Project description:ObjectivesOur study targets the potential of the local urban mosquito Aedes aegypti to experimentally transmit chikungunya virus (CHIKV), dengue virus (DENV), yellow fever virus (YFV), and Zika virus (ZIKV).MethodsWe collected eggs and adults of Ae. aegypti in Medellín, Colombia (from February to March 2020) for mosquito experimental infections with DENV, CHIKV, YFV and ZIKV and viral detection using the BioMark Dynamic arrays system.ResultsWe show that Ae. aegypti from Medellín was more prone to become infected, to disseminate and transmit CHIKV and ZIKV than DENV and YFV.ConclusionsThus, in Colombia, chikungunya is the most serious threat to public health based on our vector competence data.

Project description:Mosquito oviposition site selection is essential for vector population dynamics and malaria epidemiology. Irrigated rice cultivations provide ideal larval habitats for malaria mosquitoes, which has resulted in increased prevalence of the malaria vector, Anopheles arabiensis, in sub-Saharan Africa. The nature and origin of the cues regulating this behaviour are only now being elucidated. We show that gravid Anopheles arabiensis are attracted and oviposit in response to the odour present in the air surrounding rice. Furthermore, we identify a synthetic rice odour blend, using electrophysiological and chemical analyses, which elicits attraction and oviposition in laboratory assays, as well as attraction of free-flying gravid mosquitoes under semi-field conditions. This research highlights the intimate link between malaria vectors and agriculture. The identified volatile cues provide important substrates for the development of novel and cost-effective control measures that target female malaria mosquitoes, irrespective of indoor or outdoor feeding and resting patterns.

Project description:BACKGROUND:Most malaria vectors belong to species complexes. Sibling species often exhibit divergent behaviors dictating the measures that can be deployed effectively in their control. Despite the importance of the Anopheles funestus complex in malaria transmission in sub-Saharan Africa, sibling species have rarely been identified in the past and their vectoring potential remains understudied. METHODS:We analyzed 1149 wild-caught An. funestus (senso lato) specimens from 21 sites in Kenya, covering the major malaria endemic areas including western, central and coastal areas. Indoor and outdoor collection tools were used targeting host-seeking and resting mosquitoes. The identity of sibling species, infection with malaria Plasmodium parasites, and the host blood meal sources of engorged specimens were analyzed using PCR-based and sequencing methods. RESULTS:The most abundant sibling species collected in all study sites were Anopheles funestus (59.8%) and Anopheles rivulorum (32.4%) among the 1062 successfully amplified specimens of the An. funestus complex. Proportionally, An. funestus dominated in indoor collections whilst An. rivulorum dominated in outdoor collections. Other species identified were Anopheles leesoni (4.6%), Anopheles parensis (2.4%), Anopheles vaneedeni (0.1%) and for the first time in Kenya, Anopheles longipalpis C (0.7%). Anopheles funestus had an overall Plasmodium infection rate of 9.7% (62/636), predominantly Plasmodium falciparum (59), with two infected with Plasmodium ovale and one with Plasmodium malariae. There was no difference in the infection rate between indoor and outdoor collections. Out of 344 An. rivulorum, only one carried P. falciparum. We also detected P. falciparum infection in two non-blood-fed An. longipalpis C (2/7) which is the first record for this species in Kenya. The mean human blood indices for An. funestus and An. rivulorum were 68% (93/136) and 64% (45/70), respectively, with feeding tendencies on a broad host range including humans and domestic animals such as cow, goat, sheep, chicken and pig. CONCLUSIONS:Our findings underscore the importance of active surveillance through application of molecular approaches to unravel novel parasite-vector associations possibly contributed by cryptic species with important implications for effective malaria control and elimination.

Project description:In mammals, the serine protease plasmin degrades extracellular proteins during blood clot removal, tissue remodeling, and cell migration. The zymogen plasminogen is activated into plasmin by two serine proteases: tissue-type plasminogen activator (tPA) and urokinase-type plasminogen activator (uPA), a process regulated by plasminogen activator inhibitor 1 (PAI-1), a serine protease inhibitor that specifically inhibits tPA and uPA. Plasmodium gametes and sporozoites use tPA and uPA to activate plasminogen and parasite-bound plasmin degrades extracellular matrices, facilitating parasite motility in the mosquito and the mammalian host. Furthermore, inhibition of plasminogen activation by PAI-1 strongly blocks infection in both hosts. To block parasite utilization of plasmin, we engineered Anopheles stephensi transgenic mosquitoes constitutively secreting human PAI-1 (huPAI-1) in the midgut lumen, in the saliva, or both. Mosquitoes expressing huPAI-1 strongly reduced rodent and human Plasmodium parasite transmission to mosquitoes, showing that co-opting plasmin for mosquito infection is a conserved mechanism among Plasmodium species. huPAI-1 expression in saliva induced salivary gland deformation which affects sporozoite invasion and P. berghei transmission to mice, resulting in significant levels of protection from malaria. Targeting the interaction of malaria parasites with the fibrinolytic system using genetically engineered mosquitoes could be developed as an intervention to control malaria transmission.

