The French E3N Prospective Cohort Study

ABSTRACT: The French E3N cohort was initiated in 1990 to investigate the risk factors associated with cancer and other major non-communicable diseases in women. The participants were insured through a national health system that primarily covered teachers, and were enrolled from 1990 after returning baseline self-administered questionnaires and providing informed consent. The cohort comprised nearly 100 000 women with baseline ages ranging from 40 to 65 years. Follow-up questionnaires were sent approximately every 2-3 years after the baseline and addressed general and lifestyle characteristics together with medical events (cancer, cardiovascular diseases, diabetes, depression, fractures and asthma, among others). The follow-up questionnaire response rate remained stable at approximately 80%. A biological material bank was generated and included blood samples collected from 25 000 women and saliva samples from an additional 47 000 women. Ageing among the E3N cohort provided the opportunity to investigate factors related to agerelated diseases and conditions as well as disease survival.

Gene expression changes in adipose tissue with diet- and/or exercise-induced weight loss

Project description:An ancillary study within a randomized trial of diet, exercise, or combined diet+exercise vs. control among overweight/obese postmenopausal women. Subcutaneous adipose-tissue biopsies were performed at baseline and after 6 months and changes in adipose-tissue gene expression were determined by microarray with an emphasis on pre-specified candidate pathways, as well as by unsupervised clustering. Analyses were conducted first by randomization group, and then by degree of weight change at 6-months in all women combined. Total RNA was obtained from subcutaneous adipose tissue biopsies at baseline and 6 months. A total of 47 women were biopsied and one replicate participant was included for a total of 96 samples.

Solar exposed skin induces sex-dependent food-seeking behavior

Project description:Considering the distinct physiologies of men and women, it stands to reason that they would react differently to solar exposure, but such a study was never conducted before. Here we show that solar exposure induces food-seeking behavior, food intake and weight gain in males, but not in females, by epidemiological analysis, blood-serum proteomics, UVB-exposed mouse behavioral models and human cohort questionnaires . The underlying mechanism entails increased ghrelin secretion from skin adipocytes into the circulation. UVB irradiation led to p53 transcriptional activation of ghrelin in skin adipocytes, with mouse conditional p53-knockout abolishing UVB-induced ghrelin expression and food-seeking behavior. In females, estrogen interferes with the p53–chromatin interaction on the ghrelin promotor, thus blocking ghrelin and, consequently, food-seeking behavior in response to UVB exposure. These results identify the skin as a major mediator of human physiology in furless animals and may lead to therapeutic opportunities for sex-based treatment of endocrine-related diseases.

The Illumina Infinium MethylationEPIC array was used to obtain DNA methylation profiles across approximately 850,000 CpGs. Samples included 470 cord blood samples from infants born to women in the Southampton Women's Survery (SWS) cohort, to examine the association between DNA methylation in the infant and aspects of health and disease in early and life and childhood.

Expression data from healthy postmenopausal women on 4 different types of hormone therapy, the RET study

Project description:Postmenopausal hormone therapy (HT) is associated with many diseases and conditions, but the underlying molecular mechanisms involved are incompletely understood. The aim of the current study was to investigate the effect of 4 types of HT on gene transcription. 24 women (6 women in 4 treatment groups) received 2 mg 17M-NM-2-estradiol combined with 1 mg noresthisterone acetate (NETA), 1 mg 17M-NM-2-estradiol combined with 0.5 mg NETA, tibolone, or raloxifene hydrochloride. RNA was isolated from whole blood before treatment (baseline) and after 6 weeks on treatment. The changes in mRNA from baseline to 6 weeks were assessed with a microarray chip. 4 treatment groups with 6 women in each group were blood sampled before treatment (baseline) and after 6 weeks on treatment, that is a total of 48 samples. The gene expression data at 6 weeks were compared to the expression data at baseline for each treatment.

A pilot proteomic study reveals different protein profiles related to testosterone and gonadotropin changes in a short-term controlled healthy human cohort

Project description:Testosterone deficiency afflicts approximately 30% of men aged from 40-89 years. It is associated with type 2 diabetes, metabolic syndrome and cardiovascular disease. However, the causality of the relationship between low testosterone and metabolic diseases is unclear. We have analysed plasma samples from thirty healthy male volunteers (19-32 years of age) with pharmacologically-induced testosterone deficiency. Blood samples were collected under three conditions: normal testosterone (baseline), ‘zero’ testosterone and ‘restored’ testosterone. As a proof-of-concept, a sample pooling approach was used to obtain one pool per condition, with each pool consisting of 30 individual samples. The proteomic profiles identified significant functional differences, related to the innate immune system, the regulation of insuline-like growth factors, collagen degradation, glycolysis/gluconeogenesis process, among others.

Adult postmortem brain tissue (dorsolateral prefrontal cortex) from subjects with bipolar disorder and healthy controls

Project description:Bipolar affective disorder is a severe psychiatric disorder with a strong genetic component but unknown pathophysiology. We used microarray technology (Affymetrix HG-U133A GeneChips) to determine the expression of approximately 22 000 mRNA transcripts in post-mortem brain tissue (dorsolateral prefrontal cortex) from patients with bipolar disorder and matched healthy controls. A cohort of 70 subjects was investigated and the final analysis included 30 bipolar and 31 control subjects. Perfluorinated compounds and high fat diet in relation to CVD-relevant metabolomic pathways in the SEARCH for Diabetes in Youth study

Project description:This project aims to evaluate the association between environmental exposure to perfluorinated alkyl substances (PFCs) and the development of risk factors for cardiometabolic disease in youth diagnosed with diabetes in the SEARCH Cohort Study. The longitudinal study of newly diagnosed cases of type 1 and type 2 diabetes examines serum metabolome changes at baseline and follow-up at approximately 5 years (all >3 years from baseline). Exposures to PFCs and biological effects characterized by serum metabolome changes will be associated with known cardiometabolic risk factors in youth diagnosed with diabetes.
