Silencing of ultra-conserved long non-coding RNA Uc.291 causes delay of keratinocyte differentiation

ABSTRACT: Development of the skin includes a complicated program of cellular differentiation. Here, we study the role of Uc.291, a long non-coding RNA derived from ultra-conserved region, upregulated in suprabasal layer of the skin and during in vitro differentiation of keratinocytes. Depletion of Uc.291 causes delay of keratinocyte differentiation with decreased levels of the expression of epidermal differentiation complex (EDC) genes.

Here we study a new membrane LIM-domain containing Zn-finger protein ZNF185 which is expressed in upper layers of human skin and is up-regulated during keratinocyte differentiation in vitro. Interestingly, depletion of ZNF185 causes delay of keratinocyte differentiation with decreased levels of FLG, LOR, LCEs expression.

Project description:To further explore the differential expression profile of Transcribed ultra-conserved region (TUCR) in human colorectal cancer, we have employed TUCR microarray expression profiling as a discovery platform to identify potential differential expression profile of TUCR between human colorectal cancer tissue (CCRT) and human normal mucosal epithelial tissue of the colorectal (NT). Results showed that multiple differentially expressed TUCRs were found between CCRT and NT. In this study, We selected T-UCRs with large differences to verification and found that uc.285+ upregulation was most obvious. Then we further explored the biological function and mechanism of uc.285+ in colorectal cancer.

Intense THz pulses down-regulate genes associated with skin cancer and psoriasis: a new therapeutic avenue?

Project description:We show that exposure of artificial human skin tissue to intense, picosecond-duration THz pulses affects expression levels of numerous genes associated with non-melanoma skin cancers, psoriasis and atopic dermatitis. Genes affected by intense THz pulses include nearly half of the epidermal differentiation complex (EDC) members. EDC genes, which are mapped to the chromosomal human region 1q21, encode for proteins that partake in epidermal differentiation and are often overexpressed in conditions such as psoriasis and skin cancer. In nearly all the genes differentially expressed by exposure to intense THz pulses, the induced changes in transcription levels are opposite to disease-related changes. Total RNA from exposed artificial human skin tissues to picosecond-duration broadband (0.2–2.5 THz) THz pulses with 1 kHz repetition rate, 1/e2 spot-size diameter of 1.5 mm and pulse energies of either 1.0 ?J or 0.1 ?J. For comparison, we have exposed skin tissues to UVA pulses (400 nm, 0.1 ps, 0.024 ?J).

Expression data from intestinal mucosa of patients with UC

Project description:Ulcerative colitis (UC) is a chronic inflammatory disease of the colon with preiods of active disease followed by remission. We performed a whole-genome transcriptional analysis of colonic biopsies from patients with histologically active and inactive UC, as well as non-inflammatory controls. Ulcerative colitis patients and non-inflammatory controls were collected for RNA extraction and hybridization on Affymetrix microarrays. Inclusion criteria for UC patients were: age between 18 and 65, diagnosis of UC established at least 6 months before inclusion and exclusion of concomitant infection. Active disease was defined by endoscopic and histologic score: Mayo sub score >=2 and MATTS >=3 respectively . Inactive disease was also defined by endoscopic and histologic score: Mayo sub score =0 and MATTS <=2 respectively, and a remission state for a minimum of 5 month prior to biopsy collection, and remained inactive for at least 6 months after. Uninvolved mucosa from patients with active UC was defined as a colonic segment with completely normal endoscopic appearance, normal histology, and absence of any previous evidence of active disease. Finally, a total of 43 biopsies were analyzed: 13 healthy controls, 8 inactive UC, 7 non-involved active UC and 15 involved active UC.
