Fibular hemimelia is a rare congenital disorder with partial or complete absence of the fibula. It is usually associated with other osseous and soft tissue abnormalities of the knee joint and lower limb. Project description:Cerebral palsy-the most common physical disability of childhood-is a clinical diagnosis encompassing a heterogeneous group of neurodevelopmental disorders that cause impairments of movement and posture that persist throughout life. Despite being commonly attributed to a range of environmental factors, particularly birth asphyxia, the specific cause of cerebral palsy remains unknown in most individuals. A growing body of evidence suggests that cerebral palsy is probably caused by multiple genetic factors, similar to other neurodevelopmental disorders such as autism and intellectual disability. Recent advances in next-generation sequencing technologies have made possible rapid and cost-effective sequencing of the entire human genome. Novel cerebral palsy genes will probably be identified as more researchers and clinicians use this approach to study individuals with undiagnosed neurological disorders. As our knowledge of the underlying pathophysiological mechanisms of cerebral palsy increases, so will the possibility of developing genomically guided therapeutic interventions.

Project description:This work extends the three level powered knee and ankle prosthesis control framework previously developed by the authors by adding sitting mode. A middle level finite state based impedance controller is designed to accommodate sitting, sit-to-stand and stand-to-sit transitions. Moreover, a high level Gaussian Mixture Model based intent recognizer is developed to distinguish between standing and sitting modes and switch the middle level controllers accordingly. Experimental results with unilateral transfemoral amputee subject show that sitting down and standing up intent can be inferred from the prosthesis sensor signals by the intent recognizer. Furthermore, it is demonstrated that the prosthesis generates net active power of 50 W during standing up and dissipates up to 50 W of power during stand-to-sit transition at the knee joint.

Project description:Schistosomiasis, caused mainly by S. mansoni, S. haematobium and S. japonicum, continues to be a serious tropical disease and public health problem resulting in an unacceptably high level of morbidity in countries where it is endemic. Praziquantel, the only drug currently available for treatment, is unable to kill developing schistosomes, it does not prevent re-infection and its continued extensive use may result in the future emergence of drug-resistant parasites. This scenario provides impetus for the development and deployment of anti-schistosome vaccines to be used as part of an integrated approach for the prevention, control and eventual elimination of schistosomiasis. This review considers the present status of candidate vaccines for schistosomiasis, and provides some insight on future vaccine discovery and design.

Project description:The Wenzel model, commonly used for predicting the equilibrium contact angle (CA) of drops which penetrate the asperities of a rough surface, does not account for the liquid volume stored in the asperities. Interestingly, many previous experimental and molecular dynamics studies have noted discrepancies between observed CAs and those predicted by the Wenzel model because of this neglected liquid volume. Here, we apply a thermodynamic model to wetting of periodically patterned surfaces to derive a volume-corrected Wenzel equation in the limit of small pattern wavelength (compared to drop size). We show that the corrected equilibrium CA is smaller than that predicted by the Wenzel equation and that the reduction in CA can be significant when the liquid volume within the asperities becomes non-negligible compared to the total droplet volume. In such cases, the corrected CAs agree reasonably well with experimental observations and results of molecular dynamics simulations reported in previous studies.

Project description:To estimate k-space trajectory errors in non-Cartesian acquisitions and reconstruct distortion-free images, without trajectory measurements or gradient calibrations.The Trajectory Auto-Corrected image Reconstruction method jointly estimates k-space trajectory errors and images, based on SENSE and SPIRiT parallel imaging reconstruction. The underlying idea is that parallel imaging and oversampling in the center of k-space provides data redundancy that can be exploited to simultaneously reconstruct images and correct trajectory errors. Trajectory errors are represented as weighted sums of trajectory-dependent error basis functions, the coefficients of which are estimated using gradient-based optimization.Trajectory Auto-Corrected image Reconstruction was applied to reconstruct images and errors in golden angle radial, center-out radial, and spiral in vivo 7 Tesla brain acquisitions in five subjects. Compared to reconstructions using nominal trajectories, Trajectory auto-corrected image reconstructions contained considerably less blurring and streaking and were of similar quality to images reconstructed using measured k-space trajectories in the center-out radial and spiral cases. Reconstruction cost function reductions and improvements in normalized image gradient squared were also similar to those for images reconstructed using measured trajectories.Trajectory Auto-Corrected image Reconstruction enables non-Cartesian image reconstructions free from trajectory errors without the need for separate gradient calibrations or trajectory measurements. Project description:Cryogenic fluorescent light microscopy of flash-frozen cells stands out by artifact-free fixation and very little photobleaching of the fluorophores used. To attain the highest level of resolution, aberration-free immersion objectives with accurately matched immersion media are required, but both do not exist for imaging below the glass-transition temperature of water. Here, we resolve this challenge by combining a cryoimmersion medium, HFE-7200, which matches the refractive index of room-temperature water, with a technological concept in which the body of the objective and the front lens are not in thermal equilibrium. We implemented this concept by replacing the metallic front-lens mount of a standard bioimaging water immersion objective with an insulating ceramic mount heated around its perimeter. In this way, the objective metal housing can be maintained at room temperature, while creating a thermally shielded cold microenvironment around the sample and front lens. To demonstrate the range of potential applications, we show that our method can provide superior contrast in Escherichia coli and yeast cells expressing fluorescent proteins and resolve submicrometer structures in multicolor immunolabeled human bone osteosarcoma epithelial (U2OS) cells at [Formula: see text]C.

