Retrospective follow-up study of lymph node metastasis examination by OSNA in colorectal cancer.

ABSTRACT: Primary outcome(s): 1. Three-year disease-free survival rate and three-year overall survival rate in each pathological stages, which are diagnosed based on both of OSNA and histopahtological methods. 2. Recurrence prognostic factors in the group which is diagnosed with StageII based on histopathological method. 3. Setting of cut-offs of the sum of the quantitative results of OSNA for three-year disease-free survival rate and three-year overall survival rate. 4. Three-year disease-free survival rate and three-year overall survival rate in each diagnosed group.

Transcription profiling of human primary tumors from patients with metastatic neuroblastoma lacking MYCN amplification

Project description:Metastatic neuroblastomas lacking amplification of the MYCN proto-oncogene vary in their clinical behavior. Those diagnosed before one year of age are least aggressive and those diagnosed after two years are most aggressive. To improve current risk stratification and gain insight into the biological processes that account for this phenomenon, we analyzed expression profiles from 102 primary untreated neuroblastomas. A 55 gene model classified patients diagnosed after 12 months of age based on molecular risk with an error rate of 17.5%. The progression-free survival for this group was 16% and 79% if their tumors had molecular high or low risk profiles (p<0.001). Expression profiles of tumors with molecular high-risk features included genes relating to regulation of growth and programmed cell death. This series also includes 15 samples obtained at disease relapse. Fourteen out of the fifteen samples had molecular high risk features. Experiment Overall Design: 102 samples obtained at diagnosis were analyzed for model generation and compared to 15 obtained at relapse, using Affymetrix HG-U133A (GPL96) and HG-U133B (GPL97) platforms.

Researchers will compare another group treated with conventional preoperative chemoradiotherapy to see if the experimental group is superior to the control group in terms of 3-year disease free survival, local recurrence rate, lateral lymph node positivity rate, and 5-year overall survival rate.

Expression data from 128 Desmoids

Project description:One of the main problems in managing desmoids tumors is their locoregional aggressiveness and their high ability to recur after initial treatment. In our work, with the goal to identify molecular markers that can predict Progression-Free Survival, gene-expression screening was conducted on 128 available independent untreated primary desmoid tumors using cDNA microarray. By analyzing expression profiles, we have identified, for the first time, a gene expression signature that is able to predict Progression-Free Survival. This molecular signature identified two groups with clearly distinct Progression-Free Survival in the two sets of subjects. Patients in good prognostic group had achieved a progression-free 2-year survival rate of 86% while patients in poor prognostic group had a progression-free 2-year survival rate of 44%.

Long noncoding RNA (lncRNA) expression data from human myelodysplastic syndromes (MDS) bone marrow mononuclear cells

Project description:lncRNAs not only participate in normal hematopoiesis but also contribute to the pathogenesis of acute leukemia. However, their clinical and prognostic relevance in MDS remains unclear to date. In this study, we profiled the lncRNA expressions in 176 adult patients with primary MDS with Affymetrix GeneChip Human Transcriptome Array 2.0, and identified the lncRNAs whose expression levels were significantly associated with overall survival (OS). We then constructed a risk scoring system with the weighted sum of these lncRNAs. Patients with higher lncRNA scores had a significantly shorter OS and a higher five-year leukemic transformation rate compared with those with lower scores.

Project description:Pancreatic ductal adenocarcinoma (PDAC) remains one of the most aggressive and lethal malignant tumors all over the world, with a 5-year survival rate of less than 10%1,2. Usually diagnosed at an advanced stage, PDAC is highly resistant to current therapies, partly due to an extremely high rate of distant metastasis (>80%) at first diagnosis. The liver is the most frequent site of distant metastases for PDAC. Current therapeutic options for PDAC patients with liver metastasis are extremely limited3. Thus, understanding the molecular mechanisms underlying hepatic metastasis of PDAC is urgently needed to help us develop more effective treatments and improve survival for these patients with advanced disease.
