Project description:Arabidopsis Col-0 seedlings grown in liquid culture to growth stage 1.0 were treated with 5 M-5M Benzyladenine and incubated for the indicated time points. Roots and shoots of the seedlings were then harvested separately by taking each individual seedling with a soft forceps at the hypocotyl, dipping it into liquid nitrogen and breaking off the root at the inner wall of a collection tube that was swimming in liquid nitrogen. The shoot part was then striped from the forceps at the edge of another collection tubes.

Project description:ObjectivesTo assess the accuracy of a real-time dynamic navigation system applied in zygomatic implant (ZI) surgery and summarize device-related negative events and their management.Material and methodsPatients who presented with severely maxillary atrophy or maxillary defects and received dynamic navigation-supported ZI surgery were included. The deviations of entry, exit, and angle were measured after image data fusion. A linear mixed-effects model was used. Statistical significance was defined as p < .05. Device-related negative events and their management were also recorded and analyzed.ResultsTwo hundred and thirty-one zygomatic implants (ZIs) with navigation-guided placement were planned in 74 consecutive patients between Jan 2015 and Aug 2020. Among them, 71 patients with 221 ZIs received navigation-guided surgery finally. The deviations in entry, exit, and angle were 1.57 ± 0.71 mm, 2.1 ± 0.94 mm and 2.68 ± 1.25 degrees, respectively. Significant differences were found in entry and exit deviation according to the number of ZIs in the zygomata (p = .03 and .00, respectively). Patients with atrophic maxillary or maxillary defects showed a significant difference in exit deviation (p = .01). A total of 28 device-related negative events occurred, and one resulted in 2 ZI failures due to implant malposition. The overall survival rate of ZIs was 98.64%, and the mean follow-up time was 24.11 months (Standard Deviation [SD]: 12.62).ConclusionsThe navigation-supported ZI implantation is an accurate and reliable surgical approach. However, relevant technical negative events in the navigation process are worthy of attention.

Project description:3D printed navigational templates have facilitated the accurate treatment of orthopaedic patients. However, during practical operation, it is found that the location hole occasionally deviates from the ideal channel. As such, there will be a security risk in clinical applications. The purpose of this study was to evaluate the influence of chemical-based sterilisation methods on the dimensional accuracy of different materials and the influence of module parameters on the degree of deformation. We found that polylactic (PLA) modules sterilised with ethylene oxide (EO) would undergo micro-deformation, and these micro-deformation characteristics depend on the building direction, i.e., the module stretches in the Z direction and shrinks in the X and Y directions. Heat-resisting polylactide (HR-PLA) has the same melting temperature (Tm) as PLA, but its glass transition temperature (Tg) is greater than the EO sterilisation temperature, so there is no obvious deformation after EO sterilisation. The layer height of the module were inversely proportional to the degree of deformation in the same sterilisation method. The deformation time of the module is concentrated within 2 h after heating. The micro-deformation of the 3D printing module depends on its Tg, sterilisation temperature, and duration of the sterilisation cycle.

Project description:According to Thorkild Ramskou's theory proposed in 1967, under overcast and foggy skies, Viking seafarers might have used skylight polarization analysed with special crystals called sunstones to determine the position of the invisible Sun. After finding the occluded Sun with sunstones, its elevation angle had to be measured and its shadow had to be projected onto the horizontal surface of a sun compass. According to Ramskou's theory, these sunstones might have been birefringent calcite or dichroic cordierite or tourmaline crystals working as polarizers. It has frequently been claimed that this method might have been suitable for navigation even in cloudy weather. This hypothesis has been accepted and frequently cited for decades without any experimental support. In this work, we determined the accuracy of this hypothetical sky-polarimetric Viking navigation for 1080 different sky situations characterized by solar elevation ? and cloudiness ?, the sky polarization patterns of which were measured by full-sky imaging polarimetry. We used the earlier measured uncertainty functions of the navigation steps 1, 2 and 3 for calcite, cordierite and tourmaline sunstone crystals, respectively, and the newly measured uncertainty function of step 4 presented here. As a result, we revealed the meteorological conditions under which Vikings could have used this hypothetical navigation method. We determined the solar elevations at which the navigation uncertainties are minimal at summer solstice and spring equinox for all three sunstone types. On average, calcite sunstone ensures a more accurate sky-polarimetric navigation than tourmaline and cordierite. However, in some special cases (generally at 35°???????40°, 1 okta???????6 oktas for summer solstice, and at 20°???????25°, 0 okta???????4 oktas for spring equinox), the use of tourmaline and cordierite results in smaller navigation uncertainties than that of calcite. Generally, under clear or less cloudy skies, the sky-polarimetric navigation is more accurate, but at low solar elevations its accuracy remains relatively large even at high cloudiness. For a given ?, the absolute value of averaged peak North uncertainties dramatically decreases with increasing ? until the sign (±) change of these uncertainties. For a given ?, this absolute value can either decrease or increase with increasing ?. The most advantageous sky situations for this navigation method are at summer solstice when the solar elevation and cloudiness are 35°???????40° and 2 oktas???????3 oktas.

