CtDNA-guided Adjuvant Chemotherapy in Liver Metastasis of Colorectal Cancer

ABSTRACT: The goal of this clinical trial is to compare in resectable colorectal cancer liver metastasis patients.The main question it aims to answer is whether the 3-year progression-free survival rate (PFS) of "watching and waiting" is non-inferior to adjuvant chemotherapy in postoperative ctDNA-negative resectable colorectal cancer liver metastasis patients.Participants will undergo ctDNA testing after resection of colorectal cancer liver metastasis, and will be randomly assigned to receive adjuvant chemotherapy or "watching and waiting" treatment strategy. The researchers will compare the outcomes between the two groups to see if the PFS between the two groups is similar.

The cohort consisted of 21 patients with primary, untreated, resectable, high-risk GIST divided into two groups; patients who developed metastatic disease within three years and patients without disease relapse for more than five years follow-up after adjuvant imatinib treatment.

Project description:In the last decades platinum-based neo-adjuvant chemotherapy (NACT) has been recognized as a reliable therapeutic strategy in patients with un-resectable advanced epithelial ovarian cancer (EOC). However, the molecular changes induced by NACT at miRNA level, and their prognostic role has not been clarified until now. In order to uncover miRNAs that are altered in EOC tumor which received NACT, we performed whole-miRNA analysis on 82 FIGO Stage IIIC-IV high-grade serous (HGS) tumors, whose samples had been collected at complete primary debulking (PDS) and at interval-debulking surgery (IDS) after fter 4 courses of NACT.

Gene expression analysis of multiple myeloma patients with relapse

Project description:Total RNA was obtained from bone marrow aspirates isolated from 16 multiple myeloma patients who underwent auto bone marrow transplantation (Auto-BMT). Our study was to define altered gene expression in MM relapse, gene expression profile was examined with 16 MM patients by progression-free survival (PFS) 12 months after autologous stem cell transplantation. Patients at each end of the PFS spectrum, 4 with shortest PFS and 4 with the longest PFS, were chosen for analyses to maximize the difference between different prognostic groups.

Project description:Migrating cancer stem cells (MCSCs) are believed to be tumorigenic initiators of metastasis. However, the relevance of MCSCs for organ-specific metastasis in colorectal cancer remains unclear. Using in vivo selection, transcriptomic analysis and functional verification, we cultured colorectal cancer stem cells and discovered specific cell surface markers of MCSCs exhibiting distinct abilities to metastasize to the liver and lung. These results have important clinical implications as selection criterion for post-operative adjuvant therapy.
