Project description:Interventions: Procedure/Surgery : 1) onestep knife conventional ESD (control) ? Insert the colonoscope with the transparent cap attached. ? After puncturing the submucosal layer immediately around the lesion with a puncture needle for submucosal solution injection, inject the saline-based submucosal solution. ? When the lesion is sufficiently raised, submucosal dissection is performed using a dual knife. ? Retrieve the resected specimen, fix it on a styrofoam fixing plate, and ask the pathology department for tissue analysis. (2) onestep knife + Traction ESD (intermediate group) ? Insert the colonoscope with the transparent cap attached. ? After puncturing the submucosal layer immediately around the lesion with a puncture needle for submucosal solution injection, inject the saline-based submucosal solution. ? When the lesion is sufficiently raised, submucosal dissection is performed using a dual knife. ? When performing ESD, make an incision around the lesion. After fixing the rubber band to the endoscope clip from the outside of the patient, insert it into the channel of the colonoscope and fix the clip at the end of the pre-incision lesion using the clip. Using the second clip, hold the rubber band secured to the pre-fastened first clip and secure the clip in a position that will provide adequate traction. ? Implement additional ESD. ? Transcriptomics of Rat Striata 16 h after Gamma Knife Surgery: Distinct Bilateral Effects in the Un-irradiated Striatum

Project description:Gamma knife surgery (GKS) is used for treatment of various brain disorders. The effects of gamma irradiation to targeted and un-targeted regions were evaluated by monitoring gene expression changes in the unilateral irradiated (60 Gy) and contralateral un-irradiated striata in the rat. Striata of irradiated and control brains were dissected 16 h post-irradiation for analysis by rat whole genome 44K DNA oligo microarray. Results revealed 230 induced and 144 repressed genes in the irradiated striatum and 432 induced and 239 repressed genes in the un-irradiated striatum. The number of altered genes in un-irradiated striatum was more than that in irradiated striatum. Results of RT-PCR and western analyses suggested that gamma-irradiation caused cellular damage, including oxidative stress, in both striata of both hemispheres. Our present results indicate that unilateral irradiation during GKS produce bilateral effects as early as 16 h, the time-period analyzed, and these molecular changes in the un-irradiated striatum are ample proof.

Dual roles of HSP70 chaperone HSPA1 in quality control of nascent and newly synthesized proteins

Project description:Exposure to heat stress triggers a well-defined acute response marked by HSF1-dependent transcriptional upregulation of heat shock proteins. Cells allowed to recover acquire thermotolerance, but this adaptation is poorly understood. By quantitative proteomics, we discovered selective upregulation of HSP70-family chaperone HSPA1 and its co-factors, HSPH1 and DNAJB1, in MCF7 breast cancer cells acquiring thermotolerance. HSPA1 was found to have dual function during heat stress response: (i) during acute stress, it promotes the recruitment of the 26S proteasome to translating ribosomes, thus poising cells for rapid protein degradation and resumption of protein synthesis upon recovery; (ii) during thermotolerance, HSPA1 together with HSPH1 maintains ubiquitylated nascent/newly synthesized proteins in a soluble state required for their efficient proteasomal clearance. Consistently, deletion of HSPH1 impedes thermotolerance and esophageal tumor growth in mice, thus providing a potential explanation for the poor prognosis of digestive tract cancers with low HSPH1, and nominating HSPH1 as a cancer drug target. We propose dual roles of HSPA1 either alone or in complex with HSPH1 and DNAJB1 in promoting quality control of nascent/newly synthesized proteins and cellular thermotolerance.
