Project description:Laser-induced graphene (LIG) is an emerging technique for producing few-layer graphene or graphene-like material that has recently received increasing attention, due to its unique advantages. Subsequently, a variety of lasers and materials have been used to fabricate LIG using this technique. However, there is a lack of understanding of how different lasers (wavelengths) perform differently in the LIG conversion process. In this study, the produced LIG on polyimide (PI) under a locally water-cooled condition using a 10.6 μm CO2 infrared laser and a 355 nm ultraviolet (UV) laser are compared. The experimental investigations reveal that under the same UV and CO2 laser fluence, the ablation of PI show different results. Surface morphologies with micron-sized and nanometer pores were formed by the UV laser under different laser fluences, whereas micron-sized pores and sheet structure with fewer pores were produced by the CO2 laser. Energy dispersive spectrometry and three-dimensional topography characterization indicate that the photochemical effects were also involved in the LIG conversion with UV laser irradiation. It is also observed through experiments that the photothermal effect contributed to the formation of LIG under both lasers, and the LIG formed on PI substrates by the CO2 laser showed better quality and fewer layers.

Project description:Alumina toughened zirconia (ATZ) and zirconia toughened alumina (ZTA) are currently the materials of choice to meet the need for tough, strong, and bioinert ceramics for medical devices. However, the mechanical properties of ZrO2/Al2O3 dispersion ceramics could be considerably increased by reducing the corresponding grain sizes and by improving the homogeneity of the phase dispersion. Here, we prepare nanoparticles with an intraparticular phase distribution of Zr(1-x)Al(x)O(2-x/2) and (γ-, δ-)Al2O3 by the simultaneous gas phase condensation of laser co-vaporized zirconia and alumina raw powders. During subsequent spark plasma sintering the zirconia defect structures and transition alumina phases transform to a homogeneously distributed dispersion of tetragonal ZrO2 (52.4 vol%) and α-Al2O3 (47.6 vol%). Ceramics sintered by spark plasma sintering are completely dense with average grain sizes in the range around 250 nm. Outstanding mechanical properties (flexural strength σf = 1500 MPa, fracture toughness KIc = 6.8 MPa m(1/2)) together with a high resistance against low temperature degradation make these materials promising candidates for next generation bioceramics in total hip replacements and for dental implants.

Here, we report the case of a tracheal fistula in a patient after esophageal surgery suggested by pneumothorax and confirmed by fiberoptic bronchoscopy, and introduce lung ultrasound as a new bedside tool to sensitively detect pneumothorax.

Project description:Purpose:We wished to present the clinical applications and to evaluate the benefits of the use of a carbon dioxide (CO2) laser versus the conventional procedure for circumcision in adults, in terms of duration of surgery, surgical techniques, complications, pain and cosmetic appearance. Patients and Methods:The medical records of 482 patients who had been circumcised were retrospectively evaluated. The patients were divided into two groups: 168 patients (Group A) were circumcised with traditional techniques; and 314 patients (Group B) were circumcised using a CO2 laser. All the patients were circumcised under local anesthesia. Pain was evaluated using a verbal numerical rating scale for pain assessment. Postoperative wound swelling, bleeding, infection and pain were assessed at 4 hours, 24 hours and 7 days after surgery. Results:There were no significant differences between the two groups in terms of bleeding and infections. The difference in operating times between the groups was significant (p<0.001). Wound disruptions occurred in one patient in Group A at 3 days and two patients in Group B at 1 week. Pain scores were low and there was less pain in Group B than in Group A during the first 4 hours (1.8 vs 3.7; p<0.002). Compared with the conventional method, the CO2 laser technique was associated with much less pain at both 1 day (p<0.002) and 7 days (p<0.001) postoperatively. The cosmetic results were superior in Group B; a linear surgical scar developed in 94.9% of patients in Group B versus 61.3% in Group A (p<0.001). Conclusion:Our results show that the use of a CO2 laser was associated with a shorter operative time, less wound irritation and better cosmetic appearance compared with standard surgical techniques for circumcision.

Project description:Material suitability needs to be considered for the 3D printing of solid oral dosage forms (SODFs). This work aims to assess the suitability of a CO2 laser (λ = 10.6 μm) for selective laser sintering of SODFs containing copovidone and paracetamol. First, physicochemical characterization of powders (two grades of copovidone, two grades of paracetamol and their mixtures at various proportions) was conducted: particle size distribution, morphology, infrared absorbance, flowability, and compactness. Then, printing was launched, and printability of the powders was linked to their physicochemical characteristics. The properties of the sintered SODFs were evaluated (solid state, general aspect, porosity, hardness, drug content and release). Hence, it was found that as copovidone absorbs at the laser's wavelength, sintering was feasible without using an absorbance enhancer. Also, flowability, which mainly depends on the particle size, represents the first control line for "sinterability" as a fair flow is at least required. Low compactness of copovidone and mixtures reduces the mechanical properties of the SODFs but also increases porosity, which can modulate drug release. Moreover, the drug did not undergo degradation and demonstrated a plasticizer effect by lowering the heating temperature. In conclusion, this work proves the applicability of CO2 laser SLS printer to produce SODFs.

