Deficiency in class III PI3-kinase induces intestinal epithelial damage and inflammatory bowel disease like phenotype in zebrafish

ABSTRACT: PIK3C3 and its product phosphatidylinositol 3-phosphate (PI(3)P) play critical roles in autophagy as well as vesicular trafficking. To fully compare the gene expression differences between wild-type and pik3c3 KO zebrafish, we performed RNAseq at 6dpf, 7dpf and 8dpf before mutant died. We found that homologs of barrier-function related human IBD susceptibility genes are suppressed while the inflammation response genes are stimulated in mutant, while genes involved in bacterial sensing and autophagy pathways are not affected. Thus, the pik3c3 mutant may serve as a valuable model for epithelial injury induced IBD.
