Project description:Midlatitude storm tracks are preferred regions of intense activity of synoptic eddies shaping the day-to-day weather and several aspects of surface climate. Here statistical analyses of observationally-based atmospheric data and observed Arctic sea ice concentration (SIC) in the period 1979-2017 are used to identify linkages of a dominant mode of interannual variability in wintertime upper-tropospheric storm track activity over Eurasia (STAEA mode) to the concurrent surface climate anomalies and pre-winter Arctic SIC variations. This mode explains an exceptionally large fraction (about 70% of the variance) of the North Atlantic Oscillation (NAO) and of a dominant mode of Eurasian surface air temperature variations. As more than 50% of the variance of the STAEA mode and NAO is found to be accounted for by October SIC anomalies in the Barents/Kara Sea, it is concluded that wintertime Eurasian climate variability is to some extent predictable and that this predictability might have increased after an acceleration of the sea ice cover decline in the mid 2000s. These conclusions are supported by results from leave-1-yr-out cross-validated forecast experiments.

Project description:The assessment of climate impact on marine communities dwelling deeper than the well-studied shelf seas has been hampered by the lack of long-term data. For a long time, the prevailing expectation has been that thermal stability in deep ocean layers will delay ecosystem responses to warming. Few observational studies have challenged this view and indicated that deep organisms can respond exceptionally fast to physical change at the sea surface. To address the depth-specific impact of climate change, we investigated spatio-temporal changes in fish community structure along a bathymetry gradient of 150-1500 m between 1998 and 2016 in East Greenland. Here, the Arctic East Greenland Current and the Atlantic Irminger Current meet and mix, representing a sub-Arctic transition zone. We found the strongest signals of community reorganizations at depths between 350 and 1000 m and only weak responses in the shallowest and deepest regions. Changes were in synchrony with atmospheric warming, loss in sea ice and variability in physical sea surface conditions both within our study region and North of the Denmark Strait. These results suggest that interannual variability and long-term climate trends of the larger ecoregion can rapidly affect fish communities down to 1000-m depth through atmospheric ocean coupling and food web interactions.

Project description:The Circumpolar Biodiversity Monitoring Programme (CBMP) provides an opportunity to improve our knowledge of Arctic arthropod diversity, but initial baseline studies are required to summarise the status and trends of planned target groups of species known as Focal Ecosystem Components (FECs). We begin this process by collating available data for a relatively well-studied region in the Arctic, the North Atlantic region, summarising the diversity of key terrestrial arthropod FECs, and compiling trends for some representative species. We found the FEC classification system to be challenging to implement, but identified some key groups to target in the initial phases of the programme. Long-term data are scarce and exhibit high levels of spatial and temporal variability. Nevertheless, we found that a number of species and groups are in decline, mirroring patterns in other regions of the world. We emphasise that terrestrial arthropods require higher priority within future Arctic monitoring programmes.

Project description:Vb cyclones typically emerge in the Western Mediterranean and propagate to the Northeast into Central Europe. This paper explores the temporal characteristics of Vb cyclone occurrence based on cyclone tracks identified at the atmospheric levels of Z700 and sea level pressure, using JRA-55 reanalysis data for the period 1959-2015. The risk of Vb occurrence was significantly high in the 1960s and has remained at a lower level since then. Vb cyclones do not occur fully randomly according to a Poisson point process. Eleven well-separated and distinct clusters as well as 11 hiatus periods are identified, with average occurrence rates of 21.5 and 5.2 yrea-1, respectively. During the event of Vb, the large-scale atmospheric circulation is changed into a state favoring the development of successive Vb cyclones. Clustering is very prominent in the case of Genoan Vb cyclones in summer as well as those Vb cyclones developing over the Iberian Peninsula or the North African Coast in winter. Superposition of the polar and the subtropical jet stream over the Western Mediterranean is identified as a main feature at the onset of Vb cyclones. Vb cyclone occurrence appears to be synchronized with the Northern Atlantic Oscillation (NAO; at Z500) and Arctic Oscillation (AO; at Z1000). Clusters have occurred when both NAO and AO were negative. This relation applies to Western Mediterranean cyclones not following a Vb track as well, however to a much weaker extent. In contrast, Vb cyclone frequency was particularly low from 1988 to 1997 during a sustained positive phase of both NAO and AO.

