Project description:Nitrogen and carbon stable isotopic compositions (?15N and ?13C) of consumers have been used for physiological and food web studies. Previous studies have shown ?15N and ?13C values are affected by several biological and environmental factors during starvation, but the generality of the effect of starvation on ?15N and ?13C values has not yet been tested. Here, we performed a meta-analysis to evaluate the effects of starvation on ?15N and ?13C values of consumers, and the underlying factors that may explain the observed variation. The ?15N and ?13C values were calculated as the differences between the final ?15N and ?13C values of consumers (post-starvation) and the pre-starvation values on each experiment. Our meta-analysis showed a large variation in the ?15N and ?13C values of consumers (?15N range: -0.82 to 4.30‰; mean: 0.47‰ and ?13C range: -1.92 to 2.62‰; mean: 0.01‰). The ?15N values of most consumers increased along the length of the starvation period and were influenced by nitrogen excretion and thermoregulation types, probably because differences in nitrogen metabolism and thermoregulation affect nitrogen processing and excretion rates. None of our predictor variables accounted for the variation in ?13C values, which showed both increases and decreases due to fasting. Our findings suggest that starvation results in changes in consumer ?15N values which are mainly explained by the length of the fasting period and by nitrogen and energy metabolism, but the underlying mechanisms of the starvation effects on ?13C values seem to be more complex than previously thought.

Project description:Stable isotope analysis of feces can provide a non-invasive method for tracking the dietary habits of nearly any mammalian species. While fecal samples are often collected for macroscopic and genetic study, stable isotope analysis can also be applied to expand the knowledge of species-specific dietary ecology. It is somewhat unclear how digestion changes the isotope ratios of animals' diets, so more controlled diet studies are needed. To date, most diet-to-feces controlled stable isotope experiments have been performed on herbivores, so in this study I analyzed the carbon and nitrogen stable isotope ratios in the diet and feces of the meerkat (Suricata suricatta), a small omnivorous mammal. The carbon trophic discrimination factor between diet and feces (?13Cfeces) is calculated to be 0.1 ± 1.5‰, which is not significantly different from zero, and in turn, not different than the dietary input. On the other hand, the nitrogen trophic discrimination factor (?15Nfeces) is 1.5 ± 1.1‰, which is significantly different from zero, meaning it is different than the average dietary input. Based on data generated in this experiment and a review of the published literature, carbon isotopes of feces characterize diet, while nitrogen isotope ratios of feces are consistently higher than dietary inputs, meaning a discrimination factor needs to be taken into account. The carbon and nitrogen stable isotope values of feces are an excellent snapshot of diet that can be used in concert with other analytical methods to better understand ecology, diets, and habitat use of mammals.

Project description:Tibetan Plateau is located at a mountain region isolated from direct anthropogenic sources. Mercury concentrations and stable isotopes of carbon, nitrogen, and mercury were analyzed in sediment and biota for Nam Co and Yamdrok Lake. Biotic mercury concentrations and high food web magnification factors suggested that Tibetan Plateau is no longer a pristine site. The primary source of methylmercury was microbial production in local sediment despite the lack of direct methylmercury input. Strong ultraviolet intensity led to extensive photochemical reactions and up to 65% of methylmercury in water was photo-demethylated before entering the food webs. Biota displayed very high ?(199)Hg signatures, with some highest value (8.6%) ever in living organisms. The ?(202)Hg and ?(199)Hg in sediment and biotic samples increased with trophic positions (?(15)N) and %methylmercury. Fish total length closely correlated to ?(13)C and ?(199)Hg values due to dissimilar carbon sources and methylmercury pools in different living waters. This is the first mercury isotope study on high altitude lake ecosystems that demonstrated specific isotope fractionations of mercury under extreme environmental conditions.

Project description:Americans spend >100 billion dollars on restaurant fast food each year; fast food meals comprise a disproportionate amount of both meat and calories within the U.S. diet. We used carbon and nitrogen stable isotopes to infer the source of feed to meat animals, the source of fat within fries, and the extent of fertilization and confinement inherent to production. We sampled food from McDonald's, Burger King, and Wendy's chains, purchasing >480 servings of hamburgers, chicken sandwiches and fries within geographically distributed U.S. cities: Los Angeles, San Francisco, Denver, Detroit, Boston, and Baltimore. From the entire sample set of beef and chicken, only 12 servings of beef had delta(13)C < -21 per thousand; for these animals only was a food source other than corn possible. We observed remarkably invariant values of delta(15)N in both beef and chicken, reflecting uniform confinement and exposure to heavily fertilized feed for all animals. The delta(13)C value of fries differed significantly among restaurants indicating that the chains used different protocols for deep-frying: Wendy's clearly used only corn oil, whereas McDonald's and Burger King favored other vegetable oils; this differed from ingredient reports. Our results highlighted the overwhelming importance of corn agriculture within virtually every aspect of fast food manufacture.

