Liver Image Guided High Dose Radiation Therapy for the Treatment of Colorectal Liver Metastases

ABSTRACT: Interventions: Stereotactic Radiation; Six fractions of radiotherapy delivered with 2-3 fractions per week at least one fraction 24 hours apart per week with risk adjusted dosing based on mean liver dose; 1. 60Gy in 6# delivered over 2-3 weeks for mean liver dose < 13.3Gy 2. 51Gy in 6# delivered over 2-3 weeks for mean liver dose 13.3-15.0Gy 3. 42Gy in 6# delivered over 2-3 weeks for mean liver dose 15.1-17.5Gy Primary outcome(s): To determine that delivery of high dose stereotactic image guided radiotherapy for colo-rectal liver metastases can be performed in a multidisciplinary setting. Successful delivery of stereotactic image guided radiotherapy for liver cancer will be indicated by 0 major clinical violations and <4 minor clinical violations on the planning review. [3 months post treatment ] Study Design: Purpose: Treatment; Allocation: Non-randomised trial; Masking: Open (masking not used);Assignment: Single group;Type of endpoint: Safety

Project description:Patients with cervical cancer were treated with external beam radiotherapy (EBRT) at a dose of 45Gy in 25 fractions over five weeks, with weekly cisplatin. Biopsies were taken pre-, during and post-treatment and analysed for copy number changes. Cervical cancer patients were treated with chemoradiotherapy and disease response was followed by array CGH profiling.

Project description:We report whole transcriptome sequencing of the intraventricular septum of mouse heart 2 and 6 weeks after 25 Gy ionizing radiation. The objective of this experiment was to identify genes and pathways differentially regulated by ionizing radiation in the heart. Image-guided isocentric gamma irradiation was delivered to experimental groups under isoflurane anesthesia in a single, 25 Gy dose over approximately 15 minutes. Control group animals were experimental group littermates which received anesthesia and CT imaging but no radiation. RNA was isolated from intraventricular septum using TRIzol.

Gene expression profile of mouse Lewis lung carcinoma cell line treated with single or fractionated ionizing radiation doses

Project description:Radiotherapy is one of the most common therapies for cancer. Approximately half of all cancer patients will receive radiotherapy at some point during treatment. Consequences of IR treatment are dose dependent and different sensitivity to IR of various types of cells is well established. To reduce the damage of IR to most sensitive cells of normal (noncancerous) tissue radiotherapy is administered as fractionated dose treatment applying radiation in ~2 Gy fractions every 24 hours, 5 times per week. However, during the therapy intrinsic and acquired tumor radioresistance may result in treatment failures. Comprehensive mechanisms of the resistance to irradiation as well as mechanisms of cellular response to fractionated dose IR remain unclear. Therefore, in the present study we evaluated global gene expression changes in murine Lewis lung carcinoma LLC1 cells following X-ray irradiation of single 2 Gy or 10 Gy and 2 Gy x 5 fractionated doses. Total RNA was harvested from mouse Lewis lung carcinoma cells 4h after treatment of single (2 Gy or 10 Gy) or fractionated (5x2 Gy) ionizing radiation dose.

Project description:Data of 100 colorectal cancer patients with liver metastases who received stereotactic radiotherapy of Cyberknife or microwave ablation in the multicenter of the research group from June 2019 to May 2021 were collected, as well as follow-up data.To evaluate the clinical efficacy of stereotactic radiotherapy and microwave ablation in liver metastases.In addition, the local control rate and side effects of stereotactic radiotherapy and microwave ablation in the treatment of liver metastases were explored, and the efficacy and safety of different doses of stereotactic radiotherapy were determined.

Project description:Radiotherapy remains a standard treatment for colorectal cancer. The clinical significance of the conventional treatment is based on accumulated dose and usually consists of single daily irradiations of 1.8 - 2 Gy fractions. However the repopulation of cancer cells during and/or after the treatment suggests that the surviving fraction of cells acquire the radioresistance to cancer therapy. It has been determined a number of radioresistance markers, but only limited data exist on the acquisition of the resistance to fractionated radiation therapy. This research provides the genomic data of gene expression changes in colorectal cancer cell HCT116 line following single and multiple fractions of irradiation, aiming to identify particular genes associated with the effect of fractionated radiotherapy.