Project description:MicroPoly(A)Pure kits (Ambion) were used to prepare mRNA from P. salmonis-infected and control Atlantic salmon macrophages. Infected and control (24 h incubation) macrophage mRNA samples were subjected to two rounds of amplification using the MessageAmp aRNA kit (Ambion) and manufacturer's instructions. Approximately 400 ng of macrophage mRNA was used as template in the first round of amplification, yielding 8.1 ug of infected macrophage aRNA and 7.8 ug of control macrophage aRNA. Approximately 1 ug of first-round macrophage aRNA was used as template in the second round of amplification, yielding 81.2 ug of infected macrophage aRNA and 83.3 ug of control macrophage aRNA. Infected and control macrophage aRNA samples were visualized on agarose gels to check quality and quantity. Except for the amounts of aRNA and reverse transcription (RT) primer used, macrophage target labeling reactions and microarray hybridizations were identical. Macrophage target syntheses used 5 ug of second-round aRNA and 5 ug of random primers (Roche). RT primers were mixed with aRNA samples, and nuclease-free H2O (Invitrogen) was added to make 10 ul total volume. Samples were incubated at 70 degrees C 4 min, then placed on ice. RT reactions included 50 mM Tris HCl pH 8.3, 75 mM KCl, 3 mM MgCl2, 10 mM DTT, 0.5 mM each dATP, dCTP, and dGTP, 0.2 mM dTTP, 0.05 mM Cy3-dUTP or Cy5-dUTP (Amersham), 40U RNAguard ribonuclease inhibitor (Amersham), and 400U Superscript II Rnase H- reverse transcriptase (Invitrogen). RT reactions were incubated at 42 degrees C for 2.5 h, followed by addition of 0.025U RNase A (Sigma) and 1.5U RNase H (New England BioLabs) and incubation at 37 degrees C for 30 min. Labeled targets were cleaned using the Qiaquick PCR purification kit (Qiagen) and precipitated, with 300 mM sodium acetate, 1 ul of 20 ug/ul glycogen (Invitrogen), and 2.5 volumes of 100% ethanol, at -20 degrees C overnight. Each labeled target was recovered by centrifugation (14,000 rpm, 4 degrees C, 1 h), washed in 70% ethanol, air-dried, and resuspended in 60 ul of hybridization buffer: 50% deionized formamide, 5X SSC, 0.1% SDS, 5 ul of 5 ug/ul oligo dT (5'T18[Q-N]3'), 5 ul of 2 ug/ul BSA (Pierce), and 2 ul of 10 ug/ul sonicated human placental DNA (Sigma). Targets were incubated at 96 degrees C for 3 min, and then at 65 degrees C until applied to microarrays. Microarrays were prepared for hybridization by washing 2 X 5 min in 0.1% SDS, washing 5 X 1 min in MilliQ H2O, immersing 3 min in 95 degrees C MilliQ H2O, and drying by centrifugation (2000 rpm, 5 min, in 50 ml conical tube). Microarray hybridizations were run in the dark under HybriSlips hybridization covers (Grace Biolabs) in slide hybridization chambers (Corning) submerged in a 45 degrees C water bath for 16 h. Coverslips were floated off in 45 degrees C (1X SSC, 0.2% SDS), and arrays were washed 1 X 10 min in 45 degrees C (1X SSC, 0.2% SDS), 3 X 4 min in room temperature (0.1X SSC, 0.1% SDS), and 4 X 4 min in room temperature 0.1X SSC. Slides were dried by centrifugation as before. Fluorescent images of hybridized arrays were acquired immediately at 10 um resolution using ScanArray Express (PerkinElmer). The Cy3 and Cy5 cyanine fluors were excited at 543 nm and 633 nm respectively, and the same laser power (90%) and photomultiplier tube (PMT) settings were used for all slides in a study (PMT 72 Cy3, PMT 63 or 64 Cy5 for macrophage study). Project description:Increasing influence of Atlantic water in the Arctic Ocean has the potential to significantly impact regional water temperature and salinity. Here we use a rDNA barcoding approach to reveal how microbial communities are partitioned into distinct assemblages across a gradient of Atlantic-Polar Water influence in the Norwegian Sea. Data suggest that temperate adapted bacteria may replace cold water taxa under a future scenario of increasing Atlantic influence, but the eukaryote response is more complex. Some abundant eukaryotic cold water taxa could persist, while less abundant eukaryotic taxa may be replaced by warmer adapted temperate species. Furthermore, within lineages, different taxa display evidence of increased relative abundance in reaction to favourable conditions and we observed that rare microbial taxa are sample site rather than region specific. Our findings have significant implications for the vulnerability of polar associated community assemblages, which may change, impacting the ecosystem services they provide, under predicted increases of Atlantic mixing and warming within the Arctic region.

