Project description:Handedness and language are two well-studied examples of asymmetrical brain function in humans. Approximately 90% of humans exhibit a right-hand preference, and the vast majority shows left-hemisphere dominance for language function. Although genetic models of human handedness and language have been proposed, the actual gene expression differences between cerebral hemispheres in humans remain to be fully defined. In the present study, gene expression profiles were examined in both hemispheres of three cortical regions involved in handedness and language in humans and their homologues in rhesus macaques: ventrolateral prefrontal cortex, posterior superior temporal cortex (STC), and primary motor cortex. Although the overall pattern of gene expression was very similar between hemispheres in both humans and macaques, weighted gene correlation network analysis revealed gene co-expression modules associated with hemisphere, which are different among the three cortical regions examined. Notably, a receptor-enriched gene module in STC was particularly associated with hemisphere and showed different expression levels between hemispheres only in humans.

Project description:One of the serious constraints to realize high level of rice crop productivity in agriculture has been due to Soil Water Stress (SWS) situation that growing plants often face. In order to increase or maintain the crop productivity in SWS situation, our initial aim is to understand the drought response mechanism in different genotypes of rice. For thorough analysis of SWS situation in rice we have taken here two wild genotypes of rice namely Oryza nivara, Oryza rufipogon and three Oryza sativa indica cultivars namely Nagina-22, IR20 and Vandana, where IR20 is known to be susceptible and Vandana is known to be tolerant under SWS condition [GSE49364 and the current study]. Global analysis of transcript profiling under SWS condition reveal the actual picture of genes responsive to stress situation in different genetic background of rice. Furthermore it would help us in the selection of most desirable resource for crop breeding without compromising the yield of crop. We used the 44k rice Oligoarray from Agilent technologies to study the expression profiles from five rice genotypes during vegetative (Veg) and grain-filling (GF) stages under varying water conditions, viz. Before Stress (BS), After Stress (AS) and After Recovery (AR).

Project description:Language is a unique human capability with limited molecular understanding due to the lack of animal models and technical constraints. Transcriptomics analysis offers a comprehensive view of gene expression in specific tissues, aiding the understanding of their functions. However, such patterns have been underexplored in language-related regions of the human brain. This study conducts a comprehensive transcriptomic analysis of 125 samples from 13 language-related Brodmann areas (BAs) in both hemispheres of five human postmortem brains. The expression landscape of human language-related regions is mapped, revealing higher expression in the right hemisphere, notably BA45 (Broca’s area) and BA3/1/2 (ventral sensory-motor cortex). Integrative analysis of differentially expressed genes and language-relevant genetic discoveries provides insights into the rs62060948 locus. The findings suggest that the rs62060948-MYC-WNT3 axis plays a crucial role in language function in BA44 of the right hemisphere. Behavior tests in mice show that Wnt3 knockdown in the right auditory cortex leads to abnormal behaviors, confirming that imbalanced Wnt3 expression impacts language function. Pathway analysis indicates that Wnt3 maintains nervous system development through neuron ensheathment. This study enhances understanding of the molecular mechanisms underlying human language function and identifies potential targets for language disorder therapies.

Project description:Glioblastoma (GBM) is a deadly cancer in which cancer stem cells (CSCs) sustain tumor growth and contribute to therapeutic resistance. Protein Arginine Methyltransferase 5 (PRMT5) has recently emerged as a promising target in GBM. Using two orthogonal-acting inhibitors of PRMT5 (GSK591 or LLY-283), we show that pharmacological inhibition of PRMT5 suppresses the growth of a cohort of 46 patient-derived GBM stem cell cultures, with the proneural subtype showing greater sensitivity. We show that PRMT5 inhibition caused widespread disruption of splicing across the transcriptome, particularly affecting cell cycle gene products. We identify a GBM splicing signature that correlates with the degree of response to PRMT5 inhibition. Importantly, we demonstrate that LLY-283 is brain-penetrant and significantly prolongs the survival of mice with orthotopic patient-derived xenografts. Collectively, our findings provide a rationale for the clinical development of brain penetrant PRMT5 inhibitors as treatment for GBM.

Project description:Aims: In an effort to gain further insight into the underlying mechanisms tied to disease onset and progression of Gulf War Illness (GWI), our team evaluated GWI patient response to stress utilizing RNA-Seq. Main Methods: The protocol included blood collection before (baseline), during (peak of exercise, measured by VO2 max), and after exercise challenge (recovery – four hours post-exercise challenge). Peripheral blood mononuclear cell (PBMC) transcriptomics data were analyzed using DESeq2 and DEXseq software to understand why GWI patients process stressors differently from their healthy counterparts. Key findings: Our findings validate previously identified dysregulation of immune and inflammatory pathways among GWI patients as well as highlight novel immune and inflammatory markers of disease activity. These results provide a foundation for future research efforts in understanding GWI pathophysiology and creating targeted treatments. Significance: Gulf War Illness is a complex, chronic, and debilitating multi-system illness impacting 25%-30% of the U.S. troops deployed to the 1990-1991 Gulf War. The condition is characterized by medically unexplained fatigue and affects multiple organ systems. Because the underlying mechanisms are largely unknown, patients receive symptom-based treatment, rather than targeting fundamental biological processes. To the best of our knowledge, this is the first study that applies RNA-Seq to analyze the effect of GWI, and the response to stressors in GWI, on the transcriptomic changes in circulating immune cells.
