Project description:The TP53 c.359A>G mutation severely impairs expression of the major TP53 transcript variant encoding p53 K120R by creating a new splicing donor site. Antisense morpholino oligomer (AMO) targeting the mutation site restored normal splicing and expression of the major variant. Despite the recovery of mutant TP53 mRNA levels, the functional restoration as the tumor suppressor of p53 K120R was unclear. Given that p53 exerts its tumor suppressor function through regulation of target genes that mediate growth arrest or apoptosis, the p53 K120R mutant exhibits impaired transcriptional regulation of CDKN1A, a key target gene in growth arrest, while maintaining normal induction of the pro-apoptotic BBC3 gene. Consequently, the p53 K120R mutant protein demonstrates a phenotype characterized by defective cell growth arrest but retained apoptotic induction capacity, suggesting that the p53 K120R mutant protein may retain some tumor suppressor functionality. In addition, Lysine 120 provides a critical acetylation site linked to p53 activation. Although other studies of the oncogenic effects of p53 K120R highlight the relevance of acetylation for tumor suppression, our findings indicate that targeting mutant TP53 mRNA is a prerequisite for restoration of p53 function. In particular, despite the presence of mutations, it showed sufficient corrective effect to restore p53 functions, including apoptosis induction. In conclusion, this study highlights the potential of AMO-mediated splice correction as a therapeutic approach for TP53 mutations.

Project description:Deletion of the short arm of chromosome 17 (17p-) is one of the most critical genetic variants used in B-CLL risk stratification. The tumor suppressor TP53 maps to this region, and its loss or mutation significantly accelerates B-CLL progression, hampers response to chemotherapy, and shortens survival. While florescent in situ hybridization (FISH) analyses for 17p deletions are routinely performed during clinical diagnoses, mutational analyses of the TP53 gene is not widely available and thus its mutational status is often unknown in patients with CLL. Given the limited clinical data that exists for frontline treatment of patients with CLL harboring TP53 mutations, there is a great need to identify novel treatment strategies for this subset of patients. Herein, we use a CLL mouse model (Eμ-TCL1) in the presence or absence of a common TP53 hot-spot mutation (p53R172H, corresponding to p53R175H in humans) to study its impact on disease progression, survival, response to therapy, and dynamic loss of the remaining wild-type Trp53 allele during the course of B-CLL following BTK inhibitor (ibrutinib). We show that ibrutinib was effective in increasing survival and activated gene and cellular programs outside of the p53 pathway in both settings, and thus, did not place selective pressure on the remaining wild type Trp53 allele. These data demonstrate the potent effect of BTK-inhibition in B-CLL and more importantly, provide evidence that ibrutinib acts as an effective treatment for aggressive forms of B-CLL with TP53 mutation and potentially chemo-resistant refractory disease.

Project description:To screen the related miRNAs in the occurence and development of sebaceous gland carcinoma(SGC) of the eyelid , we have employed Human Agilient microarray expression profiling as a discovery platform to identify differentially expressed miRNAs for SGC and TP53 mtation related miRNAs. Human formalin-fixed paraffin-embedded (FFPE) SGC tissues and para-carcinoma sebaceous gland(SG) FFPE samples from SGC patients were screened using microarray assay. Human FFPE SGC tissues containing TP53 mutation and human FFPE SGC tissues without TP53 mutation were screened using microarray. The overlap of two sets of microarray differentially upregulated miRNAs were selected as our research object.

