Traditional Chinese medicine (TCM) for cancer

ABSTRACT: Study hypothesis: To examine the feasibility of conducting a large Canadian trial aimed at assessing the effectiveness of traditional Chinese herbal medicine (with specially tailored formulas) and Tai Chi on improving cancer patients’ quality of life (QoL). Hypothesis: Patients receiving Chinese herbal medicine or Tai Chi will have more energy, more appetite and more overall quality of life than patients exposed to placebo or light physical exercise. Please note that as of 04/03/2009 the anticipated start and end dates in this record were amended. The previous dates are as follows: Initial anticipated start date: 30/06/2008 Initial anticipated end date: 30/12/2008 Please also note that at this time, the disease was amended from non-small cell lung cancer to colorectal cancer. Primary outcome: The feasibility as assessed through several factors, such as co-interventions, percentage of participation and withdrawal/non-compliance, preparing decoction/placebo and keeping blinding, the possibility to stratify, reasons for not participating, male/female difference regarding compliance and participation. Primary clinical outcome: Quality of life Primary outcome measuring time point: baseline, monthly, three months post-intervention

Project description:As a traditional martial art in China, Tai Chi Chuan has excellent health benefits along with its combat function.Studies have shown that Tai Chi as an exercise prescription can significantly reduce the incidence of cancer, but the complexity of Tai Chi Chuan routines, poor disease targeting, and the lack of traditional internal mental and physical training method limit the healing of tumours and other diseases. For this reason, we have established the Medical Tai Chi Exercise Healing System（MTCEH）, which integrates Tai Chi exercise, traditional Chinese internal mental and physical training method, rehabilitation medicine and sports medicine methods, on the basis of evidence-based medicine and with the aim of targeting different tumour treatments. With the characteristics of easily learning, internal and external training, and individualised treatment of tumours, this series of routines effectively combines traditional oriental Tai Chi martial arts with tumour treatment, which opens up a beautiful and mysterious oriental healing journey for integrative medicine.

Project description:Cocksfoot grass (Dactylis glomerata) collected from Wytham, Oxford, UK, was tested for Cocksfoot streak virus infection. Small RNA of the grass was extracted and converted to DNA according to Ho, T., et al. (2008) Biochem Biophys Res Commun. 368:433-7, with primers modified to contain 454 adapter nucleotide sequences. ATAC-seq on endocardial cells from E12.5 mouse hearts in Tbx20 knockouts and wild type counterparts

Project description:We used ATAC-seq (Buenrostro et al, 2013) to identify regions of open chromatin in FACS sorted mouse endothelial cells from E12.5 hearts. Please note that the animals were injected at different dates, which resulted in different effects of the knockout, as indicated in the batch attribute of the samples. The Taiman transcriptional coactivator engages Toll signals to promote apoptosis and intertissue invasion in Drosophila

Project description:The Drosophila Taiman (Tai) protein is homologous to the human steroid-receptor coactivators SRC1-3 and activates transcription in complex with the 20-hydroxyecdysone (20E) receptor (EcR). Tai has roles in intestinal homeostasis, germline maintenance, cell motility and proliferation through interactions with EcR and the coactivator Yorkie (Yki). Tai also promotes invasion of tumor cells in adjacent organs, but this pro-invasive mechanism is undefined. Here we show that Tai expression transforms sessile pupal wing cells into an invasive mass that penetrates the adjacent thorax during a period of high 20E. Candidate analysis confirms a reliance on elements of the 20E and Hippo pathways, such as Yki and the Yki-Tai target dilp8. Screening the Tai-induced wing transcriptome detects enrichment for innate immune factors, including the Spätzle (Spz) family of secreted Toll ligands that induce apoptosis during cell competition. Tai-expressing wing cells induce immune signaling and apoptosis among adjacent thoracic cells, and genetic reduction of spz, Toll or the rpr/hid/grim pro-apoptotic factors each suppresses invasion, suggesting an intercellular Spz-Toll circuit supports killing-mediated invasion. Modeling these interactions in larval epithelia confirms that Tai kills neighboring cells via a mechanism involving Toll, Spz factors, and the Spz-inhibitor Necrotic. Tai-expressing cells evade death signals by repressing the immune deficiency (IMD) pathway, which operates in parallel to Toll to control NFkB activity, and independently regulates JNK activity. In sum, these findings suggest that Tai promotes competitive cell killing via Spz-Toll, and that this killing mechanism supports pathologic intertissue invasion in Drosophila.

Promoter capture mESC and fetal liver

Project description:Genome organization influences transcriptional regulation by facilitating interactions between gene promoters and distal regulatory elements. To analyse distal promoter contacts we used Capture Hi-C (CHi-C) to enrich for promoter-interactions in HiC libraries from mouse ESC and E14.5 fetal liver. Please note additional files included. These files were created using the following protocol: Significantly interacting regions were called using the GOTHiC BioConductor package (http://www.bioconductor.org/packages/devel/bioc/html/GOTHiC.html) as described in (Mifsud et al.).<br>Update on December 2015: the original additional file E-MTAB-2414.additional.1.zip contained an earlier iteration of processed data and not the one that was used for the published paper. The Ecdysone Receptor Coactivator Taiman Links Yorkie to Transcriptional Control of Germline Stem Cell Factors in Somatic Tissue

Project description:The Hippo pathway is a conserved signaling cas- cade that modulates tissue growth. Although its core elements are well defined, factors modulating Hippo transcriptional outputs remain elusive. Here we show that components of the steroid-responsive ecdysone (Ec) pathway modulate Hippo transcrip- tional effects in imaginal disc cells. The Ec receptor coactivator Taiman (Tai) interacts with the Hippo transcriptional coactivator Yorkie (Yki) and promotes expression of canonical Yki-responsive genes. Tai enhances Yki-driven growth, while Tai loss, or a form of Tai unable to bind Yki, suppresses Yki-driven tissue growth. This growth suppression is not corre- lated with impaired induction of canonical Hippo- responsive genes but with suppression of a distinct pro-growth program of Yki-induced/Tai-dependent genes, including the germline stem cell factors nanos and piwi. These data reveal Hippo/Ec pathway crosstalk in the form a Yki-Tai complex that collabo- ratively induces germline genes as part of a tran- scriptional program that is normally repressed in developing somatic epithelia.

Sexual dimorphism of gut microbiota dictates therapeutics efficacy of radiation injuries [RNA-seq]

Project description:Accidental or iatrogenic ionizing radiation exposure precipitates acute and chronic radiation injuries. Traditional paradigm of mitigating radiotherapy-associated side effects has ignored the gender-specific dimorphism of patients. Here we examined the effects of sexual dimorphism on therapeutic agent efficiencies in murine models. High-throughput sequencing of host mRNA showed that different treatments reprogrammed the spectrum of mRNA expression in small intestines of male or female mice, respectively. Collectively, our observations demonstrate that therapeutic strategy efficiencies for radiation toxicity might be dependent on the gender of patients. We aim to uncover the underlying mechanisms by which simvastatin or high fat diet fights against radiotherapy-induced gastrointestinal tract toxicity for male or female mice. The mice were exposed to 12 Gy total abdominal irradiation (TAI), then treated with simvastatin for male mice or fed with high fat diet for female mice. The small intestine tissues were extracted from male mice without TAI, exposed to TAI only, and exposed to TAI combined with simvastatin treatment as one cohort, and from female mice without TAI, exposed to TAI only, and exposed to TAI combined with high fat diet feeding as the other cohort.
