NREG: old banger or new vehicle for research?

ABSTRACT: The Northern Region Endoscopy Group (NREG) is a collaborative endoscopy network. NREG provides the opportunity for clinicians across the entire Northern region to undertake high quality, clinically meaningful, health service based research. NREG research is undertaken across a wide range of endoscopy units with pooling of resources and ideas. NREG has presented a large number of abstracts and is contributing to a number of National Institute for Health Research trials. NREG is collaborating with other research teams in the UK and a number of research grants have been secured. NREG aims to make a significant contribution to the field of endoscopic research over the coming years.

35 Years of TFAM Research: Old Protein, New Puzzles.

Project description:Transcription Factor A Mitochondrial (TFAM), through its contributions to mtDNA maintenance and expression, is essential for cellular bioenergetics and, therefore, for the very survival of cells. Thirty-five years of research on TFAM structure and function generated a considerable body of experimental evidence, some of which remains to be fully reconciled. Recent advancements allowed an unprecedented glimpse into the structure of TFAM complexed with promoter DNA and TFAM within the open promoter complexes. These novel insights, however, raise new questions about the function of this remarkable protein. In our review, we compile the available literature on TFAM structure and function and provide some critical analysis of the available data.

Caenorhabditis elegans in anthelmintic research - Old model, new perspectives.

Project description:For more than four decades, the free-living nematode Caenorhabditis elegans has been extensively used in anthelmintic research. Classic genetic screens and heterologous expression in the C. elegans model enormously contributed to the identification and characterization of molecular targets of all major anthelmintic drug classes. Although these findings provided substantial insights into common anthelmintic mechanisms, a breakthrough in the treatment and control of parasitic nematodes is still not in sight. Instead, we are facing increasing evidence that the enormous diversity within the phylum Nematoda cannot be recapitulated by any single free-living or parasitic species and the development of novel broad-spectrum anthelmintics is not be a simple goal. In the present review, we summarize certain milestones and challenges of the C. elegans model with focus on drug target identification, anthelmintic drug discovery and identification of resistance mechanisms. Furthermore, we present new perspectives and strategies on how current progress in C. elegans research will support future anthelmintic research.

Rural and urban perspectives on growing old: developing a new research agenda.

Project description:Urban and rural themes have played an important part in European gerontological research. This paper analyses current issues in the field of urban and rural studies as applied to understanding old age. Both dimensions are being affected by population movements of different kinds, driven to a significant degree by globalisation in its various forms. The paper summarises trends underpinning rural and urban living and evidence regarding the impact of change in these areas on daily life in old age. The article considers a number of research agendas which would advance rural and urban studies of ageing, these combining developments in geography and urban studies with those in critical gerontology. The paper argues that a revitalised rural and urban gerontology will bring forward major new themes and issues for social gerontology in the 21st century.

Complete genome sequence of Escherichia coli C - old model organism with a new application in biofilm research

Project description:Escherichia coli strain C is the last of five E. coli strains (C, K12, B, W, Crooks) designated as safe for laboratory purposes whose genome has not been sequenced. We found that E. coli C forms more robust biofilms than the other four laboratory strains. Here we present the complete genomic sequence of this strain in which we utilized high resolution optical mapping to confirm a large inversion in comparison to other strains. DNA sequence comparison revealed the absence of several genes involved in biofilm formation, such as antigen 43, waaSBOJYZUL for LPS synthesis, and cpsB for curli synthesis. The main difference affecting biofilm formation is the presence of an IS3-like insertion sequence in front of the carbon storage regulator csrA gene. This insertion is located 86 bp upstream of the csrA start codon inside the -35 region of P4 promoter and blocks the transcription from the sigma32 and sigma70 promoters P1-P3 located further upstream. Analysis of gene expression profiles in planktonic and biofilm attached cells by the RNAseq method allows better understanding of this regulatory pathway in E. coli.

Bridging animal and clinical research during SARS-CoV-2 pandemic: A new-old challenge.

Project description:Many milestones in medical history rest on animal modeling of human diseases. The SARS-CoV-2 pandemic has evoked a tremendous investigative effort primarily centered on clinical studies. However, several animal SARS-CoV-2/COVID-19 models have been developed and pre-clinical findings aimed at supporting clinical evidence rapidly emerge. In this review, we characterize the existing animal models exposing their relevance and limitations as well as outline their utility in COVID-19 drug and vaccine development. Concurrently, we summarize the status of clinical trial research and discuss the novel tactics utilized in the largest multi-center trials aiming to accelerate generation of reliable results that may subsequently shape COVID-19 clinical treatment practices. We also highlight areas of improvement for animal studies in order to elevate their translational utility. In pandemics, to optimize the use of strained resources in a short time-frame, optimizing and strengthening the synergy between the preclinical and clinical domains is pivotal.