Project description:While mutations in the KRAS oncogene are amongst the most prevalent in human cancer, there are few successful treatments to target these tumors. It is also likely that heterogeneity in KRAS-mutant tumor biology significantly contributes to the response to therapy. We hypothesized that presence of commonly co-occurring mutations in STK11 and TP53 tumor suppressors may represent a significant source of heterogeneity in KRAS-mutant tumors. To address this, we utilized a large cohort of resected tumors from 442 lung adenocarcinoma patients with data including annotation of prevalent driver mutations (KRAS, EGFR) and tumor suppressor mutations (STK11 and TP53), microarray-based gene expression and clinical covariates including overall survival (OS). Specifically, we determined impact of STK11 and TP53 mutations on a new KRAS mutation-associated gene expression signature as well as previously defined signatures of tumor cell proliferation and immune surveillance responses. Interestingly, STK11, but not TP53 mutations, were associated with highly elevated expression of KRAS mutation-associated genes. Mutations in TP53 and STK11 also impacted tumor biology regardless of KRAS status, with TP53 strongly associated with enhanced proliferation and STK11 with suppression of immune surveillance. These findings illustrate the remarkably distinct ways through which tumor suppressor mutations may contribute to heterogeneity in KRAS-mutant tumor biology. In addition, these studies point to novel associations between gene mutations and immune surveillance that could impact the response to immunotherapy.

Project description:The TP53 gene is frequently mutated in human cancer. Research has focused predominantly on six major “hotspot” codons, accounting for only ~30% of cancer-associated p53 mutations. To comprehensively characterize the consequences of the p53 mutation spectrum, we created a synthetically designed library and measured the functional impact of ~10,000 DNA-binding domain (DBD) p53 variants in human cells in culture and in vivo. Our results highlight the differential outcome of distinct p53 mutations in human patients and elucidate the selective pressure driving p53 conservation throughout evolution. Furthermore, while loss of anti-proliferative functionality largely correlates with the occurrence of cancer-associated p53 mutations, we observe that selective gain-of-function may further favor particular mutants in vivo. Finally, when combined with additional acquired p53 mutations, seemingly neutral TP53 SNPs may modulate phenotypic outcome and presumably tumor progression.

Project description:The physiological effects of the many germline mutations of TP53, encoding the tumor suppressor protein p53, are poorly understood. Here, we report generating a p53 R178C knockin mouse modeling the human TP53 R181C mutation that is notable for its prevalence and prior molecular characterization. Consistent with its weak cancer penetrance in humans, homozygous p53178C/C mice showed a modest increase in tumorigenesis but, surprisingly, were lean with decreased body fat content. They displayed evidence of increased lipolysis and upregulation of fatty acid metabolism in their inguinal white adipose tissue (iWAT). Gene expression and chromatin immunoprecipitation sequencing (ChIP-Seq) analyses showed that the mutant p53 bound and transactivated Beta-3-Adrenergic Receptor (ADRB3), a gene that is known to promote lipolysis and is associated with obesity. The current study reveals that a germline mutation of p53 can affect fat metabolism which can impact cancer development.

Project description:Multiple family members with cancer or individuals with multiple primary cancers are indicative of potential genetic etiology1. Germline mutations in TP53 cause a rare high penetrance cancer syndrome, Li Fraumeni Syndrome (LFS)2. We identified a TP53 tetramerization domain (TD) missense mutation c.1000G>C;p.G334R, in a family with LFS-associated cancers. Twenty-one additional probands were identified, and available tumors showed biallelic somatic inactivation of TP53. The majority of families were of Ashkenazi Jewish descent, and the TP53 c.1000G>C allele was found on a commonly inherited haplotype. While classical p53 target gene activation was maintained in p.G334R mutant cell lines treated with Nutlin-3a, a subset of p53 target genes, including PCLO, PLTP, PLXNB3 and LCN15, showed defective transactivation. Structural analysis demonstrated thermal instability of the mutant TD, and the G334R mutant protein showed increased preponderance of mutant conformation protein. Project description:Heterozygous p53-R280T mutations frequently occur in many nasopharyngeal carcinoma cell lines and nasopharyngeal carcinoma patients. However, the role of this mutation in the progression of nasopharyngeal carcinoma remains unclear. In this study, we successfully generated the tp53 knockout nasopharyngeal carcinoma (NPC) cells by CRISPR/Cas9-mediated genome editing and found that knockout of heterozygous tp53-R280T inhibited the proliferation of NPC cells significantly in vivo and in vitro. Mechanistic analyses indicated that heterozygous p53-R280T can activate the PI3K/Akt Signaling pathway in NPC cells. In conclusion, our findings provide a mechanistic insight into the role of heterozygous p53-R280T in NPC progression.
