Physical Activities by Technology Help (PATH)

ABSTRACT: PATH is a research study for cancer survivors to help participants to become more active. Studies suggested an association between inactivity and cancer. The investigators created new novel ways and technologies that may help participants to become more active. The three methods the investigators are studying are: 1) participant become active on her/his own; educational material will be provided; 2) working with a programmed health coach over the phone via text messages; and 3) using digital voice assist to help participant become more active. The digital voice assist will be delivered via Amazon Alexa on Echo speaker (it is the famous intelligent voice that you see in superball commercial by Alec Baldwin). This study is funded by the State of Maryland.

DNA methylation profiling by HELP-seq in Dnmt1+/+ mouse embryonic fibroblasts

Project description:DNA methylation and the Polycomb Repression System are epigenetic mechanisms that play important roles in maintaining transcriptional repression. Recent evidence suggests that DNA methylation can attenuate the binding of Polycomb protein components to chromatin and thus plays a role in determining their genomic targeting. However, whether this role of DNA methylation is important in the context of transcriptional regulation is unclear. By genome-wide mapping of the Polycomb Repressive Complex 2 (PRC2)-signature histone mark, H3K27me3, in severely DNA hypomethylated mouse somatic cells, we show that hypomethylation leads to widespread H3K27me3 redistribution, in a manner that reflects the local DNA methylation status in wild-type cells. Unexpectedly, we observe striking loss of H3K27me3 and PRC2 from Polycomb-target gene promoters in DNA hypomethylated cells, including Hox gene clusters. Importantly, we show that many of these genes become ectopically expressed in DNA hypomethylated cells, consistent with loss of Polycomb-mediated repression. An intact DNA methylome is required for appropriate Polycomb-mediated gene repression by constraining PRC2 targeting. These observations identify a previously unappreciated role for DNA methylation in gene regulation and therefore influence our understanding of how this epigenetic mechanism contributes to normal development and disease. CD4 + T-cell help explains clinical benefit of specific intra-tumoral dendritic cell states in human cancer

Project description:Classical type 1 dendritic cells (cDC1) have a low abundance in the tumor microenvironment (TME), but positively correlate with patient survival. We hypothesized that the ability of cDC1 to relay CD4+ T-cell help for the cytotoxic T lymphocyte (CTL) response explains their clinical benefit. In vitro, contact with activated CD4+ T-cells enabled human cDC1, but no other DC types, to induce a CTL response to cell-associated tumor antigens. Single cell transcriptomics revealed that CD4+ T-cell help uniquely optimized cDC1 and no other DC types in many functions that support antigen cross-presentation and T-cell priming. We robustly identified “helped” cDC1 in a multitude of human cancer types by the overlap in their transcriptomic signature with that of recently defined, positively prognostic tumor-infiltrating DC states. As predicted from the functional effects of CD4+ T-cell help, the transcriptomic signature of “helped” cDC1 correlated with tumor infiltration by CTL and Thelper(h)-1 cells, overall survival and response to PD-1-targeting immunotherapy. Transcriptional basis of CD4 T cel help for memory CD8 T cell formation

Project description:CD4+ T-cell help is required for the generation of CD8+ cytotoxic T lymphocyte (CTL) memory. We here reveal how “help” signals delivered during priming impact memory differentiation of CTLs, as informed by genome-wide analyses. “Help” signals promoted the IL-15-dependent maintenance of central memory (TCM) cells. However, they had a much larger impact on the generation of effector memory (TEM) cells and their gene expression program . CD4+ T-cell help created TEM cells that produced Granzyme B and IFNg after antigen-independent recall with IL-12 and IL-18. Furthermore, “helped” memory CTLs expressed the effector program characteristic of “helped” primary CTLs upon recall with MHC class I-restricted antigen only, due to both epigenetic imprinting and sustained mRNA expression of relevant genes . Thus, CD4+ T-cell help delivered during priming creates CD8+ TEM cells with innate and help-independent recall capacities.

CD4 help is required for the generation of a transcriptionally distinct cytolytic CD8 T cell subset that arises during chronic viral infection

Project description:CD4 help is crucial to sustain the function of exhausted CD8 T cells, yet the detailed mechanisms by which CD4 help regulates CD8 T cell differentiation and function during chronic viral infection remain elusive. Here, using single cell RNA-sequencing (scRNA-seq) we show that CD8 T cells responding to chronic viral infection are more heterogeneous than previously appreciated, with three transcriptionally distinct subsets dominating the late phase of the antiviral response. Importantly, our findings uncover the formation of a previously unrecognized CD8 T cell subset that exhibits potent cytolytic function. This subset is characterized by high expression of killer cell lectin-like receptors Klre1 and Klra9, the chemokine receptor CX3CR1, and the transcription factors T-bet and Zeb2. Notably, our data further demonstrate that the formation of this cytotoxic subset is critically dependent on CD4 help and that exploitation of this developmental pathway may be used therapeutically to enhance the killer function of CD8 T cells infiltrated into the tumor. These findings uncover novel cellular and molecular mechanisms of how CD4 help regulates CD8 T cell differentiation during persistent infection and have implications towards optimizing the generation of protective CD8 T cells in immunotherapy.

Induction of autonomous memory alveolar macrophages requires T-cell help and is critical to trained immunity

Project description:Innate immune memory is a new important area of research. However, innate immune memory at the major mucosal sites remains poorly understood. Here we show that respiratory viral infection induces long-lasting memory alveolar macrophages (AM). Memory AM are programed to express high MHC II, carry a defense-ready gene signature, and produce, upon re-stimulation, neutrophil chemokines. Using a multitude of approaches, we reveal that the priming, but not maintenance, of memory AM requires the help from effector CD8 T cells. T cells jump-start this process in AM via IFN-γ production. We further find that formation/maintenance of memory AM are independent of monocytes or bone marrow progenitors. Finally, we demonstrate that memory AM are poised for robust trained immunity against bacterial infection in the lung via rapid induction of chemokines and neutrophilia. Our study thus establishes a new paradigm of immunological memory formation whereby adaptive T-lymphocytes render innate memory of mucosal-associated macrophages.
