Project description:While separating sounds into frequency components and subsequently converting them into patterns of neural firing, the mammalian cochlea processes signal components in ways that depend strongly on frequency. Indeed, both the temporal structure of the response to transient stimuli and the sharpness of frequency tuning differ dramatically between the apical and basal (i.e., the low- and high-frequency) regions of the cochlea. Although the mechanisms that give rise to these pronounced differences remain incompletely understood, they are generally attributed to tonotopic variations in the constituent hair cells or cytoarchitecture of the organ of Corti. As counterpoint to this view, we present a general acoustic treatment of the horn-like geometry of the cochlea, accompanied by a simple 3-D model to elucidate the theoretical predictions. We show that the main apical/basal functional differences can be accounted for by the known spatial gradients of cochlear dimensions, without the need to invoke mechanical specializations of the sensory tissue. Furthermore, our analysis demonstrates that through its functional resemblance to an ear horn (aka ear trumpet), the geometry of the cochlear duct manifests tapering symmetry, a felicitous design principle that may have evolved not only to aid the analysis of natural sounds but to enhance the sensitivity of hearing.

Project description:Signaling pathways relay information about changes in the external environment so that cells can respond appropriately. How much information a pathway can carry depends on its bandwidth. We designed a microfluidic device to reliably change the environment of single cells over a range of frequencies. Using this device, we measured the bandwidth of the Saccharomyces cerevisiae signaling pathway that responds to high osmolarity. This prototypical pathway, the HOG pathway, is shown to act as a low-pass filter, integrating the signal when it changes rapidly and following it faithfully when it changes more slowly. We study the dependence of the pathway's bandwidth on its architecture. We measure previously unknown bounds on all of the in vivo reaction rates acting in this pathway. We find that the two-component Ssk1 branch of this pathway is capable of fast signal integration, whereas the kinase Ste11 branch is not. Our experimental techniques can be applied to other signaling pathways, allowing the measurement of their in vivo kinetics and the quantification of their information capacity.

Project description:BACKGROUND: Auditory evoked responses can be modulated by both the sequencing and the signal-to-noise ratio of auditory stimuli. Constant sequencing as well as intense masking sounds basically lead to N1m response amplitude reduction. However, the interaction between these two factors has not been investigated so far. Here, we presented subjects tone stimuli of different frequencies, which were either concatenated in blocks of constant frequency or in blocks of randomly changing frequencies. The tones were presented either in silence or together with broad-band noises of varying levels. RESULTS: In silence, tones presented with random sequencing elicited a larger N1m response than tones presented with constant sequencing. With increasing noise level, this difference decreased and even vanished in the condition where noise intensity exceeded the tone intensity by 10 dB. Furthermore, under noisy conditions, the N1m latency was shorter in the constant sequencing condition compared to the random sequencing condition. CONCLUSIONS: Besides the well-known neural habituation mechanisms, bottom-up driven attention plays an important role during auditory processing in noisy environments. This bottom-up driven attention would allow us to track a certain auditory signal in noisy situations without voluntarily paying attention to the auditory modality.

Project description:BackgroundDual processing theory of human cognition postulates that reasoning and decision-making can be described as a function of both an intuitive, experiential, affective system (system I) and/or an analytical, deliberative (system II) processing system. To date no formal descriptive model of medical decision-making based on dual processing theory has been developed. Here we postulate such a model and apply it to a common clinical situation: whether treatment should be administered to the patient who may or may not have a disease.MethodsWe developed a mathematical model in which we linked a recently proposed descriptive psychological model of cognition with the threshold model of medical decision-making and show how this approach can be used to better understand decision-making at the bedside and explain the widespread variation in treatments observed in clinical practice.ResultsWe show that physician's beliefs about whether to treat at higher (lower) probability levels compared to the prescriptive therapeutic thresholds obtained via system II processing is moderated by system I and the ratio of benefit and harms as evaluated by both system I and II. Under some conditions, the system I decision maker's threshold may dramatically drop below the expected utility threshold derived by system II. This can explain the overtreatment often seen in the contemporary practice. The opposite can also occur as in the situations where empirical evidence is considered unreliable, or when cognitive processes of decision-makers are biased through recent experience: the threshold will increase relative to the normative threshold value derived via system II using expected utility threshold. This inclination for the higher diagnostic certainty may, in turn, explain undertreatment that is also documented in the current medical practice.ConclusionsWe have developed the first dual processing model of medical decision-making that has potential to enrich the current medical decision-making field, which is still to the large extent dominated by expected utility theory. The model also provides a platform for reconciling two groups of competing dual processing theories (parallel competitive with default-interventionalist theories).

