The hypothesis tested whether the presence of polyphenols in the fruit and vegetable juice concentrate could reduce the expression of systemic inflammatory genes in the blood of Obese patients with high levels of plasma CRP (≥3.0). Results provide evidence that systemic inflammatory genes/ and or pathways may be modulated by the fruit and vegetable juice concentrate.

Project description:Transcriptome analysis of Eggplant cv. PPL during fruit development at 0, 5, 10, 20 and 50 dpa. Eggplant is third most important solanaceae crop species after potato and tomato. It is a versatile crop adapted to different agro-climatic regions and can be grown throughout the year. Unripe eggplant fruit is consumed as cooked vegetable in various ways. It is low in calories and fats, contains mostly water, some protein, fibre and carbohydrates. To decipher molecular mechanisms involved in fruit development eggplant fruit were collected at 0, 5, 10, 20 and 50 dpa and gene expression profiles were analyzed using Affymetrix tomato GeneChip Genome array.

Project description:Previous studies have suggested that increases in maternal cortisol or maternal stress in late pregnancy increase the risk of stillbirth at term. In an ovine model with increased maternal cortisol over the last 0.20 of gestation, we have previously found evidence of disruption of fetal serum and cardiac metabolomics, and of expression of genes related to mitochondrial function and metabolism in biceps femoris, diaphragm and cardiac muscle. The present studies were designed to test for effects of chronically increased maternal cortisol on gene expression and metabolomics in placentomes near term. We hypothesized that changes in placenta may underlie or contribute to the alterations in fetal serum metabolomics, and thereby contribute to changes in striated muscle metabolism. Multi-omic analysis indicates that amino acid metabolism, particularly of branched chain amino acids and glutamate occur in placenta; changes in amino acid metabolism, degradation or biosynthesis in placenta were consistent changes in valine, isoleucine, leucine and glycine in fetal serum. The analysis also indicates changes in glycerophospholipid metabolism, and suggests changes in ER stress and antioxidant status in the placenta. These findings suggest that changes in placental function occurring with excess maternal cortisol in late gestation may contribute to metabolic dysfunction in the fetus at birth.

Project description:Hybridization of one kidney of cortisol treated fish vs. one kidney of control fish. Kidneys were collected from untreated juvenile sea bream (n=4) and from fish, which received for 72h a coconut-oil implant containing 10mg/Kg (fish wet weight) (n=4) cortisol. Experiments were carried out at the University of the Algarve, Portugal in accordance with National legislation for the welfare of animals. Experiments were conducted in two 125 l cylindriconical tanks supplied with a continuous through-flow of oxygenated seawater at 20+1 °C using juvenile sea bream (25 g + 3 g) adapted for 1 week to the experimental conditions. One tank contained 8 untreated fish (control) and the other tank 8 cortisol treated fish and the end of experiments fish were removed form tanks, decapitated and the kidneys rapidly removed and place in RNAlater (Qiagen) at â20 °C. No mortality occurred in the control tank but 2 fish died in the cortisol treated tank.

Project description:Background: The prevalence of individuals allergic to latex, exhibiting cross-hypersensitivity with plant-derived food has been frequently reported as the so-called latex-fruit syndrome. Nonetheless, molecular mechanisms underlying allergy to latex and/or fruit are poorly understood. Objective: The aims of this study were to identify candidate genes that may be associated with the pathogenesis of allergy to latex and /or vegetable food, and to assess if similar molecular pathways are involved in both types of hypersensitivity. Methods: DNA microarray analysis was performed to screen the molecular profiles of peripheral blood mononuclear cells isolated from patients with allergy to latex, to fruit, or with latex-fruit syndrome, and from control healthy subjects. Results: Gene expression profiling identified an overlapping dataset of genes commonly regulated in all the atopic patients enrolled in this study, suggesting that similar molecular mechanisms are involved in the pathogenesis of allergy to the fruit and /or latex. Several regulators of the innate and acquired immunity reported to polarize the immunological response towards a Th2-mediated immune response were overexpressed in patients. Furthermore, evidences suggested that T regulatory cell expression might be defective in allergic patients, as a consequence of a dysregulation of some inflammatory cytokines. Finally, several transcription factors that may be responsible for the Th1/Th2 imbalance were modulated in allergic patients. Conclusion and clinical implications: This study identified relevant genes that may help to elucidate the molecular mechanisms underlying allergic disease. Knowledges of critical targets, along with transcription factors regulating gene activity may facilitate the development of new therapies. Experiment Overall Design: This study aimed at the understanding of the molecular pathways involved in allergy to latex and fruit. As latex and vegetable food allergens cross-react, it could be hypothesized that similar pathways are involved in both types of hypersensitivity. Experiment Overall Design: For this purpose, patients allergic to latex (n=6), allergic to vegetable food (n=5), or suffering from latex-fruit syndrome (n=6) were enrolled from the Unit of Allergy of the Gemelli Hospital of Rome. Moreover, 4 healthy volunteers were added to this study. Five ml of blood were harvested from each individual, and PBMCs were isolated on ficoll gradient. Following sample preparation (as recommended by the manufacturer), each sample was hybridized on Affymetrix human focus array. Data were processed with Affymetrix MAS 5.0 software and averaged intensity of signals from biological replicates were calculated for further analysis. Project description:Although oral administration of Bifidobacterium longum (B. longum) relieves irritable bowel syndrome (IBS) symptoms in a clinical setting, the underlying mechanisms remain undetermined. Herein, we confirmed that B. longum ameliorated defecation habits and alleviated visceral hypersensitivity in water avoidance stress (WAS) rats. Further analysis revealed that B. longum enhanced mucosal repair, promoted the production of lysozyme, and ameliorated microbiota dysbiosis in WAS rats. These activities are all closely correlated with Paneth cell function. In vitro, we incubated primary cultured enteroids with B. longum and found that this bacterium promoted the proliferation of these organoids, which may be attributed to the up-regulated expression of the stem niche factors WNT3A and TGF-β which are serected by the Paneth cells. On the basis of our findings, we propose that B. longum relieves IBS by restoring the antimicrobial activity and stem niche maintenance functions of Paneth cells.

