Epidemiological and Molecular Colorectal Cancer Registry

ABSTRACT: This is a Registry that invites patients undergoing colorectal surgery for colorectal cancer. Epidemiological data is collected. The Registry includes tumor tissue and blood banks for analyzing different genetic mutations and disease-specific biomarkers.

Project description:Expression profiling of whole blood cells isolated from patients piror to undergoing cardiac catheterization. The Cathgen Registry is a single-center coronary catheter-lab cohort being run at Duke University for the purpose of identifying biomarkers associated with coronary disease. 3 condition experiment: Cases (2) are patients with ?70% stenosis in >1 major vessel or ?50% stenosis in >2 arteries; intermediates (1) are patients with luminal stenosis >25% but less than 50%; controls (0) have luminal stenosis of <=25%

NSABP Patient Registry and Biospecimen Profiling Repository

Project description:This is a study to collect and analyze tissue specimens from metastatic colorectal cancer (mCRC) patients for the development of a molecularly profiled tissue repository for the primary purpose of maintaining a patient registry for future clinical trials based on the molecular profile of the tumors. The main purpose of testing these tissue specimens is to identify genetic alterations or biomarkers associated with colorectal cancer such that if new agent(s) become available, particularly those that target these genetic alterations/biomarkers, participants may be offered the opportunity to take part in a National Surgical Adjuvant Breast and Bowel Project (NSABP) treatment protocol.

Exome Genotyping Study of Scripps Venous Thrombosis Registry using Axiom® Exome Genotyping Array

Project description:Younger age and VTE recurrence are more likely to be caused by genetic risk factors than secondary VTE in older patients who more likely have comorbidities. When the exome rare variant genotyping database of the Scripps VTE Registry for adults < 55 yrs old was generated and analyzed for single nucleotide polymorphisms (SNPs). Two F5 related SNPs (rs6025, factor V Leiden and rs6687813) exceeded significance (FDR (false discovery rate) p < 0.05). No other variants met genome-wide significance. When the data for the subgroup of cases with recurrent VTE that are more likely to have genetic risk factors than cases with a single VTE episode were compared to controls (N=211 controls and N=32 recurrent VTE cases), 28 SNPs, including the F5 rs6025 SNP, achieved significance (FDR p < 0.05).

Low paternal dietary folate alters the sperm epigenome and is associated with negative pregnancy outcomes

Project description:Epidemiological studies suggest that a fatherâ€™s diet can influence offspring health. A proposed mechanism for paternal transmission of environmental information is via the sperm epigenome. The epigenome includes heritable information such as DNA methylation. We hypothesized that the dietary supply of methyl donors would alter epigenetic reprogramming in sperm. This hypothesis was examined by feeding male mice a folate deficient (FD) and folate sufficient (FS) diets and then generating and analyzing DNA methylation profiles of their sperm.
