Project description:Advanced paternal age has been shown to be a significant risk factor for neurodevelopmental psychiatric disorders, particularly autism. We have recently shown that mice conceived by old fathers display behavioral abnormalities which resemble key diagnostic symptoms of human autism. De novo mutations and epigenetic alterations increase in the male germ line during ageing and are thought to mediate the effect of paternal age on occurrence of diseases occurrence. Because the placenta carry a predominantly fetal genetic background, age-related mutagenesis and epigenetic errors might negatively influence placental physiology and in turn perturb fetal brain development. Here, we examined the impact of paternal age on placental mRNA transcriptome. Project description:Animal models provide opportunity to study neurobiological aspects of human alcoholism. Changes in gene expression have been implicated in mediating brain function, including reward system and addiction. The current study aimed to identify novel genes that may underlie ethanol preference. Microarray analysis comparing gene expression in nucleus accumbens (NAc), hippocampus (HP) and prefrontal medial cortex (mPFC) was performed in two rat strains selected for extreme levels of ethanol preference - Warsaw High Preferring (WHP) and Warsaw Low Preferring (WLP). The identified candidate genes may underlie differential ethanol preference in rat model of alcoholism. Project description:The PICCOLINO is a randomized health services study performed within the framework of the Polish Colonoscopy Screening Program (PCSP) in Poland.
Within the study 12,298 eligible persons between 55 and 64 years of age will be drawn from the Population Registry and randomly assigned in a 1:1:1 ratio to receive an invitation to participate in one of the three screening strategies: (I) postal invitation to colonoscopy and a re-invitation to colonoscopy for initial non-responders, (II) postal invitation for screening using fecal immunochemical test (FIT) for non-responders and subjects refusing a colonoscopy, or (III) postal invitation offering a choice between FIT and colonoscopy.
Colonoscopies will be performed in seven local centers participating in the study. FITs will be analyzed in the central laboratory. Subjects with positive FIT result will be recommended to undergo colonoscopy.
The primary outcome is participation with CRC screening within 18 weeks after enrollment, defined as completion of colonoscopy, or completion of FIT along with colonoscopy for positive FIT result. Secondary outcome will be diagnostic yield for advanced neoplasia (CRC or advanced adenoma). The study has been approved by a local bioethics committee.

Project description:Advanced Maternal Age (AMA) is a risk factor for neurological and neuropsychiatric disorders in offspring. We developed a mouse model to investigate whether pregnancy at advanced age may provoke behavioral and brain gene expression changes in offspring. Mice conceived by 15- to 18-month-old (15-18M) or 3M control females were delivered by cesarean section and were all fostered after birth by 3M dams. Genome-wide mRNA expression was analyzed using microarrays in the hippocampus of 4M male offspring. In conclusion, pregnancy at advanced age yields offspring with altered patterns of gene expression in the hippocampus, typical of human neuropsychiatric and neurodegenerative disorders. The effects were not reversed by the postnatal maternal care provided by young foster mothers. This suggests that prenatal conditions associated with AMA may negatively affect fetal brain development and hence postnatal behaviors. We included 2 comparisons: 1) placenta of female APA vs placenta of female control; 2) placenta of male APA vs placenta of male control. Each comparison was composed of 3 biological replicates. To minimize family bias, poolings contained at most one placenta per sex from each dam to a minimum of one and a maximum of three placentas per group/sex.

Project description:Smoking cessation is the most effective measure for reducing the risk of smoking-related diseases but switching to less harmful products (modified-risk tobacco products) can be an alternative for smokers who would otherwise not quit. In an 18-month chronic carcinogenicity/toxicity study in A/J mice (OECD Test Guideline 453), we assessed aerosol from the Tobacco Heating System 2.2 (THS 2.2), a candidate modified-risk tobacco product based on the heat-not-burn principle, compared with 3R4F cigarette smoke (CS). To capture toxicity- and disease-relevant mechanisms, we complemented standard toxicology endpoints with in-depth systems toxicology analyses. Briefly, female A/J mice were exposed to fresh air (Sham), three concentrations of THS 2.2 aerosol corresponding to nicotine concentrations of 6.7 (Low; L), 13.4 (Medium; M), and 26.8 (High; H) µg nicotine\L test atmosphere, or to one concentration of 3R4F CS (13.4 µg nicotine\L test atmosphere) in whole-body exposure chambers for 6 h per day, 5 days per week, and up to 18 months. Interim dissections were scheduled for months 1, 5, and 10. Male mice were exposed to either fresh air (Sham) or to the high THS 2.2 aerosol concentration for 15 months. Chronic toxicity and carcinogenicity endpoints outlined in the OECD protocol were assessed; detailed findings will be reported separately (Wong et al.). For evaluation of non-OECD endpoints following a systems toxicology approach (proteomics, transcriptomics, and genomics), several tissues were collected for further analysis. Housing and all procedures involving animals were performed in accordance with the approved Institutional Animal Care and Use Committee protocol in an Agri-Food & Veterinary Authority of Singapore-licensed and Association for Assessment and Accreditation of Laboratory Animal Care International-accredited facility, where the care and use of the animals for scientific purposes were in accordance with the National Advisory Committee for Laboratory Animal Research (NACLAR) Guideline (NACLAR, 2004). Animals allocated to the omics endpoints (N=8-20) were dissected within 16-24 h of the last exposure and following randomization of all planned necropsies for the dissection time point in question. All efforts to minimize potential nucleic acid and protein degradation were made, and samples were frozen as rapidly as possible once ex vivo. Lungs collected after month 1 were snap-frozen and the right caudal lobe was used for proteomics analysis. These are the protein expression data for the respiratory nasal epithelium assessed by iTRAQ®-based quantitative proteomics.
