Primary outcome(s): The primary outcome of the study is the accuracy of the CAD-CNN system for predicting histology of diminutive colorectal polyps (1-5mm) compared with the accuracy of the prediction of the endoscopist. Both the CAD-CNN system and the endoscopist will use NBI for their predictions.
Accuracy is defined as the percentage of correctly predicted optical diagnoses of the CAD-CNN system and/or endoscopist compared to the gold standard pathology. For the calculation of the accuracy, adenomas and SSLs will be dichotomised as neoplastic polyps, while HPs and other non-neoplastic histology are considered non-neoplastic.

Project description:Primary outcome(s): This trial will be triple primary endpoint clinical trial. 1.When EC-CAD was used under methylene blue staining for polyps of 5 mm or less in diameter in the sigmoid colon from the rectum, it was verified that the result diagnosed with high confidence (probability 90% or more) was NPV 90% or more. 2.When observing a polyp of 5 mm or less in diameter with magnifying narrow band imaging (NBI) and NBI combined endocytoscopy (EC-NBI), verify that the ratio at which trainee can acquire a focused endoscope video for 3 seconds or more at EC-NBI. 3.When EC-CAD is used under methylene blue staining for colon lesions of 20 mm or more in diameter, we verify that the diagnostic ability for invasive cancer is over 90% PPV.

Project description:Aims: Exosomes exist in almost all body fluid and contain diverse biological contents which may be reflective of disease state. Circular RNAs (CircRNAs) are stable in structure and have a long half-life in exosomes without degradation, thus making them reliable biomarkers. However, the potential of exosomal circRNAs as biomarkers of coronary artery disease (CAD) remains to be established. Here we aimed to investigate the expression levels and the potential use of exosomal circRNAs as diagnostic biomarkers of CAD. Main methods: CircRNAs expression levels in exosomes obtained from three plasma samples from CAD patients and three paired controls were analyzed using RNA sequencing. Exosomal circRNAs obtained in the profiling phase were then verified in two-center validation cohorts. Finally, the ability of exosomal circRNAs, adjusting for Framingham Heart Study (FHS) risk factors, was determined to discriminate between CAD patients and non-CAD controls. Key findings: 355 circRNAs were differentially expressed between these two groups: 164 were upregulated, and 191 were downregulated. Here we selected potential circRNAs (fold change > 4, P < 0.05) as candidate biomarkers for further validation. Our data showed that only hsa_circ_0005540 was significantly associated with CAD (P < 0.0001). After adjustment for risk factors, hsa_circ_0005540 showed a high discriminatory power for CAD in ROC analyses (AUC = 0.853; 95% confidence interval [CI] = 0.799 - 0.906, P < 0.001). Significance: Our results suggest that plasma exosomal hsa_circ_0005540 can be used as a promising diagnostic biomarker of CAD.

Project description:A number of scientific applications are performance-limited by expressions that repeatedly call costly elementary functions. Lookup table (LUT) optimization accelerates the evaluation of such functions by reusing previously computed results. LUT methods can speed up applications that tolerate an approximation of function results, thereby achieving a high level of fuzzy reuse. One problem with LUT optimization is the difficulty of controlling the tradeoff between performance and accuracy. The current practice of manual LUT optimization adds programming effort by requiring extensive experimentation to make this tradeoff, and such hand tuning can obfuscate algorithms. In this paper we describe a methodology and tool implementation to improve the application of software LUT optimization. Our Mesa tool implements source-to-source transformations for C or C++ code to automate the tedious and error-prone aspects of LUT generation such as domain profiling, error analysis, and code generation. We evaluate Mesa with five scientific applications. Our results show a performance improvement of 3.0 × and 6.9 × for two molecular biology algorithms, 1.4 × for a molecular dynamics program, 2.1 × to 2.8 × for a neural network application, and 4.6 × for a hydrology calculation. We find that Mesa enables LUT optimization with more control over accuracy and less effort than manual approaches.

Project description:Previously, while studying the development, especially of corticospinal neurons, it was concluded that the main compensatory mechanism after unilateral brain injury in rat at neonatal stage was due to in part by nonlesioned ipsilateral corticospinal neurons that escaped selection by axonal elimination or neuronal apoptosis (Yoshikawa et al., 2011, Dev Neuroscience). However, above result suggesting compensatory mechanism in neonate brain could not be correlated with high functional recovery. Therefore, we asked - what is the difference among neonate and adult in context of functional recovery and mechanism/s therein, especially focused on the corticospinal neurons? Toward this goal, in the present study, we have utilized a unilateral brain injury mice model and compared motor functional recovery mechanism post-neonatal brain injury (hereafter termed, NBI) with adult brain injury (hereafter termed, ABI). Three analyses were perfomed: i) quantitative behavioral analysis of the forelimb movements using the ladder walking test, where the corticospinal system has been suggested to play an important role ii) a neuroanatomical retrogradely tracing analysis of unlesioned side corticospinal neurons; and iii) differentially global gene expressions profiling in the unlesioned-side neocortex (rostral from bregma) in NBI and ABI using a high-throughput DNA microarray approach on a 8x60 K whole genome Agilent DNA chip using a two-color (Cy3/Cy5) dye-swap approach. Behavioral data confirmed the higher recovery ability in NBI over ABI is related to non-lesional frontal neocoetex including rostral caudal forelimb area. With a overall aim to unravel the underlying molecular events, in the present study a first inventory of the differential expressed gene expressions genome-wide in the NBI and ABI model mice has been provided.

Project description:Coronary artery disease (CAD) remains a leading cause of mortality worldwide. The current clinical diagnosis differentiates among the types of CAD mainly dependents on coronary angiography. High cost limit it to a select population. On one hand, a sizable portion of individuals who underwent invasive angiography had been shown to have normal coronary arteries. On the other hand, episodes of myocardial ischemia or infarction are possible after atypical symptoms in some patients with CAD, especially in patients who are elderly or have diabetes. Additionally, the transition from coronary stability to instability is less well understood. To identify the marker proteins for CAD progression, proteomic technique was applied.