Research on the spatial association network structure for innovation efficiency of China's new energy vehicle industry and its influencing factors.

Project description:It is of great significance to study the spatial network of the new energy vehicle (NEV) industry innovation efficiency and its factors to promote the rational allocation of innovative resources and the coordinated development of Chinese NEV industry. First, the Super Efficiency Data Envelope Analysis model is used to measure innovation efficiency in the NEV industry in Chinese provinces, and based on the results, the improved gravity model is applied to construct a spatial correlation network. Then, by applying social network analysis (SNA) to study NEV industry development node spatial correlations, we conclude that there is no overall hierarchical structure. The SNA are applied to examine spatial correlations with respect to NEV industry innovation efficiency in each province, and to analyze the role and position of each province in the spatial correlation network. Finally, the influencing factors of spatial correlation of the innovation efficiency of China's NEV industry has been discussed. The result shows that the difference in spatial distance and R&D investment has a significant impact on the spatial correlation of the NEV industry.

An empirical study on new-energy vehicle users' preferences of in-vehicle interaction input methods.

Project description:New energy sources are transforming the automotive market. This shift has also expanded the possibilities for in-vehicle interaction. Through a literature review, this study categorizes the in-vehicle interaction activities into three types: driving tasks, comfort tasks, and entertainment tasks. This study conducted empirical survey of 377 users to understand their preferences of in-vehicle interaction input methods inside new energy vehicles. The results show that gender, educational level, income, driver's license type and driving experience have significant influence on the perception and preference of the in-vehicle interaction input methods. However, age and experience with new energy vehicle didn't show significant results. The findings of this study can assist manufacturers in developing targeted solutions and meeting the personalized needs of users in future vehicle market segments.

Chess databases as a research vehicle in psychology: Modeling large data.

Project description:The game of chess has often been used for psychological investigations, particularly in cognitive science. The clear-cut rules and well-defined environment of chess provide a model for investigations of basic cognitive processes, such as perception, memory, and problem solving, while the precise rating system for the measurement of skill has enabled investigations of individual differences and expertise-related effects. In the present study, we focus on another appealing feature of chess-namely, the large archive databases associated with the game. The German national chess database presented in this study represents a fruitful ground for the investigation of multiple longitudinal research questions, since it collects the data of over 130,000 players and spans over 25 years. The German chess database collects the data of all players, including hobby players, and all tournaments played. This results in a rich and complete collection of the skill, age, and activity of the whole population of chess players in Germany. The database therefore complements the commonly used expertise approach in cognitive science by opening up new possibilities for the investigation of multiple factors that underlie expertise and skill acquisition. Since large datasets are not common in psychology, their introduction also raises the question of optimal and efficient statistical analysis. We offer the database for download and illustrate how it can be used by providing concrete examples and a step-by-step tutorial using different statistical analyses on a range of topics, including skill development over the lifetime, birth cohort effects, effects of activity and inactivity on skill, and gender differences.

Hematopoietic-cell transplantation for lymphoma in the era of genetically engineered cellular therapy: it's not quite time to scrap the old vehicle for the new car.

Project description:PURPOSE OF REVIEW:Second-line platinum-based salvage chemotherapy followed by high-dose chemotherapy and autologous hematopoietic-cell transplantation (AHCT) has remained the standard of care (SOC) for relapsed and primary refractory (r/r) diffuse large B-cell lymphoma (DLBCL) for greater than 2 decades. In the postrituximab era, this strategy has yielded disappointing outcomes for r/r patients with curability in less one-quarter of the patients by intention-to-treat. RECENT FINDINGS:Given the Food and Drug Administration (FDA) approval of chimeric antigen receptor (CAR) modified T cells directed against CD19 (CD19 CAR T) for DLBCL following two lines of therapy and/or failed AHCT, encouragement with this therapy in the second line for r/r patients has naturally prompted randomized phase III studies against the aforementioned SOC. The predominant hurdle to procession to AHCT is chemotherapy sensitivity after platinum-based salvage therapy. SUMMARY:In this review, we will discuss recent investigations to improve response rates in r/r DLBCL with the intent of proceeding to potentially curative AHCT, as well as investigations to decrease progression post-AHCT. In addition, data regarding currently FDA approved CD19 CAR T cells will be reviewed. Within 2-3 years, we will know if the multicenter/multinational studies of CD19 CAR T will replace SOC salvage therapy and AHCT in the second-line. The role of allogeneic HCT will also be briefly reviewed in the context of these therapies.
