Synthetic IL-22 signaling revealed biological activity of homodimeric IL-10 receptor 2 and functional cross-talk with the IL-6 receptor gp130

Project description:Cytokine signaling is transmitted by cell surface receptors which function as natural biological switches to control among others mainly immune related processes. Recently, we have designed synthetic cytokine receptors (SyCyRs) consisting of GFP- and mCherry-nanobodies fused to trans-membrane and intracellular domains of cytokine receptors, which phenocopied cytokine signaling induced by non-physiological homo- and heterodimeric GFP-mCherry ligands. Interleukin 22 signals via IL-22Rα1 and the common IL-10R2, belongs to the IL-10 cytokine family and is critically involved in tissue regeneration. IL-22 SyCyRs phenocopied native IL-22 signal transduction as shown by induction of cytokine-dependent cellular proliferation, signal transduction and transcriptome analysis. Whereas homodimeric IL-22Rα1 SyCyRs failed to activate signaling, homodimerization of the second IL-22 signaling chain, SyCyR(IL-10R2), which was considered to not induce signal transduction, lead to induction of signal transduction. Interestingly, the SyCyR(IL-10R2) and SyCyR(IL-22Rα1) were able to form functional heterodimeric receptor signaling complexes with the synthetic IL-6 receptor chain SyCyR(gp130). In summary, we demonstrated that IL-22 signaling can be phenocopied by synthetic cytokine receptors. Further we identified a novel IL-10R2 homodimeric receptor complex and receptor cross-talk with gp130.

Geothermometry of calcite spar at 10-50 °C.

Project description:Carbonate geothermometry is a fundamental tool for quantitative assessment of the geothermal and geochemical evolution of diagenetic and hydrothermal systems, but it remains difficult to obtain accurate and precise formation temperatures of low-temperature calcite samples (below ~ 40 to 60 °C). Here, we apply three geothermometry methods (∆47-thermometry, nucleation-assisted fluid inclusion microthermometry-hereafter NA-FIM-and oxygen isotope thermometry) to slow-growing subaqueous calcite spar samples to cross-validate these methods down to 10 °C. Temperatures derived by NA-FIM and Δ47-thermometry agree within the 95% confidence interval, except for one sample. Regression analyses suggest that the real uncertainty of ∆47-thermometry exceeds the 1 SE analytical uncertainty and is around ± 6.6 °C for calcite spar that formed at 10-50 °C. The application of δ18O thermometry was limited to a few samples that contained sufficient primary fluid inclusions. It yielded broadly consistent results for two samples with two other geothermometers, and showed higher temperature for the third spar. We also found that calcite with steep rhombohedral morphologies is characteristic of low temperatures (11-13 °C), whereas blunt rhombohedra prevail in the 10-29 °C domain, and the scalenohedral habit dominates > 30 °C. This suggests that the calcite crystal morphology can be used to qualitatively distinguish between low- and higher-temperature calcite.

Novel ultra-sensitive detectors in the 10-50 ?m wavelength range.

Project description:We have developed novel single-photon detectors in the 10-50 ?m wavelength region. The detectors are charge-sensitive infrared phototransistors (CSIPs) fabricated in GaAs/AlGaAs double quantum well (QW) structures, in which a photo-generated hole (+e) in the floating gate (upper QW) modulates the conductance of a capacitively-coupled channel located underneath (lower QW). The excellent noise equivalent power (NEP = 8.3 × 10(-19) W/Hz(1/2)) and specific detectivity (D(*) = 8 × 10(14) cm Hz(1/2)/W) are demonstrated for 15 micron detection up to 23 K, which are by a few orders of magnitude better than those of other state-of-the-art high-sensitivity detectors. The dynamic range exceeds 10(6) (~aW to pW) by repeatedly resetting the accumulated holes in the upper QW. Simple device structure makes the detectors feasible for array fabrication: Furthermore, monolithic integration with reading circuits will be possible.
