Gene expression data from striatal regions of MPTP-intoxicated mouse brain by acupuncture

Project description:Acupuncture at acupoints GB34 and LR3 has been reported to inhibit nigrostriatal degeneration in parkinsonism models, yet the genes related to this preventive effect of acupuncture on the nigrostriatal dopaminergic system remain elusive. We investigated gene expression profile changes in the striatal region of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced parkinsonism models after acupuncture at the acupoints GB34 and LR3 C57BL/6 mice were divided into four experimental groups; â C: Control, â¡ M: MPTP-treatment only, â¢ MA: MPTP- and acupuncture-treatment at acupoints GB34 and LR3, â£ MNA: MPTP- and acupuncture-treatment at non-acupoints. Total RNA was isolated from two brains' striatal regions of each experimental group (4 experimental group Ã 2 samples of each experimental group = total 8 samples).

Gene expression data from thalamic regions of MPTP-intoxicated mouse brain by acupuncture

Project description:Acupuncture stimulations at GB34 and LR3 inhibit the reduction of tyrosine hydroxylase in the nigrostriatal dopaminergic neurons in the parkinsonism animal models. Especially, behavioral tests showed that acupuncture stimulations improved the motor dysfunction in a previous study. The thalamus is a crucial area for the motor circuit and has been identified as one of the most markedly damaged areas in Parkinson’s disease (PD), so acupuncture stimulations might also have an effect on the thalamic damage. We investigated gene expression profile changes in the thalamic region of 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP)-induced parkinsonism models after acupuncture at the acupoints GB34 and LR3 C57BL/6 mice were divided into four experimental groups; ① C: Control, ② M: MPTP-treatment only, ③ MA: MPTP- and acupuncture-treatment at acupoints GB34 and LR3, ④ MNA: MPTP- and acupuncture-treatment at non-acupoints. Project description:It has been shown that acupuncture at acupoints GB34 and LR3 inhibits the degeneration of nigrostriatal neurons in an 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) mouse model of Parkinsonâ??s disease. This preventative effect of peripheral acupuncture stimulation is hypothesized to be transmitted through the spinal cord to the nigrostriatal neurons. Genome-wide profiling of gene expression under electro-acupuncture pretreatment on myocardial I/R injury in rats

Project description:Purpose:Acupuncture exerts cardioprotective effect on myocardial I/R injury. In order to elucidate the potential mechanisms, RNA-seq by next generation sequencing was used to identify the rat genome-wide alterations by EA at Neiguan acupoint pretreatment in this study Methods: Adult male Sprague Dawley rats (250-300g) were divided into four groups: Sham operation(SO), I/R, electro-acupuncture at Neiguan pretreatment (EA) and electro-acupuncture at non-acupoint (NA) groups and myocardium mRNA profiles of rats in each group were generated by deep sequencing,using Illumina Hiseq 2000. The sequence reads that passed quality filters were analyzed at the transcript isoform level with two methods: Burrows–Wheeler Aligner (BWA) followed by ANOVA (ANOVA) and TopHat followed by Cufflinks. Results: Using an optimized data analysis workflow, we mapped about 20~43 million sequence reads per sample to the mouse genome (UCSC RN4) and identified 26190 22895 25160 22852 transcripts in the SO, I/R, EA and NA rats with TopHat and Cufflinks workflow respectively. Approximately 32% and 22% of the transcripts showed differential expression between the SO and I/R, I/R and EA , I/R and NA, respectively with a |log2 fold change |≥1. Conclusions: Our results for the first time generated genome-wide gene expression profiles both in the rat I/R model and in acupuncture treatment by high throughput sequencing. The optimized data analysis workflows reported here should provide a framework for acupuncture investigations of expression profiles.

Transcriptome analysis and differentially expressed gene screening for hypoxic-ischemic brain damage in rats treated with acupuncture

Project description:Purpose: Hypoxic-ischemic brain damage (HIBD) is one of the most common critical diseases in neonates and has high mortality and disability rates. Increasing evidence shows that long non-coding RNAs(lncRNA) plays an important role in the development of HIBD. Recently, acupuncture therapy has been found to be effective in the treatment of HIBD. However, the mechanism of lncRNA in acupuncture treatment of HIBD is still unclear. Method: In this study, a whole transcriptometric RNA-sequencing technique was applied to investigate transcriptome changes caused by acupuncture in PFC. Results: A total of 48 mRNAs (7 up-regulated and 41 down-regulated) and 65 lncRNAs (17 up-regulated and 48 down-regulated) was identified relate to acupuncture group and model group. According to Kyoto Encyclopedia of Genes and Genomes (KEGG) and Gene Ontology (GO) analysis, we found several lncRNAs and their target mRNAs were related to PI3K-Akt signaling pathway, TNF signaling pathway and NOD-like receptor signaling pathway, etc. Conclusion: we found several significant lncRNAs whose corresponding target genes might be the targets of acupuncture therapy for HIBD. The results of our study may provide new perspectives on the mechanism of acupuncture and affect the diagnosis and therapy of HIBD
