Project description:X-ray microscopes are powerful tools in the nano-inspection of materials owing to their ultra-high resolution at the molecular level. However, the focusing efficiency of binary zone plate lenses as key components in such probes is merely 5% in practice, hindering their application in advanced scientific research. Although kinoform zone plate lenses are in principle supposed to possess high efficiency beyond binary ones, little progress has been reported so far due to the shortage of both a theoretical calculation approach and greyscale lithography for generating fine three-dimensional (3D) kinoform zones of the lenses. This paper reports our theoretical work for a modified beam propagation method to compute the focusing performance and state-of-the-art 3D greyscale electron beam lithography for kinoform zone plate lenses. Three different zone shapes - binary, kinoform and top-flat kinoform (nicknamed the trapezoid-kinoform) - were compared both theoretically and experimentally. Theoretical calculations suggest, for the first time, that the trapezoid-kinoform zone plate gives rise to the highest focusing efficiency among the three lenses, which was proved by optical characterization of the fabricated lens with hard X-rays. As high as 40% of the focusing efficiency by Au trapezoid-kinoform lenses with resolution of 250 nm at 8 keV has been achieved, which is two times higher than that of binary zone plate lenses. The origin of the enhanced efficiency in the trapezoid-kinoform zone plate lens was explained by the joint contributions from both the refraction through the kinoform slope and the diffraction through the top flat part of the trapezoid-kinoform zone plate. Such a breakthrough in focusing efficiency sheds light on the further development of X-ray lenses with both high resolution and high efficiency.

Project description:In this study, we report the characteristics of acoustic jets obtained through a mesoscale (radius less than 5 wavelengths) ABS cylinder made with a 3D printer. We have analyzed the influence of cylinder size on the characteristic parameters of an acoustic jet, such as maximum acoustic intensity at focus, Full Width at Half Maximum and length of Acoustic Jet. FWHM below 0.5 wavelength in AJ was experimentally obtained. It has been observed that there are two operating regimes depending on the cylinder radius: the resonant and the non-resonant. In the resonant regime, the excitation of Whispering Gallery Modes results in optimal parameter values of the acoustic jet. However, as it is a resonant regime, any minimal variation in cylinder size, working frequency or refractive index would make resonance disappear. In non-resonant mode, a phononic crystal has been embedded inside the cylinder and the characteristic parameters of the acoustic jet have been studied. These have been observed to improve. Finally, we have shown that curved acoustic jets can be obtained with the ABS cylinder with a phononic crystal embedded inside.

Project description:Diffractive zone plates have a wide range of applications from focusing x-ray to extreme UV radiation. The Gabor zone plate, which suppresses the higher-order foci to a pair of conjugate foci, is an attractive alternative to the conventional Fresnel zone plate. In this work, we developed a novel type of Beynon Gabor zone plate based on perfectly absorbing carbon nanotube forest. Lensing performances of 0, 8 and 20 sector Gabor zone plates were experimentally analyzed. Numerical investigations of Beynon Gabor zone plate configurations were in agreement with the experimental results. A high-contrast focal spot having 487 times higher intensity than the average background was obtained.

Project description:Overcoming the diffraction limit, which enables focusing much less than the wavelength, requires tailoring the evanescent spectrum of an aperture's field distribution. We model and simulate a corrugated near field plate, which can generate a sub-wavelength focus in inhomogeneous background media. All reactive coupling, between the metasurface near field plate and the focusing domain and among the corrugations in the metasurface, is taken into consideration with the finite element method, which we solve in combination with a constraint to generate a desired focus. Various geometries for the near field plate are considered and we demonstrate that the proposed method can effectively create a deeply sub-wavelength focus within a layered medium having properties resembling brain tissue. Such a device could find use as a detector of biological signals or for hyperthermic treatment near the skin surface.

Project description:Laser-plasma technology promises a drastic reduction of the size of high-energy electron accelerators. It could make free-electron lasers available to a broad scientific community and push further the limits of electron accelerators for high-energy physics. Furthermore, the unique femtosecond nature of the source makes it a promising tool for the study of ultrafast phenomena. However, applications are hindered by the lack of suitable lens to transport this kind of high-current electron beams mainly due to their divergence. Here we show that this issue can be solved by using a laser-plasma lens in which the field gradients are five order of magnitude larger than in conventional optics. We demonstrate a reduction of the divergence by nearly a factor of three, which should allow for an efficient coupling of the beam with a conventional beam transport line.

Project description:Variable light focusing is the ability to flexibly select the focal distance of a lens. This feature presents technical challenges, but is significant for optical interrogation of three-dimensional objects. Numerous lens designs have been proposed to provide flexible light focusing, including zoom, fluid, and liquid-crystal lenses. Although these lenses are useful for macroscale applications, they have limited utility in micron-scale applications due to restricted modulation range and exacting requirements for fabrication and control. Here, we present a holographic focusing method that enables variable light focusing without any physical modification to the lens element. In this method, a scattering layer couples low-angle (transverse wave vector) components into a full angular spectrum, and a digital optical phase conjugation (DOPC) system characterizes and plays back the wavefront that focuses through the scattering layer. We demonstrate micron-scale light focusing and patterning over a wide range of focal distances of 22-51 mm. The interferometric nature of the focusing scheme also enables an aberration-free scattering lens. The proposed method provides a unique variable focusing capability for imaging thick specimens or selective photoactivation of neuronal networks.

