Alterations in the Transcriptional Profile of the Liver Tissue and the Therapeutic Effects of Propolis Extracts in Alcohol-induced Steatosis in Rats

ABSTRACT: Gene expression Array analysis has been an effective technique for exploring treatment-associated transcriptional changes in tissues. The main goal of this study is to explore the expression changes of transcripts in the liver tissue of rats which were treated with the ethanolic extraction of propolis (EEP) as well as alcohol. We aim to find a subset of genes which responded to the treatment of EEP and may be useful candidate genes in explaining the protective effects of EEP on fatty liver.

Project description:We showed that Brazilian green propolis attenuated Aβ-induced memory and learning impairment. Comprehensive gene expression analysis demonstrated that Brazilian green propolis treatment suppressed inflammation and immune responses via immune cells such as microglia and astrocytes in the brain. These results indicate the potential of Brazilian green propolis as a promising ingredient for preventing AD-type dementia.

So far, several line of studies demonstrated that propolis improved glucose metabolism in obese models. Comprehensive proteome analysis of propolis elucidates the origin and function of its proteins

Project description:Propolis is a resinous honeybee product, rich in polyphenolic compounds and with high economic value. Although extensive studies regarding the chemical composition of different propolis extracts have been carried out, the propolis proteome remained unknown. The present study aimed at characterizing the proteome of two geographically-distinct propolis originating from Belgium and Iran. Propolis extracts were analysed by SDS-PAGE followed by MALDI-TOF MS (mass spectrometry) and LC-ESI-MS. Our in-depth proteomic analysis led to the identification of honeybee venom and royal jelly proteins, and numerous plant-defense proteins in propolis, extending our understanding on the antimicrobial properties of this complex natural substance. The proteome of both propolis was mainly made up of proteins belonging to the poplar tree. The poplar PR-2 (β-1,3-glucanases) and PR-8 (acidic endochitinases) proteins were found to be the major components of both propolis extracts, while bee venom (Api m 1) and royal jelly allergens (MJRP 1 and 2) were only identified in the propolis from Belgium. Overall, our proteome analyses revealed that propolis extracts from Belgium and Iran shared a core protein composition originating from the poplar proteome. The majority of identified proteins are involved in the plant defense against pathogens, some belonging to well-known pathogenesis-related families.

Project description:The aim of this study was to analyze the influence of PADMA28 ethanolic extracts on HepG2 gene expression. PADMA28 (Swissmedic Nr. 58436) is an Indo-Tibetan polyherbal preparation used for the treatment of symptoms associated with circulatory disorders. Triplicate cultures of HepG2 cells were treated with either PADMA28 or EtOH solvent control for 18 hours. After cell harvest, the triplicates were pooled and total RNA was extracted. The RNA was analyzed using Affymetrix Human Genome U133 Plus 2.0 microarrays (one array per treatment).

Parasite Killing of Leishmania (V) braziliensis by Standardized Propolis Extracts.

Project description:Treatments based on antimonials to cutaneous leishmaniasis (CL) entail a range of toxic side effects. Propolis, a natural compound widely used in traditional medical applications, exhibits a range of biological effects, including activity against infectious agents. The aim of this study was to test the potential leishmanicidal effects of different propolis extracts against Leishmania (Viannia) braziliensis promastigotes and intracellular amastigotes in vitro. Stationary-phase L. (V) braziliensis promastigotes were incubated with medium alone or treated with dry, alcoholic, or glycolic propolis extract (10, 50, or 100 μg/mL) for 96 h. Our data showed that all extracts exhibited a dose-dependent effect on the viability of L. (V) braziliensis promastigotes, while controlling the parasite burden inside infected macrophages. Dry propolis extract significantly modified the inflammatory profile of murine macrophages by downmodulating TGF-β and IL-10 production, while upmodulating TNF-α. All three types of propolis extract were found to reduce nitric oxide and superoxide levels in activated L. braziliensis-infected macrophages. Altogether, our results showed that propolis extracts exhibited a leishmanicidal effect against both stages of L. (V) braziliensis. The low cell toxicity and efficient microbicidal effect of alcoholic or glycolic propolis extracts make them candidates to an additive treatment for cutaneous leishmaniasis.
