ABSTRACT: The raccoon dog (Nyctereutes procyonoides) is an introduced, invasive species in Europe. Literature data show that raccoon dogs act as a reservoir of many dangerous parasites, including nematodes of the genus Trichinella. The aims of the study were to determine the prevalence of Trichinella spp. infection in raccoon dogs collected from the Głęboki Bród Forest District between 2013 and 2016, and to evaluate their distribution in the muscle tissue of the host. The larvae of Trichinella spp. were detected in 45 raccoon dogs (39.82%), and all of them were identified as T. britovi. No mixed infection was observed. The intensity of infection ranged from 0.02 to 622.92 larvae per gram (LPG), and the highest mean was observed in the tongue and lower forelimb in both examined sexes. The raccoon dog may play a significant role as a reservoir of T. britovi in the wildlife in the examined area.

Project description:The present study compares the immunogenic patterns of muscle larvae excretory-secretory proteins (ML E-S) from T. spiralis and T. britovi recognized by Trichinella-infected human sera. Samples were analyzed using two-dimensional electrophoresis (2-DE) coupled with 2D-immunoblot and liquid chromatography-tandem mass spectrometry LC-MS/MS analysis, two ELISA procedures and a confirmatory 1D-immunoblot test. Sera were obtained from nine patients with a history of ingestion of raw or undercooked meat who presented typical clinical manifestations of trichinellosis and from eleven healthy people. Specific anti-Trichinella IgG antibodies were detected in all samples tested with the Home-ELISA kits, but in only four samples for the commercially-available kit. The 1D-immunoblot results indicated that all nine serum samples were positive for T. spiralis ML E-S antigens, expressed as the presence of specific bands. In contrast, eight of the serum samples with T. britovi E-S ML antigens were positive, with one serum sample taken from a patient at 33dpi (days post infection) being negative. To identify immunoreactive proteins that are specifically recognized by host antibodies, both species of ML E-S proteins were subjected to 2D-immunoblotting with human serum taken at 49 dpi. The sera recognized 22 protein spots for T. spiralis and 18 for T. britovi in 2D-immunoblot analysis. Their molecular weights (MW) ranged from 50 to 60 kDa. LC-MS/MS analysis identified both common and specifically-recognized immunoreactive proteins: transmembrane serine protease 9, serine protease, antigen targeted by protective antibodies and Actin-1 partial were shared for both Trichinella species; hypothetical protein T01_7775 and P49 antigen, partial those specific to T. spiralis; deoxyribonuclease-2-alpha and hypothetical protein T03_17187/T12_7360 were specific to T. britovi. Our results demonstrate the value of 2-DE and 2D-immunblot as versatile tools for pinpointing factors contributing to the parasite-host relationship by comparing the secretomes of different Trichinella species.

Project description:Numerous bird species, often rare or endangered, rely on the presence of standing and downed deadwood for shelter, nesting, and foraging. Habitat quality was evaluated on the basis of deadwood volume, the density of large standing deadwood, and the space filling index (SFI). The SFI reflects the degree of space filling of the bottom layers taking into account tree trunks, seedlings, saplings, ground vegetation, stumps, and downed deadwood. Analysis encompassed all special protection areas (SPAs) in Poland (a total of 107 SPAs containing 7974 sample plots monitored under the National Forest Inventory). An additional in-depth analysis was conducted for 30 SPAs with the greatest share of forest habitats. The studied indicators varied substantially both between and within individual SPAs, with deadwood volume ranging from 1.3 to 50.5 m3 ha-1 (mean of 9.0 m3 ha-1) and the density of large standing deadwood (diameter at breast height ≥ 30 cm) from 0.1 to 16.0 ind ha-1 (mean of 2.2 ind ha-1). These values were relatively low compared to the density of living trees with corresponding dimensions (111 ind ha-1). SFI analysis indicated high or very high space filling of the bottom forest layers on 14-56% of sample plots in a given SPA. The presence of deadwood was found to be significantly positively affected by SPA location in the mountains, a greater proportion of sites with higher fertility, a greater share of forest area under strict protection, as well as higher stand volume within a given SPA. The correlation between deadwood volume and the density of birds (primary and secondary cavity nesters) in individual SPAs was positive (R = 0.60). As compared to lowland areas, SPAs in mountain areas are generally characterized by high stand volumes, a greater density of large living trees, and a greater amount of diverse deadwood. In those areas conservation measures should involve continuous monitoring and diagnosing of any problems associated with the populations of individual bird species; focused efforts should be implemented to support those species that exhibit unfavorable population trends. In most lowland SPAs measures aimed at the improvement of site conditions for birds must be more extensive than in the mountains, with a low abundance of dead trees (especially large ones). These parameters can be improved by retaining some senescent stands in managed forests until their natural death and implementing a strict protection regime in areas of high conservation value.

Project description:Woodland salamanders are among the most abundant vertebrate animals in temperate deciduous forests of eastern North America. Because of their abundance, woodland salamanders are responsible for the transformation of nutrients and translocation of energy between highly disparate levels of trophic organization: detrital food webs and high-order predators. However, the spatial extent of woodland salamanders' role in the ecosystem is likely contingent upon the distribution of their biomass throughout the forest. We sought to determine if natural environmental gradients influence the fine-scale distribution and density of Southern Ravine Salamanders (Plethodon richmondi) and Cumberland Plateau Salamanders (P. kentucki). We addressed this objective by constructing occupancy, co-occurrence, and abundance models from temporally replicated surveys within an old-growth forest in the Cumberland Plateau region of Kentucky. We found that Plethodon richmondi had a more restricted fine-scale distribution than P. kentucki (mean occupancy probability [ ?¯^ ] = 0.737) and exhibited variable density, from <250 to >1000 individuals per hectare, associated with increased soil moisture and reduced solar exposure due to slope face. While more ubiquitously distributed ( ?¯^ = 0.95), P. kentucki density varied from <400 to >1,000 individuals per hectare and was inversely related to increased solar exposure from canopy disturbance and landscape convexity. Our data suggest co-occurrence patterns of P. richmondi and P. kentucki are influenced primarily by abiotic conditions within the forest, and that populations likely occur independently and without evidence of biotic interaction. Given the critical role that woodland salamanders play in the maintenance of forest health, regions that support large populations of woodland salamanders, such as those highlighted in this study-mesic forest stands on north-to-east facing slopes with dense canopy and abundant natural cover, may provide enhanced ecosystem services and support the stability of the total forest.
