Project description:The sustainable development of modern aquaculture must rely on a significant reduction of the fish meal (FM) used in aquafeed formulations. However, FM substitution with alternative ingredients in diets for carnivorous fish species often showed reduced nutrient absorption, significantly perturbed metabolisms and histological changes at both hepatic and intestinal level. In the present study, adult rainbow trout (Oncorhynchus mykiss) were fed three different experimental aquafeed formulations. A control diet with higher FM content (27.3%) than two test formulations in which fish meal was substituted with two more sustainable and promising alternatives: insect meal (Hermetia illucens larvae=10.1%, FM=11.6%) and poultry by-products meal (PBM=14.8%; FM=11.7%). Combined metabolomics and proteomics analyses of fish liver, together with histological examination of liver and intestine demonstrated that a well balanced formulation of nutrients in the three diets allowed high metabolic compatibility of either substitutions, paving the way for innovative and sustainable use of novel raw materials for the fish feed industry. Results show that the main metabolic pathways of nutrient absorption and catabolism were essentially unaltered by alternative feed ingredients, and also histological alterations were negligible. It is demonstrated that substitution of fish meal with sustainable alternatives does not impact on fish metabolism, given proper efforts are put in fulfilling nutritional requirements of rainbow trout.

Project description:Frass is the by-product of the larval meal industry and includes larval waste, exoskeleton sheds, and residual feed ingredients. Experimental frass was derived from the larvae of black solder flies (Hermetia illucens) fed distillers dried grains. A 10-week study was conducted to evaluate the effect of dietary levels of frass on the global gene expression of channel catfish, Ictalurus punctatus. Three diets containing 0, 50, and 300 g frass per kg diet were fed to channel catfish (5.24 ± 0.04 g) in quadruplicate aquaria to apparent satiation twice daily. Intestine (n=12 in pools of 3) and liver (n=12 in pools of 3) tissues were taken from fish at the end of the trial and processed for high-throughput Illumina RNA sequencing (RNAseq). Pairwise comparisons identified both up- and down-regulated genes in frass diets compared to no frass controls.

Project description:In the context of replacing fish meal and fish oil in feeds for aquaculture, rainbow trout alevins received from first-feeding onwards, one of the three experimental diets: V (100% plant-based), C (mix of FM-FO & plant ingredients) or M (100% FM-FO based). The long term effects of such dietary replacement on the intestinal (mid gut) and hepatic transcriptome were studied in juveniles after a 7-month feeding trial at 7°C.

Project description:Interventions: The general rehabilitation program will consist of a multidisciplinary education and exercise program, which will be provided as group sessions twice weekly for 8 weeks. The education component will include 1-hour nutritional advice and management provided by a dietitian; 1-hour exercise education regarding the importance of exercise to well-being, bladder and bowel advice, and adherence to exercise provided by a physiotherapist; 1-hour fatigue management and return to work strategies provided by an occupational therapist; and 5-hours health behavior change and self-management strategies provided by a psychologist. The exercise component will consist of a moderate intensity (Borg Rating of Perceived Exertion (RPE) Scale 12-14), physiotherapist-supervised, combined aerobic and resistance training program. The exercise program will be individualized based on the results from baseline outcome measures. Participants will be required to attend an one hour education session per week and an one hour exercise session per week over the 8-week intervention period. The home-based walking program requires participants to perform daily walking with duration and intensity individually prescribed based on the results of baseline walking tests. To encourage adherence to the home-based walking program, 6 telephone motivational coaching sessions (1 to 2 calls per month) will be provided from a physiotherapist, between data collection time-points 2 (immediately post-intervention) and 3 (6 months following baseline) . Daily feedback for participants regarding distance and number of steps taken will be encouraged with a simple pedometer.
Primary outcome(s): Functional capacity as assessed using the six-minute walk test [At baseline, immediately post-intervention, and at 6 months following baseline];Feasibility as assessed by the recruitment rate, adherence, withdrawals, and adverse events. There are minimal risks associated with the rehabilitation and measurement tests. Participants may feel some general muscle discomfort after exercise, which is a normal response to exercise.[At baseline, immediately post-intervention, and at 6, 12, 24, and 60 months following baseline]

Project description:We performed RNA-seq, H3K27ac ChIP-seq, and HNF4a ChIP-seq on jejunal intestinal epithelial cells, which are primarily responsible for the absorption of fatty acids, in four conditions: Germ-free (GF), Germ-free plus high fat meal (GF+HFM), ex-GF colonized with a conventional microbiota for 2 weeks (Colonized, CV), and Colonized plus high fat meal (CV+HFM). We, for the first time, map genomewide HFM responsive regulatory regions in the intestine. We identify that in the absence of microbes the HNF4a transcriptional program supports a FAO program in enterocytes while suppressing a proliferation program.

Project description:Purpose: Aim of this study was to determine the miRNAs associated with average daily gain in Kangal Akkaraman lambs. Depending on the average daily gain, two groups were formed as low average daily gain (LDG) and high average daily gain (HDG). Methods: Peripheral blood mononuclear cells (pbmc) were separated from blood samples obtained from lambs and RNA was isolated with TRI Reagent (Sigma). Isolated total RNAs from LDG (n = 17) and HDG (n = 21) groups were pooled. miRNA libraries (LDG and HDG groups) were prepared with NEBNext® Multiplex Small RNA Library Prep Set for Illumina® (NEB, USA.) and sequenced with Illumina Novaseq. Readings were mapped with the Bowtie program using the Oar_v3.1 sheep reference genome. Differential expression analysis of the two groups was performed using the DESeq R package (1.8.3). Results: Using an optimized data analysis workflow, we mapped about 18 million sequence reads in HDG and 21 million sequence reads n LDG samples to the ovis aries genome (oar_v3.1). In total, 129 known miRNAs were identified, 134 novel miRNAs were predicted, and 40 differentially expressed miRNAs were found between the groups. Conclusions: Our study represents the first detailed analysis of Kangal type Akkaraman sheep PBMC miRNAs with RNA-seq technology. The obtained sequence reads would provide data for futher breeding studies related to average daily gain in Kangal Akkraman sheep.

Project description:For the purpose of mechanism-based risk assessment, we investigated temporal concentration-dependent responses in cultures of PHH and HepaRG cells exposed to three cosmetics ingredients, 2,7-naphthalenediol (NPT), triclosan (TCS) and butylated hydroxytoluene (BHT), that are suspected to have liver injury liability. To facilitate the identification of early KEs and visualisation of their development over time, samples were collected at 4 time points, namely 8 and 24 hours (single exposure), 48 and 72 hours (daily repeated exposure), after exposure to a broad concentration range. Concentrations were selected based on the estimated Cmax of the cosmetic ingredients. The selected maximum concentration was set at approximately 100x Cmax, whilst the minimum tested concentration was approximately 0.2x Cmax. The large concentration range allowed to apply benchmark concentration (BMC) modelling on individual genes as well as gene co-expression networks to derive in vitro transcriptomics benchmark concentrations (BMCs) and assess their suitability to be used as PoD in chemical risk assessment.

Project description:Human health effects from chronic exposure to pesticide residues are little investigated. We compared standard histopathology and serum biochemistry measures and multi-omics analyses in an in vivo subchronic toxicity test of a glyphosate, its formulated product MON 52276, and mixture of six pesticide active ingredients frequently detected in foodstuffs (azoxystrobin, boscalid, chlorpyrifos, glyphosate, imidacloprid and thiabendazole). Sprague-Dawley rats were administered with the pesticide mixture with each ingredient at its regulatory permitted acceptable daily intake.
