Project description:Adequate bowel cleansing is important for a completeness of colonoscopy and detection of colon polyps. Inadequate bowel preparation leads to longer duration of colonoscopy and obscured mucosal visualization resulting in missed lesions. Bowel cleansing agents are simply classified into the large volume, iso-osmotic polyethylene glycol (PEG) based solutions or the small volume osmotically active agents, such as sodium picosulphate with magnesium citrate (SPMC). There are rare reports that compare directly conventional polyethylene glycol (PEG) solution and sodium picosulphate with magnesium citrate (SPMC) for bowel preparation in korea. The aim of this study is to compare the efficacy, safety, and tolerability of different regimens of SPMC and PEG solution.

Proteomics of Extracellular Vesicles Harvested with ExtraPEG

Project description:Polyethylene glycol (PEG) is commonly used to isolate virus particles from culture medium. This project adapted and optimized the virus procedure for harvesting extracellular vesicles (of similar biophysical properties to viruses) from media. An 8% PEG solution harvested the purest vesicles. Followed by a wash step (ultracentrifugation of concentrated vesicles in saline), this method produced preparations comparable or superior to commonly used methods in terms of purity, abundance, and cost. It is also highly adaptable, allowing the procedure to be used for enriching vesicles from a variety of biological fluids. As proof of principle, extracellular vesicle protein was successfully harvested from HeLa cell medium using the described PEG-based harvest method and analyzed by mass spectrophotometry.

Arabidopsis thaliana peptide SP2 and ethyl acetate extraction clean up.

Project description:The success of bottom-up proteomic analysis frequently depends on the efficient removal of contaminants from protein or peptide samples before LC-MS/MS. For a peptide clean-up workflow, the single-pot solid-phase-enhanced peptide sample preparation on carboxylate-modified paramagnetic beads (termed SP2) was evaluated for sodium dodecyl sulfate or polyethylene glycol removal from Arabidopsis thaliana tryptic peptides. The robust and efficient 40-min SP2 protocol, tested for a 10 ng, 250ng and 10µg peptide sample, was proposed and benchmarked thoroughly against the ethyl acetate extraction protocol. The SP2 protocol on carboxylated magnetic beads proved to be the robust approach even for simultaneous removal of massive sodium dodecyl sulfate (SDS) and polyethylene glycol (PEG) contaminations from AT peptide samples in respect of the LC-MS/MS data outperforming ethyl acetate extraction.