Project description:The coconut tree (Cocos nucifera L.) is an ancient palm species that since early times had been integrally exploited due to their many benefits to the habitants of the tropical and subtropical areas (Niral and Jerard, 2018). The coconut together with the oil palm (Elaeis guineensis) and date palm (Phoenix dactylifera L.) belongs to the Arecaceae family and represent a natural source of oils and carbohydrates both, highly demanded in food and pharmaceutical industries. The coconut shows high morphological variations, but grouping normally in two groups, according to their morphology and growth habits, the “Tall” and the “Dwarf” varieties. Frequently, hybrids are considered as a third variety resulting from cross pollination between Tall and Dwarfs. Tall varieties begin to flowering at 5-7 years after planting, and continue emitting inflorescences and fruits up to 80 to 100 years. When adults, tall palms reach heights between 20-30 m; producing fruits from medium to large in size with abundant solid endosperm and high oil content. In case of dwarf palm varieties, they start flowering and fruiting at 3-4 years after planting, and continue producing by almost 50 years. As adults, their height is 8-10 m, producing fruits from small to medium size with moderate amounts of solid endosperm and less oil content than tall varieties. The solid endosperm, also named as “coconut meat” and when it is dry “copra”, is the source of medium chain saturated fatty acids (MCSFA), e.g. lauric acid (C12:0), myristic acid (C14:0) and palmitic acid (C16:0), among others, and their content of total fatty acids is higher in Tall than in Dwarf varieties. Coconut Oil contains high levels of lauric acid and exhibit characteristics as increased oxidative stability, low melting points and formation of stable emulsions, all them highly appreciated in the food and chemical industries (Kumar, 2011; Reynolds et al., 2019). Moreover, due to its high content of antioxidants such as tocopherol and beta-carotene, coconut oil exhibit various health benefits, such as antibacterial, antiviral, and cardiovascular protection. Coconut fruit growth and ripening is intrinsically related with development of the different seed tissues, i.e., endosperms, embryo and pericarp. The endosperms of the coconut fruit did not accumulate synchronically with fruit maturation.

Project description:Cucumber (Cucumis sativus L.) is an important vegetable crop bearing fleshy pepo fruits that harvested immature. The fruit length is one of the most important agricultural traits that directly determine the fruit yield and affects fruit quality, but the regulatory mechanism of fruit length variation remains elusive. Here we found a FRUITFULL-like MADS-box gene CsFUL1 functions as a key repressor for fruit length regulation in cucumber. The expression of CsFUL1 is highly enriched in male flowers and fruits, and negatively correlated with fruit length in different cucumber lines. Notably, a key SNP in CsFUL1 was selected during cucumber domestication for long fruit. Ectopic expression of CsFUL1 was unable to rescue the indehiscent fruit phenotype of ful-1 in Arabidopsis. Overexpression of CsFUL1 resulted in increased floral organs and reduced fruit length, whereas knockdown of CsFUL1 led to elongated fruit in cucumber. Transcriptome and biochemical analyses showed that CsFUL1 regulates fruit length through two pathways: one by inhibiting the PIN-FORWED (PIN1/7)-mediated auxin transport and thus downregulates auxin related genes in the fruit, and the other by forming a tetramer with other MADS-box genes to repress the CsSUP-mediated cell division and cell expansion. In addition, we found that CsFUL1 promotes locule number variation through the classical CsWUS-CsCLV pathway. Our findings uncover the regulatory commonality and specificity during development of different fruit types, and provide an important candidate gene to customize fruit length during cucumber breeding.