Project description:There is limited information that provides a comprehensive understanding of the trophic ecology of Mexican Pacific harbor seal (Phoca vitulina richardii) colonies. While scat analysis has been used to determine the diet of some colonies, the integrative characterization of its feeding habits on broader temporal and spatial scales remains limited. We examined potential feeding grounds, trophic niche width, and overlap, and inferred the degree of dietary specialization using stable carbon and nitrogen isotope ratios (δ13C and δ15N) in this subspecies. We analyzed δ13C and δ15N on fur samples from pups collected at five sites along the western coast of the Baja California Peninsula, Mexico. Fur of natal coat of Pacific harbor seal pups begins to grow during the seventh month in utero until the last stage of gestation. Therefore pup fur is a good proxy for the mother's feeding habits in winter (~December to March), based on the timing of gestation for the subspecies in this region. Our results indicated that the δ13C and δ15N values differed significantly among sampling sites, with the highest mean δ15N value occurring at the southernmost site, reflecting a well-characterized north to south latitudinal 15N-enrichment in the food web. The tendency identified in δ13C values, in which the northern colonies showed the most enriched values, suggests nearshore and benthic-demersal feeding habits. A low variance in δ13C and δ15N values for each colony (<1‰) and relatively small standard ellipse areas suggest a specialized foraging behavior in adult female Pacific harbor seals in Mexican waters.

Project description:Knowing the distribution of marine animals is central to understanding climatic and other environmental influences on population ecology. This information has proven difficult to gain through capture-based methods biased by capture location. Here we show that marine location can be inferred from animal tissues. As the carbon isotope composition of animal tissues varies with sea surface temperature, marine location can be identified by matching time series of carbon isotopes measured in tissues to sea surface temperature records. Applying this technique to populations of Atlantic salmon (Salmo salar L.) produces isotopically-derived maps of oceanic feeding grounds, consistent with the current understanding of salmon migrations, that additionally reveal geographic segregation in feeding grounds between individual philopatric populations and age-classes. Carbon isotope ratios can be used to identify the location of open ocean feeding grounds for any pelagic animals for which tissue archives and matching records of sea surface temperature are available.

Project description:Stable isotope measurements are increasingly being used to gain insights into the nutritional ecology of many wildlife species and their role in ecosystem structure and function. Such studies require estimations of trophic discrimination factors (i.e. differences in the isotopic ratio between the consumer and its diet). Although trophic discrimination factors are tissue- and species-specific, researchers often rely on generalized, and fixed trophic discrimination factors that have not been experimentally derived. In this experimental study, captive wild boar (Sus scrofa) were fed a controlled diet of corn (Zea mays), a popular and increasingly dominant food source for wild boar in the Czech Republic and elsewhere in Europe, and trophic discrimination factors for stable carbon (?13C) and nitrogen (?15N) isotopes were determined from hair samples. The mean ?13C and ?15N in wild boar hair were -2.3‰ and +3.5‰, respectively. Also, in order to facilitate future derivations of isotopic measurements along wild boar hair, we calculated the average hair growth rate to be 1.1 mm d(-1). Our results serve as a baseline for interpreting isotopic patterns of free-ranging wild boar in current European agricultural landscapes. However, future research is needed in order to provide a broader understanding of the processes underlying the variation in trophic discrimination factors of carbon and nitrogen across of variety of diet types.

Project description:At the regional scale, although environmental factors are known to shape the distributions of belowground communities in terrestrial ecosystems, these environmental factors account for relatively low percentages of the variation in belowground communities. More of this variation might be explained by considering ecosystem stable isotopic values, which can provide insight into environmental conditions. Here, we investigated ecosystem (plant and soil) δ13C and δ15N values and belowground communities (microbes and nematodes) as well as environmental factors (climates, soils, and plants) across the Mongolian Plateau. The regression analyses showed that plant isotopic values were more closely associated with belowground communities than soil isotopic values, while ecosystem δ13C values were more closely associated with the belowground communities than ecosystem δ15N values. We also found isotopic values were more closely associated with nematode communities than microbial communities. Variation partioning analyses indicated that environmental variables together explained 16-45% of total variation in belowground communities. After isotopic variables were added as predictors to the variation partition analyses, the explanation of the variance was improved by14-24% for microbial communities and was improved by 23-44% for nematode communities. These findings indicate that isotopic values could be used to predict the properties of belowground communities at a regional scale.

Project description:Food choices affect the isotopic composition of the body with each food item leaving its distinct isotopic imprint. The common view is that the natural abundance of the stable isotopes of nitrogen (expressed as δ15N) is higher in animals than in plants that constitute our contemporary diets. Higher δ15N is thus increasingly viewed as a biomarker for meat and fish intake. Here we show that organic compared to conventional farming increases plant δ15N to an extent that can appreciably impact the performance of δ15N as a biomarker. The error that can arise when organic plants are consumed was modelled for the entire range of proportions of plant versus animal protein intake, and accounting for various intakes of organic and conventionally grown crops. This mass balance model allows the interpretation of differences in δ15N in light of organic food consumption. Our approach shows that the relationship between δ15N and meat and fish intake is highly contextual and susceptible to variation at the population, community or group level. We recommend that fertilization practices and organic plant consumption must not be overlooked when using δ15N as a biomarker for meat and fish intake or to assess compliance to nutritional interventions.

Project description:Stable carbon and nitrogen isotopes are often used to make inferences of past environments and social patterns. We analyze ? 13C and ? 15N values in contemporary kukui (Aleurites moluccanus) endocarp to examine the effects of site environment. Results from across environmental transects on Hawai'i Island show strong patterns for both stable isotopes. For ? 13C a robust linear relationship with elevation is exhibited, strengthened by the inclusion of rainfall. This relationship breaks down at a minimum threshold of annual rainfall, possible relating to physiological responses to drought. For ? 15N, the only significant relationship observed pertains to substrate age. The endocarp from kukui is one of the most readily identified plant remains in the Pacific archaeological records and is often targeted for radiocarbon dating. We discuss the potential implications of our results regarding ancient climate, inferred diets, and habitat composition.
