Project description:Breeding schemes for meat production in rabbits involved a three-way cross of specialized lines in which a paternal line inseminates maternal crossbred females. Paternal line or terminal sire are selected for growth traits, being the males used for the production of fertile dose at the insemination centres and farms. So high growth rate males must produce, in addition, semen in sufficient quantity and quality to meet the demand of insemination and, nevertheless, several studies have been showed that selection for growth have effects on reproduction performance in female and males. In rabbits, negative effects has been observed in ovulation induction, prenatal survival and genetic correlation to fertility. Many factors influence the production and quality of rabbit semen, management as collection frequency, environment (season or photoperiod, nutrition and genetic. Most of the previous studies have been focused in the effects of selection on the seminal and sperm parameters but, little attention has been paid to the protein seminal plasma or sperm composition and if these changes could be affect the fertility of seminal doses obtained from the paternal males. The aim of this study was to evaluate if selection program by daily gain in fattening period has changed seminal traits, plasma and sperm proteoma and, the fertility of semen when it is used in artificial insemination. To do this we uses two re-derived groups of paternal males obtained from vitrified embryos with a difference of 18 generations between both groups.

Project description:Allele frequency analysis reveals that the breed is enhanced for genes controlling traits associated with the poodle-type coat, which are perceived to have an association with hypoallergenicity, with no strong signatures of selection for Labrador retriever (LAB) traits. This study provides a blueprint for understanding how dog breeds are formed, highlighting the limited scope of trait selection in defining seemingly new breeds.

Project description:Complex traits and diseases can be influenced by both genetics and environment. However, given the large number of environmental stimuli and power challenges for gene-by-environment testing, it remains a critical challenge to identify and prioritize specific disease-relevant environmental exposures. We propose a novel framework for leveraging signals from transcriptional responses to environmental perturbations to identify disease-relevant perturbations that can modulate genetic risk for complex traits and inform the functions of genetic variants associated with complex traits. We perturbed human skeletal muscle, fat, and liver relevant cell lines with 21 perturbations affecting insulin resistance, glucose homeostasis, and metabolic regulation in humans and identified thousands of environmentally responsive genes. By combining these data with GWAS from 31 distinct polygenic traits, we show that the heritability of multiple traits is enriched in regions surrounding genes responsive to specific perturbations and, further, that environmentally responsive genes are enriched for associations with specific diseases and phenotypes from the GWAS catalog. Overall, we demonstrate the advantages of large-scale characterization of transcriptional changes in diversely stimulated and pathologically relevant cells to identify disease-relevant perturbations.

Project description:<p>Marine sponges can host abundant and diverse microbiomes, which can largely influence the metabolism and other phenotypic traits of the host. However, information on the potential relationships between sponge microbiomes and metabolic signatures, other than secondary metabolites explored for biotechnological purposes, needs further investigation. Applying an integrated approach, we investigated the microbiomes associated with 4 ubiquitous Mediterranean sponge species (i.e., Petrosia ficiformis, Chondrosia reniformis, Crambe crambe and Chondrilla nucula), correlated with their metabolomic patterns (in terms of lipidomics) and microbial predicted functions. Microscopy observations of sponge tissues revealed differences in microbial abundances, which, however, were only partially linked to their diversity assessed through metabarcoding. The microbiomes of the 4 sponges showed a species-specific composition and a different core size, which was independent from the microbial diversity of the surrounding seawater. Predicted functions of the associated microbiomes allowed identifying 2 functional host clusters: one more related to heterotrophic pathways and the other more linked to phototrophic activities. Differences in the microbiomes were also associated with different metabolic profiles, mostly due to specific compounds characterizing the host and its microbiome. Overall, this study provides new insights on the functionality of sponges and their prokaryotic symbioses’, and in particular, it discloses a descriptive sketch of the diverse compartments forming the sponge holobiont.</p>
