GenomEUtwin

ABSTRACT: GenomEUtwin is EU FP6 funded study aiming to capitalize special advantages of Europe in population genetics by efficient collaboration of twin researchers, genetic epidemiologists, molecular geneticists and mathematicians. Our goal is to identify critical genetic and life-style risk factors for common diseases using European strengths in genetics, epidemiology and biocomputing.

A cost-efficient way to increase power in a genetic association study is to pool controls from different sources. The genotyping effort can then be directed to large case series. Project description:European populations and epidemiological cohorts are of special significance in the current era of genomic research aiming to characterize the background of common human diseases. The genome sequence, detailed information of genetic variations between individuals, high-throughput molecular technologies and novel statistical strategies create new possibilities to define genetic and life-style risk factors behind common health problems. Studies of large population cohorts are needed to transform the genetic information to detailed understanding of the predisposing factors in diseases affecting most human populations. European twin cohorts provide a unique competitive advantage for investigations of the role of genetics and environment or life style in the etiology of common diseases. This project will apply and develop new molecular and statistical strategies to analyze unique European twin and other population cohorts to define and characterise the genetic, environmental and life-style components in the background of health problems like obesity, migraine, coronary heart disease and stroke, representing major health care problems worldwide.

Expression data from phenotypically discordant monozygotic twin lymphoblasts

Project description:Human intelligence demonstrates one of the highest heritabilities among human quantitative traits. Phenotypically discordant monozygotic twins provide a way to identify loci responsible for normal-range intelligence. We conducted array-based genome-wide gene expression analysis aiming to identify genes displaying significant difference among monozygotic twin pairs manifesting between-co-twins IQ differences. Total RNA was isolated from B-lymphoblastoid cell lines and applied to Affymetirx arrays after reverse transcription. 17 twin pairs (34 subjects) were recruited in this study.

Brisbane Systems Genetics Study

Project description:Brisbane Systems Genetics Study comprises of a total of 862 individuals from 374 families. Families consist of combinations of both MZ and DZ twin pairs, their siblings and for 72 families their parents. Whole blood for expression profiling was collected directly into PAXgene tubes and total RNA was extracted using the WB gene RNA purification kit. RNA from all samples was run on an Agilent Bioanalyzer to assess quality.

Enrichment of NICD4 interacting proteins by three different methods.

Project description:There are several methods to bring down the bait and interactors from the cell lysates, each with its own advantages and disadvantages. Recently, the most commonly used method is one step AP, TAP and proximity-labeling. In order to analyze the Strengths and weaknesses of the three methods, fusion proteins were constructed by NICD4 with FLAG, SFB (S tag-2x Flag tag-SBP tag) and TurboID respectively, NICD4 and its interactors were obtained by purification. By analyzing these proteins, we believe that SFB-TAP is the most reliable and effective method to identify interacting proteins.

Large-scale non-targeted serum metabolomics in the Swedish Twin Registry

Project description:The TwinGene project, conducted between 2004 and 2008, is a population-based Swedish study of twins born between 1911 and 1958. The study participants have previously participated in a telephone interview called Screening Across the Lifespan Twin Study, conducted between 1998 and 2002. To be included in TwinGene, both twins within a pair had to be alive. In total, 12591 individuals participated by donating blood to the study, and by answering questionnaires about life style and health. For the purpose of metabolomic profiling, we designed a case-cohort of incident coronary heart disease, diabetes, dementia and ischemic stroke events and a matched sub-cohort (controls) stratified on age and sex. In the final analysis we included serum samples from 2,139 individuals.