Project description:BackgroundThroughout large parts of sub-Saharan Africa, seasonal malaria transmission follows mosquito density, approaching zero during the dry season and peaking during the wet season. The mechanisms by which malaria mosquitoes survive the long dry season, when no larval sites are available remain largely unknown, despite being long recognized as a critical target for vector control. Previous work in the West African Sahel has led to the hypothesis that Anopheles coluzzii (formerly M-form Anopheles gambiae) undergoes aestivation (dry-season diapause), while Anopheles gambiae (s.s.) (formerly S-form An. gambiae) and Anopheles arabiensis repopulate each wet season via long-distance migration. The environmental cues used by these species to signal the oncoming dry season have not been determined; however, studies, mostly addressing mosquitoes from temperate zones, have highlighted photoperiod and temperature as the most common token stimuli for diapause initiation. We subjected newly established colonies of An. coluzzii and An. arabiensis from the Sahel to changes in photoperiod to assess and compare their responses in terms of longevity and other relevant phenotypes.ResultsOur results showed that short photoperiod alone and to a lesser extent, lower nightly temperature (representing the early dry season), significantly increased longevity of An. coluzzii (by ~30%, P < 0.001) but not of An. arabiensis. Further, dry season conditions increased body size but not relative lipid content of An. coluzzii, whereas body size of An. arabiensis decreased under these conditions.ConclusionsThese species-specific responses underscore the capacity of tropical anophelines to detect mild changes (~1 h) in photoperiod and thus support the role of photoperiod as a token stimulus for An. coluzzii in induction of aestivation, although, these responses fall short of a complete recapitulation of aestivation under laboratory conditions.

Project description:BackgroundFor malaria control in Africa it is crucial to characterise the dispersal of its most efficient vector, Anopheles gambiae, in order to target interventions and assess their impact spatially. Our study is, we believe, the first to present a statistical model of dispersal probability against distance from breeding habitat to human settlements for this important disease vector.Methods/principal findingsWe undertook post-hoc analyses of mosquito catches made in The Gambia to derive statistical dispersal functions for An. gambiae sensu lato collected in 48 villages at varying distances to alluvial larval habitat along the River Gambia. The proportion dispersing declined exponentially with distance, and we estimated that 90% of movements were within 1.7 km. Although a 'heavy-tailed' distribution is considered biologically more plausible due to active dispersal by mosquitoes seeking blood meals, there was no statistical basis for choosing it over a negative exponential distribution. Using a simple random walk model with daily survival and movements previously recorded in Burkina Faso, we were able to reproduce the dispersal probabilities observed in The Gambia.Conclusions/significanceOur results provide an important quantification of the probability of An. gambiae s.l. dispersal in a rural African setting typical of many parts of the continent. However, dispersal will be landscape specific and in order to generalise to other spatial configurations of habitat and hosts it will be necessary to produce tractable models of mosquito movements for operational use. We show that simple random walk models have potential. Consequently, there is a pressing need for new empirical studies of An. gambiae survival and movements in different settings to drive this development.

Project description:Variation in vectorial capacity for human malaria among Anopheles mosquito species is determined by many factors, including behavior, immunity, and life history. To investigate the genomic basis of vectorial capacity and explore new avenues for vector control, we sequenced the genomes of 16 anopheline mosquito species from diverse locations spanning ~100 million years of evolution. Comparative analyses show faster rates of gene gain and loss, elevated gene shuffling on the X chromosome, and more intron losses, relative to Drosophila. Some determinants of vectorial capacity, such as chemosensory genes, do not show elevated turnover but instead diversify through protein-sequence changes. This dynamism of anopheline genes and genomes may contribute to their flexible capacity to take advantage of new ecological niches, including adapting to humans as primary hosts.

Project description:Reports of the expansion of the Asia malaria vector Anopheles stephensi mosquito into new geographic areas are increasing, which poses a threat to the elimination of urban malaria. Efficient surveillance of this vector in affected areas and early detection in new geographic areas is key to containing and controlling this species. To overcome the practical difficulties associated with the morphological identification of immature stages and adults of An. stephensi mosquitoes, we developed a species-specific PCR and a real-time PCR targeting a unique segment of the second internal transcribed spacer lacking homology to any other organism. Both PCRs can be used to identify An. stephensi mosquitoes individually or in pooled samples of mixed species, including when present in extremely low proportions (1:500). This study also reports a method for selective amplification and sequencing of partial ribosomal DNA from An. stephensi mosquitoes for their confirmation in pooled samples of mixed species.