Project description:To characterize the molecular signature of the choriodecidua remodeling in third trimestry of pregnancy before labour, we have employed whole genome microarray expression profiling. Fetal membranes were collected from women undergoing a emergency cesarean delivery before term (before 32 weeks of gestation and between 33-35 weeks of gestation), for preeclampsia and/or in utero growth retardation. During surgery, a Kocher's forceps was placed on membranes overlying the cervix and the area surrounding the forceps was dissected in the operating room under sterile conditions, the site, pinched by the forceps, being discarded. Fetal membranes in a remote zone were also sampled. Isolated RNAs were hybridized to the Aligent SurePrint G3 Human GE 8x60K Microarray.

First, we improved LentiMutate by making the lentiviral RT more error-prone. Next, we applied this technique to two anti-cancer drugs, imatinib and AMG 510. We find novel deletions in BCR-ABL that confer resistance to BCR-ABL inhibitors and point mutations in the AMG 510 binding pocket or oncogenic non-G12C mutations, in KRAS-G12C or wild-type KRAS, respectively, that confer resistance to AMG 510. LentiMutate may prove highly valuable to clinical and preclinical cancer drug development

Project description:BackgroundAs current augmented-reality (AR) smart glasses are self-contained, powerful computers that project 3-dimensional holograms that can maintain their position in physical space, they could theoretically be used as a low-cost, stand-alone neuronavigation system.ObjectiveTo determine feasibility and accuracy of holographic neuronavigation (HN) using AR smart glasses.MethodsWe programmed a fully functioning neuronavigation system on commercially available smart glasses (HoloLens®, Microsoft, Redmond, Washington) and tested its accuracy and feasibility in the operating room. The fiducial registration error (FRE) was measured for both HN and conventional neuronavigation (CN) (Brainlab, Munich, Germany) by using point-based registration on a plastic head model. Subsequently, we measured HN and CN FRE on 3 patients.ResultsA stereoscopic view of the holograms was successfully achieved in all experiments. In plastic head measurements, the mean HN FRE was 7.2 ± 1.8 mm compared to the mean CN FRE of 1.9 ± 0.45 (mean difference: -5.3 mm; 95% confidence interval [CI]: -6.7 to -3.9). In the 3 patients, the mean HN FRE was 4.4 ± 2.5 mm compared to the mean CN FRE of 3.6 ± 0.5 (mean difference: -0.8 mm; 95% CI: -3.0 to 4.6).ConclusionOwing to the potential benefits and promising results, we believe that HN could eventually find application in operating rooms. However, several improvements will have to be made before the device can be used in clinical practice.

Project description:Total RNAs from the Citrus red mite at the embryo, larva, nymph and adult stages were used to construct small RNA libraries for Solexa sequencing. Several categories of sRNAs were identified, including 594 known microRNAs (miRNAs) grouped into 206 families and 31 novel miRNAs. In addition, according to bioinformatics analysis and S-Poly(T) miRNA assays, the expression level of many miRNAs varied among the developmental stages. Furthermore, the prediction of miRNAs target genes and their functional annotation indicated that miRNAs are involved in the regulation of multiple pathways in the Citrus red mite.
