Genomic profiling of subcutaneous patient derived xenograft models of solid childhood cancer

ABSTRACT: We established 70 subcutaneous PDX models from 16 different pediatric solid tumor subtypes and applied whole-exome sequencing (WES), whole-genome sequencing (WGS) and RNA sequencing (RNA-seq) to both PDX, matched patient tumor (PT) and germline samples. PDXs can generally recapitulate the genomic and transcriptomic features of their matched PT. However, we found a high somatic mutation discrepancy between PDXs and paired PTs in some cases and showed evidence of clonal selection.