Project description:Gardnerella vaginalis has long been associated with bacterial vaginosis, a condition that increases the risk of women to preterm birth, sexually transmitted infections, and other adverse sequelae. However, G. vaginalis is also commonly found in healthy asymptomatic women of all ages. This raises the question if genetic differences among strains might distinguish potentially pathogenic from commensal strains. To disentangle the diversity of G. vaginalis, we invoked the concept of ecotypes-lineages of genetically and ecologically distinct strains within a named species-to better understand their evolutionary history and identify functional characteristics. We compared the genomes of G. vaginalis to six species in the closely related Bifidobacterium genus and found that G. vaginalis has a large accessory genome relative to Bifidobacterium, including many unique genes possibly involved in metabolism, drug resistance, and virulence. We then performed a comparative genomic analysis of 35 strains of G. vaginalis to infer a phylogeny based on the combined analysis of the core genome, using nucleotide substitution models, and the accessory genome, using gene gain/loss models. With the inferred tree topology, we performed comparisons of functional gene content among lineages that diverged at varying depths in the phylogeny and found significant differences in the representation of genes putatively involved in pathogenicity. Our functional enrichment analysis suggests that some lineages of G. vaginalis may possess enhanced pathogenic capabilities, including genes involved in mucus degradation like sialidases, while others may be commensal strains, lacking many of these pathogenic capabilities. The combined phylogenetic evidence and functional enrichment analysis allowed us to identify distinct ecotypes that have evolved in G. vaginalis as the result of the differential gene gain/loss for specific functions, including the capability to cause disease. We finally discuss how this analysis framework could be used to gain insight into the etiology of bacterial vaginosis and improve diagnosis.

Project description:Growth plate chondrocytes were isolated from the distal metacarpus of young dairy cattle (all under 10 mo of age), the chondrocytes were released from the extracellular matrix by digestion with Collagenase P for 4 hours, and the various zones of the growth plate were separated by density centrifugation. The least-dense Hypertrophic Zone (HZ) cells were compared to the most-dense Reserve Zone (RZ) cells. 6 pairs of HZ vs RZ were compared by microarray.

Project description:Focusing light into scattering media, although challenging, is highly desirable in many realms. With the invention of time-reversed ultrasonically encoded (TRUE) optical focusing, acousto-optic modulation was demonstrated as a promising guidestar mechanism for achieving noninvasive and addressable optical focusing into scattering media. Here, we report a new ultrasound-assisted technique, ultrasound-induced field perturbation optical focusing, abbreviated as UFP. Unlike in conventional TRUE optical focusing, where only the weak frequency-shifted first-order diffracted photons due to acousto-optic modulation are useful, here UFP leverages the brighter zeroth-order photons diffracted by an ultrasonic guidestar as information carriers to guide optical focusing. We find that the zeroth-order diffracted photons, although not frequency-shifted, do have a field perturbation caused by the existence of the ultrasonic guidestar. By detecting and time-reversing the differential field of the frequency-unshifted photons when the ultrasound is alternately ON and OFF, we can focus light to the position where the field perturbation occurs inside the scattering medium. We demonstrate here that UFP optical focusing has superior performance to conventional TRUE optical focusing, which benefits from the more intense zeroth-order photons. We further show that UFP optical focusing can be easily and flexibly developed into double-shot realization or even single-shot realization, which is desirable for high-speed wavefront shaping. This new method upsets conventional thinking on the utility of an ultrasonic guidestar and broadens the horizon of light control in scattering media. We hope that it provides a more efficient and flexible mechanism for implementing ultrasound-guided wavefront shaping.

Project description:In this study, we report the first experimental realization of an ultrathin (0.14λ, λ = 1.482 mm means wavelength at 1 MHz in the water medium) subwavelength focusing acoustic lens that can surpass the Rayleigh diffraction limit (0.61λ/NA, NA means numerical aperture). It is termed a Super-Oscillatory Acoustic Lens (SOAL), and it operates in the megasonic range. The SOAL represents an interesting feature allowing the achievement of subwavelength focusing without the need to operate in close proximity to the object to be imaged. The optimal layout of the SOAL is obtained by utilizing a systematic design approach, referred to here as topology optimization. To this end, the optimization formulation is newly defined. The optimized SOAL is fabricated using a photo-etching process and its subwavelength focusing performance is verified experimentally via an acoustic intensity measurement system. From these measurements, we found that the proposed optimized SOAL can achieve superior focusing features with a Full Width at Half Maximum (FWHM) of ~0.40λ/NA ≃ 0.84 mm (for our SOAL, NA = 0.707) with the transmission efficiency of 26.5%.