Project description:Understanding how the human speech production system is related to the human auditory system has been a perennial subject of inquiry. To investigate the production-perception link, in this paper, a computational analysis has been performed using the articulatory movement data obtained during speech production with concurrently recorded acoustic speech signals from multiple subjects in three different languages: English, Cantonese, and Georgian. The form of articulatory gestures during speech production varies across languages, and this variation is considered to be reflected in the articulatory position and kinematics. The auditory processing of the acoustic speech signal is modeled by a parametric representation of the cochlear filterbank which allows for realizing various candidate filterbank structures by changing the parameter value. Using mathematical communication theory, it is found that the uncertainty about the articulatory gestures in each language is maximally reduced when the acoustic speech signal is represented using the output of a filterbank similar to the empirically established cochlear filterbank in the human auditory system. Possible interpretations of this finding are discussed.

Project description:Autism spectrum disorder (ASD) is characterised by social communication difficulties. These difficulties have been mainly explained by cognitive, motivational, and emotional alterations in ASD. The communication difficulties could, however, also be associated with altered sensory processing of communication signals. Here, we assessed the functional integrity of auditory sensory pathway nuclei in ASD in three independent functional magnetic resonance imaging experiments. We focused on two aspects of auditory communication that are impaired in ASD: voice identity perception, and recognising speech-in-noise. We found reduced processing in adults with ASD as compared to typically developed control groups (pairwise matched on sex, age, and full-scale IQ) in the central midbrain structure of the auditory pathway (inferior colliculus [IC]). The right IC responded less in the ASD as compared to the control group for voice identity, in contrast to speech recognition. The right IC also responded less in the ASD as compared to the control group when passively listening to vocal in contrast to non-vocal sounds. Within the control group, the left and right IC responded more when recognising speech-in-noise as compared to when recognising speech without additional noise. In the ASD group, this was only the case in the left, but not the right IC. The results show that communication signal processing in ASD is associated with reduced subcortical sensory functioning in the midbrain. The results highlight the importance of considering sensory processing alterations in explaining communication difficulties, which are at the core of ASD.

Project description:Hearing perception in individuals with auditory hallucinations has not been well studied. Auditory hallucinations have previously been shown to involve primary auditory cortex activation. This activation suggests that auditory hallucinations activate the terminal of the auditory pathway as if auditory signals are submitted from the cochlea, and that a hallucinatory event is therefore perceived as hearing. The primary auditory cortex is stimulated by some unknown source that is outside of the auditory pathway. The current study aimed to assess the outcomes of stimulating the primary auditory cortex through the auditory pathway in individuals who have experienced auditory hallucinations. Sixteen patients with schizophrenia underwent functional magnetic resonance imaging (fMRI) sessions, as well as hallucination assessments. During the fMRI session, auditory stimuli were presented in one-second intervals at times when scanner noise was absent. Participants listened to auditory stimuli of sine waves (SW) (4-5.5kHz), English words (EW), and acoustically reversed English words (arEW) in a block design fashion. The arEW were employed to deliver the sound of a human voice with minimal linguistic components. Patients' auditory hallucination severity was assessed by the auditory hallucination item of the Brief Psychiatric Rating Scale (BPRS). During perception of arEW when compared with perception of SW, bilateral activation of the globus pallidus correlated with severity of auditory hallucinations. EW when compared with arEW did not correlate with auditory hallucination severity. Our findings suggest that the sensitivity of the globus pallidus to the human voice is associated with the severity of auditory hallucination.

