Multiple genetic loci have been associated with body mass index (BMI) and obesity. The aim of this study was to investigate the effects of established adult BMI and childhood obesity loci in a Greek adolescent cohort. Project description:The TEENAGE study is a cross-sectional study comprising healthy Greek adolescents. The study was conducted according to the guidelines specified in the Declaration of Helsinki and all procedures involving human subjects were approved by the Institutional Review Board of Harokopio University and the Greek Ministry of Education, Lifelong Learning and Religious Affairs. The TEENAGE study target population comprised adolescent students aged 13 to 15 years attending the first three classes of public secondary schools located in the wider Athens area of Attica. The regions across Attica from which students were recruited were physically similar and all urban in nature. Sampling of regions and schools selected to participate was random. Students were enrolled from the wider urban region to ensure that they were coming from families with varying educational background and socio-economic status. Moreover, in order to exclude any vast economic disparities, all participating schools were exclusively public schools, which are free of charge. A total of twenty-four schools participated in the study out of 285 junior secondary public schools in Attica. Anthropometric, medical and dietary information was obtained. DNA samples were genotyped using Illumina HumanOmniExpress BeadChips (Illumina, San Diego, CA, USA) at the Wellcome Trust Sanger Institute, Hinxton, UK. Genotype calling algorithm used was Illuminus (Teo et al., 2007).

PCB 52 has been found in the indoor air of schools where adolescent students and staff are being actively exposed. This study examined the livers of adolescent Sprague-Dawley rats after sub-acute exposure to PCB 52 via nose-only inhalation. Rats were exposed for 4 hours daily for 28 consecutive days. RNA extracted from both male and female livers were submitted for RNA sequencing to compare the effects of PCB 52 on the expression of genes in the liver.

Effects of 28-day nose-only inhalation of PCB52 (2,2′,5,5′-Tetrachlorobiphenyl) on the brain transcriptome

Project description:Banned from mass production, polychlorinated biphenyls (PCBs) still pose a neurological health threat decades later. PCB52, a volatile compound, has been found in the indoor air of schools where adolescent students and staff are exposed. The adolescent brain is vulnerable and highly influenced by the environment around it. This study used adolescent Sprague-Dawley rats to determine the neurological effects of a subacute (28-day) PCB52 inhalation exposure. Rats were exposed 4 h a day for 28 consecutive days with neurobehavioral tests conducted during the last 5 days of exposure. Based on the data including brain weights, PCB52 and metabolite levels, gene expression in the striatum and cerebellum, and neurobehavioral tests (Y-maze, HBT, OFT, and EPM); a subacute inhalation of PCB52 results in sex-specific differences in response to exposure. Our results indicate multilevel PCB52 influence that is different in adolescent males and females; from gene expression to downstream innate mechanisms driving behavior.

RNA extracted from both male and female fat depots were submitted for RNA sequencing to compare the effects of PCB 52 on the expression of genes in the fat depots.

Project description:<p>While many genetic and metabolic studies focus on understanding disease states, this genome wide association study (GWAS) was designed to understand the influence of genetic variants on circulating metabolites and factors in normal, healthy individuals. Over 2500 healthy, ethnically Irish college students attending Trinity College Dublin completed a health and diet questionnaire and provided a non-fasting blood sample to be used for genetic and metabolic analysis. The questionnaire was designed to collect information on age, sex, height, weight, medical conditions, smoking, dietary habits, and consumption of alcohol, fortified foods, and supplements. Extracted DNA was genotyped using the Illumina HumanOmni1-Quad Beadchip. Metabolites were measured using a number of methods and focused on those relevant to the folate/vitamin B12 pathway.</p>

Parenting and Healthy Teenage Lifestyles.

Project description:How can one promote adolescent adjustment toward a healthy lifestyle? The first step is to locate the healthy habit configuration within the family environment. The hypothesis is that, if adolescent lifestyles are assumed autonomously during adolescence, then it is very likely that they will last throughout life. How does this relate to parenting styles? After reviewing the literature of the last four decades on adolescent behavioral autonomy and scientific articles that link healthy lifestyles with parenting, several conclusions have been reached, such as the relevance of recovering the biopsychosocial richness of healthy lifestyles, the need to use a dialogue strategy to resolve discrepancies between adolescents and their parents, and the adequacy of the personalistic parenting style to promote adjusted adolescent behavioral autonomy, and with it maintain healthy lifestyles in the long term.