Project description:The export of deep water from the Arctic to the Atlantic contributes to the formation of North Atlantic Deep Water, a crucial component of global ocean circulation. Records of protactinium-231 (231Pa) and thorium-230 (230Th) in Arctic sediments can provide a measure of this export, but well-constrained sedimentary budgets of these isotopes have been difficult to achieve in the Arctic Ocean. Previous studies revealed a deficit of 231Pa in central Arctic sediments, implying that some 231Pa is either transported to the margins, where it may be removed in areas of higher particle flux, or exported from the Arctic via deep water advection. Here we investigate this "missing sink" of Arctic 231Pa and find moderately increased 231Pa deposition along Arctic margins. Nonetheless, we determine that most 231Pa missing from the central basin must be lost via advection into the Nordic Seas, requiring deep water advection of 1.1 - 6.4 Sv through Fram Strait.

Project description:In 2019, it was estimated that more than 50 million captive Atlantic salmon in Norway died in the final stage of their production in marine cages. This mortality represents a significant economic loss for producers and a need to improve welfare for farmed salmon. Single adverse events, such as algal blooms or infectious disease outbreaks, can explain mass mortality in salmon cages. However, little is known about the production, health, or environmental factors that contribute to their baseline mortality during the sea phase. Here we conducted a retrospective study including 1627 Atlantic salmon cohorts put to sea in 2014-2019. We found that sea lice treatments were associated with Atlantic salmon mortality. In particular, the trend towards non-medicinal sea lice treatments, including thermal delousing, increases Atlantic salmon mortality in the same month the treatment is applied. There were differences in mortality among production zones. Stocking month and weight were other important factors, with the lowest mortality in smaller salmon stocked in August-October. Sea surface temperature and salinity also influenced Atlantic salmon mortality. Knowledge of what affects baseline mortality in Norwegian aquaculture can be used as part of syndromic surveillance and to inform salmon producers on farming practices that can reduce mortality.

Project description:Although the Atlantic puffin Fratercula arctica is well studied throughout its temperate and low Arctic breeding range, few have studied the species in its far northern distribution. This study is the first to present data on the migratory movements of the "large-billed" subspecies, F. a. naumanni, that breeds in the high Arctic and which has significantly larger body size than those farther south. During 2013-2015, migration tracks were collected from nine adult puffins (6 males and 3 females) tagged with geolocators in northwest Greenland. Overall, female puffins traveled farther than males on their annual migration, with one female puffin traveling over 13,600 km, which was nearly a third farther than any tagged male in our study. Differential migration was observed in migratory phenology and route, with males using a form of chain migration with acute synchrony between individuals while females appeared to largely use leap-frog migration and showed little synchrony between individuals. Extreme sexual segregation in wintering areas was evidenced by two females that migrated to the southern limit of the species' range while the six males remained at the northern limit, and wintered along the sea ice edge during portions of the non-breeding season. Male puffins thus wintered in regions with sea surface temperatures up to 10° C cooler than female puffins, and in areas with generally colder sea surface temperatures when compared to previously known wintering areas of temperate and low Arctic puffin breeding populations. The degree to which body size enables male F. a. naumanni to remain in colder waters likely reflects differing life history constraints between sexes and populations (i.e., subspecies). Further study is warranted to investigate how recent changes in climate have further exacerbated the observed differences between sexes in high Arctic puffins and possibly other marine avian species.
