates a unique wavy dielectrophoretic force field that is remarkably capable of performing desired loss-free (loss being 0.5% of existing one) manipulation of droplets of surface tension from 18.9 to 98.0 mN m-1 and volume from 1 nl to 1000 μl, functioning as a "magic" wetting-proof hand to navigate, fuse, pinch, and cleave fluids on demand, enabling cargo carriers with droplet wheels and upgrading the limit of maximum concentration of deliverable protein by 4000-fold.