Project description:Underestimation of reliability is discussed from the viewpoint of deflation in estimates of reliability caused by artificial systematic technical or mechanical error in the estimates of correlation (MEC). Most traditional estimators of reliability embed product-moment correlation coefficient (PMC) in the form of item-score correlation (Rit) or principal component or factor loading (λ i ). PMC is known to be severely affected by several sources of deflation such as the difficulty level of the item and discrepancy of the scales of the variables of interest and, hence, the estimates by Rit and λ i are always deflated in the settings related to estimating reliability. As a short-cut to deflation-corrected estimators of reliability, this article suggests a procedure where Rit and λ i in the estimators of reliability are replaced by alternative estimators of correlation that are less deflated. These estimators are called deflation-corrected estimators of reliability (DCER). Several families of DCERs are proposed and their behavior is studied by using polychoric correlation coefficient, Goodman-Kruskal gamma, and Somers delta as examples of MEC-corrected coefficients of correlation.

Project description:Lower-limb strength is a marker of functional decline in elders. This work studies the feasibility of using the quasi-periodic nature of the distance between a subjects' back and the chair backrest during a 30-s chair-stand test (CST) to carry out unsupervised measurements based on readings from a low-cost ultrasound sensor. The device comprises an ultrasound sensor, an Arduino UNO board, and a Bluetooth module. Sit-to-stand transitions are identified by filtering the signal with a moving minimum filter and comparing the output to an adaptive threshold. An inter-rater reliability (IRR) study was carried out to validate the device ability to count the same number of valid transitions as the gold-standard manual count. A group of elders (age: mean (m) = 80.79 years old, SD = 5.38; gender: 21 female and seven male) were asked to perform a 30-s CST using the device while a trained nurse manually counted valid transitions. Ultimately, a moving minimum filter was necessary to cancel the effect of outliers, likely produced because older people tend to produce more motion artefacts and, thus, noisier signals. While the intra-class correlation coefficient (ICC) for this study was good (ICC = 0.86, 95% confidence interval (CI) = 0.73, 0.93), it is not yet clear whether the results are sufficient to support clinical decision-making.

Project description:Metallo-?-Lactamases (MBLs) are class B?-lactamases that hydrolyze almost all clinically-available?-lactam antibiotics. MBLs feature the distinctive ??/?? sandwich fold of the metallo-hydrolase/oxidoreductase superfamily and possess a shallow active-site groove containing one or two divalent zinc ions, flanked by flexible loops. According to sequence identity and zinc ion dependence, MBLs are classified into three subclasses (B1, B2 and B3), of which the B1 subclass enzymes have emerged as the most clinically significant. Differences among the active site architectures, the nature of zinc ligands, and the catalytic mechanisms have limited the development of a common inhibitor. In this review, we will describe the molecular epidemiology and structural studies of the most prominent representatives of class B1 MBLs (NDM-1, IMP-1 and VIM-2) and describe the implications for inhibitor design to counter this growing clinical threat.

Project description:In this review, we examine the current status of Staphylococcus aureus vaccine development and the prospects for future vaccines. Examination of the clinical trials to date show that murine models have not predicted success in humans for active or passive immunization. A key factor in the failure to develop a vaccine to prevent S. aureus infections comes from our relatively limited knowledge of human protective immunity. More recent reports on the elements of the human immune response to staphylococci are analysed. In addition, there is some controversy concerning the role of antibodies for protecting humans, and these data are reviewed. From a review of the current state of understanding of staphylococcal immunity, a working model is proposed. Some new work has provided some initial candidate biomarker(s) to predict outcomes of invasive infections and to predict the efficacy of antibiotic therapy in humans. We conclude by looking to the future through the perspective of lessons gleaned from the clinical vaccine trials.