Project description:Canavan Disease (CD) is a leukodystrophy caused by homozygous null mutations in the gene encoding aspartoacylase (ASPA). ASPA-deficiency is characterized by severe psychomotor retardation, and excessive levels of the ASPA substrate N-acetylaspartate (NAA). ASPA is an oligodendrocyte marker and it is believed that CD has a central etiology. However, ASPA is also expressed by Schwann cells and ASPA-deficiency in the periphery might therefore contribute to the complex CD pathology. In this study, we assessed peripheral and central auditory function in the AspalacZ/lacZ rodent model of CD using auditory brainstem response (ABR). Increased ABR thresholds and the virtual loss of waveform peaks 4 and 5 from AspalacZ/lacZ mice, indicated altered central auditory processing in mutant mice compared with Aspawt/wt controls and altered central auditory processing. Analysis of ABR latencies recorded from AspalacZ/lacZ mice revealed that the speed of nerve conduction was unchanged in the peripheral part of the auditory pathway, and impaired in the CNS. Histological analyses confirmed that ASPA was expressed in oligodendrocytes and Schwann cells of the auditory system. In keeping with our physiological results, the cellular organization of the cochlea, including the organ of Corti, was preserved and the spiral ganglion nerve fibres were normal in ASPA-deficient mice. In contrast, we detected substantial hypomyelination in the central auditory system of AspalacZ/lacZ mice. In summary, our data suggest that the lack of ASPA in the CNS is responsible for the observed hearing deficits, while ASPA-deficiency in the cochlear nerve fibres is tolerated both morphologically and functionally.

Project description:Neural processing of sounds in the dorsal and ventral streams of the (human) auditory cortex is optimized for analyzing fine-grained temporal and spectral information, respectively. Here we use a Wilson and Cowan firing-rate modeling framework to simulate spectro-temporal processing of sounds in these auditory streams and to investigate the link between neural population activity and behavioral results of psychoacoustic experiments. The proposed model consisted of two core (A1 and R, representing primary areas) and two belt (Slow and Fast, representing rostral and caudal processing respectively) areas, differing in terms of their spectral and temporal response properties. First, we simulated the responses to amplitude modulated (AM) noise and tones. In agreement with electrophysiological results, we observed an area-dependent transition from a temporal (synchronization) to a rate code when moving from low to high modulation rates. Simulated neural responses in a task of amplitude modulation detection suggested that thresholds derived from population responses in core areas closely resembled those of psychoacoustic experiments in human listeners. For tones, simulated modulation threshold functions were found to be dependent on the carrier frequency. Second, we simulated the responses to complex tones with missing fundamental stimuli and found that synchronization of responses in the Fast area accurately encoded pitch, with the strength of synchronization depending on number and order of harmonic components. Finally, using speech stimuli, we showed that the spectral and temporal structure of the speech was reflected in parallel by the modeled areas. The analyses highlighted that the Slow stream coded with high spectral precision the aspects of the speech signal characterized by slow temporal changes (e.g., prosody), while the Fast stream encoded primarily the faster changes (e.g., phonemes, consonants, temporal pitch). Interestingly, the pitch of a speaker was encoded both spatially (i.e., tonotopically) in Slow area and temporally in Fast area. Overall, performed simulations showed that the model is valuable for generating hypotheses on how the different cortical areas/streams may contribute toward behaviorally relevant aspects of auditory processing. The model can be used in combination with physiological models of neurovascular coupling to generate predictions for human functional MRI experiments.

Project description:In the present preregistered study, we evaluated the possibility of a shared cognitive mechanism during verbal and non-verbal tasks and therefore the implication of domain-general cognitive control during language comprehension. We hypothesized that a behavioral cost will be observed during a dual-task including both verbal and non-verbal difficult processing. Specifically, to test this claim, we designed a dual-task paradigm involving: an auditory language comprehension task (sentence comprehension) and a non-verbal Flanker task (including congruent and incongruent trials). We manipulated sentence ambiguity and evaluated if the ambiguity effect modified behavioral performances in the non-verbal Flanker task. Under the assumption that ambiguous sentences induce a more difficult process than unambiguous sentences, we expected non-verbal flanker task performances to be impaired only when a simultaneous difficult language processing is performed. This would be specifically reflected by a performance cost during incongruent Flanker items only during ambiguous sentence presentation. Conversely, we observed a facilitatory effect for the incongruent Flanker items during ambiguous sentence suggesting better non-verbal inhibitory performances when an ambiguous sentence was simultaneously processed. Exploratory data analysis suggests that this effect is not only related to a more difficult language processing but also to the previous (n-1) Flanker item. Indeed, results showed that incongruent n-1 Flanker items led to a facilitation of the incongruent synchronized Flanker items only when ambiguous sentences were conjointly presented. This result, even if it needs to be corroborated in future studies, suggests that the recruitment of executive control mechanisms facilitates subsequent executive control implication during difficult language processing. The present study suggests a common executive control mechanism during difficult verbal and non-verbal tasks.