Project description:Genetically modified mice susceptible to atherosclerosis are widely used in atherosclerosis research. Although the atherosclerotic lesions in these animals show similarities to those in humans, comprehensive expression profile analysis of these lesions is limited by their very small size. In this communication, we have developed an approach to analyze global gene expression in mouse lesions by a combination of (a) laser capture microdissection (LCM) to isolate RNA from specific lesions, and (b) an efficient RNA amplification method that reliably yields sufficient quantities of high quality cRNA for quantitative real-time PCR (qPCR), as well as for microarray analysis. The RNA passed multiple quality controls and the expression profile observed in lesional cells compared with the whole artery encompasses genes that are characteristic of a macrophage/foam cell population. We believe that this method represents a useful new tool for the unbiased analysis of global gene expression of specific sub-regions in atherosclerotic lesions in different rodent models.

Project description:How developmental programs reactivate in regeneration is a fundamental question in biology. We addressed this question through the study of Wound Induced Hair follicle Neogenesis (WIHN), an adult organogenesis model where stem cells regenerate entirely new hair follicles de novo following deep wounding. The exact mechanism is uncertain. Here we show that self-noncoding dsRNA activates the anti-viral receptor TLR3 to induce intrinsic retinoic acid (RA) synthesis in a gradient that predicts new hair follicle formation after wounding in mice. Additionally, in humans, rejuvenation lasers induce gene expression signatures for dsRNA and RA, with measurable increases in intrinsic RA synthesis. These results demonstrate a novel stimulus for retinoic acid synthesis by non-coding dsRNA, relevant to their broad functions in development and immunity. The goal of this study is to investigate whole genome analysis in human skin with CO2 laser treatment, which leads to enhanced skin rejuvenation. Under Hopkins IRB (IRB00028768), 17 Caucasian women Fitzpatrick types I-III with average age of 55 and moderate to severe baseline photoaging were enrolled. Laser treatments and biopsy schedule were as listed, with treatments occurring both to the arm, and arm biopsies used for gene expression data listed.

Project description:Fractional CO2 laser, as a typical ablative laser, has been used to assist in the treatment of many skin diseases, such as photoaging, atrophic scar, hypertrophic scar, superficial pigmentation, vitiligo, and so on. However, the dynamic changes in skin function after fractional CO2 laser treatment are still unclear. This study explored the changes in local skin function and possible regulatory mechanisms after fractional CO2 laser treatment for 1, 3, 5 and 7 days through transcriptome high-throughput sequencing. The results showed that fractional CO2 laser tended to transform the “lesions” into “normal skin”, regulate the skin barrier, coordinate the rearrangement of collagen, enhance the local microvascular circulation, activate the immune system to secrete a large number of cytokines, and act as an auxiliary tool to assist drug transport. According to the basic principle of destruction before reconstruction, fractional CO2 laser plays a key role of balancer in skin reconstruction. In conclusion, the gene expression profiles of fractional CO2 laser-treated skin undergo dynamic changes over time. Skin barrier, collagen arrangement, drug metabolism, immune system and skin defense system all present obvious characteristics of timing change after fractional CO2 laser treatment.

Project description:Background Development of the flexible CO2 fiber has presented new opportunities for the use of precision laser cutting in cranial procedures. The efficacy of the CO2 scalpel is further enhanced by combining it with a fluid removal suction capability. Objectives We report our experience with a novel CO2 laser-suction device. Methods The novel laser-suction device was designed in conjunction with OmniGuide Inc. (Cambridge, Massachusetts, USA). We performed a case review of its use in firm tumors that were resistant to resection by bipolar, suction, and ultrasonic aspirator. Results The laser-suction device was applied in three tumors where resection with ultrasonic aspiration failed. Tumor resection using the laser-suction device was successful in all three cases. There were no complications related to the laser-suction device. There were no instances of intraoperative device malfunction. Discussion The CO2 laser combined with suction is a useful instrument for resection of firm tumors that prove to be resistant to ultrasonic aspiration. We also find it to be useful in settings where precise tissue incisions are desired with minimal manipulation. In our experience, the surgical efficiency of the CO2 laser is improved by the laser-suction device. This device allows the surgeon to utilize a suction device and laser in a single hand and enables concurrent use of bipolar electrocautery without repeated instrument changes.

Project description:Lead (Pb) is the most prevalent heavy metal pollutant in the natural environment. Pb is not a fundamental element for plants, but they absorb it when it is present in their environment, having no known physiological activity. The aim of our research was to evaluate the efficacy of laser photoacoustic spectroscopy as a tool to monitor changes induced by Pb in plant respiration by highlighting two molecular markers (C2H4 and CO2). To better understand Pb phytotoxicity, we monitored the plantlets evolution as well as the morphology of the root cells. Firstly, we showed that the treatment hinders the plantlet's development. Furthermore, using laser photoacoustic spectroscopy, we found a decrease in the concentration of C2H4 and CO2 vapors measured in the respiration of treated plants. Finally, fluorescence microscopy results showed that in Pb treated plantlets, the cell roots morphology is clearly altered as compared with the untreated ones. All the results are well correlated and can help further in understanding Pb phytotoxicity.
