Project description:In this project the main focus is on assessing sexual functioning and the quality of sexual life after the treatment of colorectal cancer in patients and their partners.
Patients and their partners complete questionnaires concerning sexual functioning, quality of life, body image, fatigue, anxiety, depressive symptoms, personality factors, and demographic factors. Questionnaires are completed before surgical treatment, 6 weeks, 3 months, 6 months, and 12 months after diagnosis.
The results of this prospective study will give insight in 1) the incidence of sexual problems and the extent patients with colorectal cancer and their partners are bothered by these problems across time, 2) the effect of different treatment modalities on sexual functioning, 3) the relation between sexual problems and quality of life, 4) the determinants of sexual problems and the quality of sexual life adopting the biopsychosocial approach of patients with colorectal cancer who have been treated with surgery, radiation and/or chemotherapy, and more specifically to the role of personality and patient factors and sexual functioning/the quality of sexual life.

Project description:Purpose: T. gondii has a complex life cycle typified by an asexual development taking place in vertebrate and a sexual reproduction occurring exclusively in felids and thereby is less studied. The developmental transitions rely on changes in gene expression patterns, and recent studies have assigned roles for chromatin shapers, including histone modifications, in establishing specific programs for a given stage. Here, we identified T. gondii microrchidia (MORC) protein as an upstream transcriptional repressor of sexual commitment. MORC, in partnership with Apetala (AP2) transcription factors, was shown to recruit the histone deacetylase HDAC3, thereby impeding the chromatin accessibility of the genes predestined to be exclusively expressed in sexual stages. As such, MORC-depleted cells underwent marked transcriptional changes, resulting in the expression of a specific repertoire of genes, thus revealing a shift from asexual proliferation to sexual differentiation. MORC acts as a master regulator that directs the hierarchical expression of secondary AP2 factors, with these latter potentially contributing to the unidirectionality of the life cycle. Thus, MORC plays a cardinal role in the T. gondii life cycle, and its conditional depletion offers a way to study the parasite’s sexual development in vitro, and proposes an alternative to the requirement of cat infections.

Project description:Sexual Schmidtea mediterranea Trunk Regeneration. Sexual biotype Schmidtea mediterranea trunk fragments were sequenced at 0 day, 3 day, 5 day and 7 day post amputation along with whole worm juvenile animals in order identify sexual specific transcripts. Juvenile worms lack sex organs, testes and ovaries present in D0 trunk fragments. Days 3-7 represent time points of degeneration and regeneration of the sexual organs, testes and ovaries.

Project description:Pseudo-nitzschia multistriata is a planktonic diatom species with a diplontic life cycle comprising a short sexual phase during which gametes are produced from the encounter of two diploid cells of opposite mating type (MT). Gene expression studies have highlighted the presence of substantial changes occurring at the onset of sexual reproduction. Collectively, there is clear evidence of a chemical cross talk which mediates the mating step.Herein, we have hypothesized that these major transcriptomic changes are associated to variations in the amount and nature of the metabolites produced by the cells. In order to investigate such chemical diversity in an unbiased manner, we undertook an untargeted metabolomics approach. Liquid chromatography – tandem mass spectrometry was applied to investigate the differences in the metabolic profiles between control cells in the vegetative phase (MT+ and MT-) and mixed strains of opposite MTs (cross) undergoing sexual reproduction. Of the 2408 high-quality features obtained, 70 known metabolites could be identified based on in-house libraries and online databases; molecular networking proved valuable to classify additional 47 non-identified features.The present exploratory study provides an overview of the chemical diversity detectable in P. multistriata, presenting a dynamic picture of the changes occurring during sexual reproduction. In particular, the reduction of phytol detected in the cross can be linked to the general downregulation of photosynthesis during sexual reproduction observed elsewhere. The dysregulation of other metabolites stands as a starting point for the understanding of the mechanisms regulating sexual reproduction: a metabolite that was tentatively identified as 7-dehydrodesmosterol exhibited the highest upregulation in the comparison between the two MTs of opposite sex, as well as between MT+ vs cross.The effects of extraction solvents and harvesting times on the ensemble of endo-metabolites were also analyzed.

Project description:Study hypothesis: Evidence suggests that increased physical activity following diagnosis and treatment for colorectal cancer may reduce the risk of disease recurrence after 2.7 years and improve survival by up to 50%. Studies have also shown an association between increased physical activity and other health outcomes for people living with colorectal cancer, for example improved quality of life and improved mental well-being.
Partners may play a role in affecting physical activity behaviour change. The family can be a significant determinant of individual health behaviours. Shared familial environmental factors have been shown to partly account for similarities in physical activity levels amongst family members.
Research has also shown that spousal support is an important factor in smoking cessation, use of health services and weight reduction. Partners may be also a receptive target for physical activity behaviour change. Physical activity consultations may help to increase the physical activity levels and improve the health outcomes of the partners of people living with colorectal cancer as well as the patient.
Primary aim:
To evaluate the feasibility of conducting a randomised controlled trial of physical activity consultations with people living with colorectal cancer and their partners.
Research questions:
1. Is it possible to recruit patients who have completed all surgery and treatment for colorectal cancer in the last 24 months?
2. What would a likely response rate be for a future randomised controlled trial?
3. What are the demographic characteristics of those who are recruited to the study?
4. Can participants be retained in the study for the full 6 months?
5. Is it possible to conduct joint physical activity consultations with people living with colorectal cancer and their partners?
6. Are the outcome measures able to be used and acceptable for use with people living with colorectal cancer and their partners?
7. What would the effect size be for a future power calculation?
Secondary aim:
To evaluate the effects of physical activity consultations on physical activity levels, mental health, quality of life and body composition.
Research questions:
1. What is the effect of physical activity consultations on the physical activity levels of people living with colorectal cancer and their partners?
2. What is the effect of physical activity consultations on depression and anxiety?
3. What is the effect of physical activity consultations on quality of life?
4. What is the effect of physical activity consultations on body composition?
5. Do cancer and other health risk perceptions, intra-couple support and self-efficacy predict any changes in physical activity levels?
Primary outcome(s): Feasibility will be measured by calculating the following rates:
1. Recruitment rate (number of participants randomised/number of eligible participants)
2. Completion rate (number of participants completing baseline and 6 month intervention assessments/number of participants randomised)
3. Adherence rate (number of physical activity consultations and telephone calls conducted/number of physical activity consultations and telephone calls scheduled)

Project description:Malaria parasites go through major transitions during their complex life cycle, yet the underlying differentiation pathways remain obscure. Here we apply single cell transcriptomics to unravel events that initiate sexual development in preparation for transmission of the parasite from human to mosquito. This proof-of-concept study provides a template to capture transcriptional diversity in heterogeneous parasite populations, with major implications for our understanding of parasite biology and the ongoing malaria elimination campaign.

Project description:Sexual reproduction of Toxoplasma gondii, which is restricted to the small intestine of felids, is sparsely documented in studies involving domestic cats, which also raises ethical concerns. Chromatin modifiers dictate the developmental fate of the parasite during its multistage life cycle, but their targeting to stage-specific cistromes is poorly described. Here we show that AP2XII-1 and AP2XI-2, two transcription factors expressed in tachyzoites, a stage that causes acute toxoplasmosis, silence genes specific to merozoites, a developmental stage critical for sexual commitment and transmission to the next host, including humans. Their conditional and simultaneous depletion leads to a drastic change in the transcriptional program, promoting a complete transition from tachyzoites to merozoites, as revealed by mass spectrometry-based proteomic analyses of the parasites.
