Whole Blood Gene Expression Analysis of Controls and CRC Patients

Project description:Colorectal cancer is the third most common malignancy and the fourth most common cause of cancer mortality worldwide. In 2008, more than one million cases were newly diagnosed, and more than 600,000 people died from the disease. Given its slow development from removable precancerous lesions and curable early stages, screening for CRC has the potential to reduce both the incidence and mortality of the disease. However, compliance with current screening methods remains poor and there is a clear need for an accurate in vitro blood test to increase participation in colorectal cancer screening. In this study, we performed genome-wide gene expression profiling of peripheral blood samples from 100 healthy controls and 100 colorectal cancer patients using PAXgeneTM technology and Affymetrix GeneChip® microarrays. We show that monitoring gene expression in blood results in distinct transcriptional profiles between the controls and cancer patients. Thus, the microarray-based blood gene expression profiling holds great promise for developing novel biomarkers for colorectal cancer detection.

Comparison of Apis mellifera workers and queens brain development at fourth instar larvae

Project description:In Apis mellifera, the female eggs can develop into workers or queen depending on the diet offered during early development. The outputs of the developed honeybee females are two morphs with particular morphological traits and related physiology. The differential feeding regime experienced by the queen and the worker larvae of the honeybee Apis mellifera shapes a complex endocrine response cascade that ultimately sets up differences in brain morphologies. Herein we report on aspects of brain morphogenesis during larval development and the brain gene expression signature of fourth instar larvae (L4) of both castes, a developmental stage characterized by the greatest differences in juvenile hormone (JH) titers between castes Using results from the hybridization of whole genome-based oligonucleotide arrays with RNA samples from brain of fourth instar larvae honeybees of both castes we present a list of differentially expressed genes. Analysis used one dye-swap combination to compare workers and queens brain development at fourth instar larvae when juvenile hormone titers is higher in queens.

Project description:Transcriptional profiling of COLO 320 xenograft tumor cells comparing control COLO 320 xenograft without co-implanted rat MSCs with COLO 320 xenograft with co-implanted rat MSCs. The latter makes co-implanted MSCs visualization possible by using MSCs labeled by GFP under FACS and single cell microscopy. Two-condition experiment, COLO 320 xenograft without rat MSCs [COLO320 MSC(-)] vs. COLO 320 xenograft with rat MSCs [COLO320 MSC(+)]. Biological replicates: 1 control, 1 sample, paired xengraft tumor cells grown and harvested from the same mouse host. One replicate per array.

A new antibiotic from an uncultured bacterium binds to an immutable target

Project description:Antimicrobial resistance is a leading mortality factor worldwide. Here we report the discovery of clovibactin, a new antibiotic, isolated from uncultured soil bacteria. Clovibactin efficiently kills drug-resistant Gram-positivebacterial pathogens without detectable resistance. Using biochemical assays,solid-state NMR, and atomic force microscopy, we dissect its mode of action. Clovibactin blocks cell wall synthesis by targeting pyrophosphate of multiple essential peptidoglycan precursors (C55PP, Lipid II, LipidWTA). Clovibactin uses anunusual hydrophobic interface to tightly wrap aroundpyrophosphate, butbypasses the variable structural elements of precursors, accounting for the lack of resistance. Selective and efficient target binding is achieved by the sequestration of precursors into supramolecular fibrils that only form on bacterial membranes that contain lipid-anchored pyrophosphate groups.Uncultured bacteria offer a rich reservoir of antibiotics with new mechanisms of action that could replenish the antimicrobial discovery pipeline.
