Expression data from human dermal fibroblast subpopulations

ABSTRACT: Fibroblasts are the main dermis-resident cells, yet they remain poorly characterized. Fundamentally, fibroblasts originate from the same population of mesenchymal cells but fibroblast subpopulations have been identified. Papillary and reticular fibroblasts were characterized based on their respective location in the papillary and reticular dermis. Here, we identified a new subset of fibroblasts, located in human papillary dermis and displaying specific features such as their organization into cell clusters. We used microarrays to detail the gene expression profiles of the novel fibroblast subpopulation we have identified compared to papillary and reticualr fibroblasts and notably the relative contribution of the different fibroblast subpopulations to the extracellular matrix of the dermis.

Human dermal fibroblast subpopulations display distinct gene signatures related to cell behaviours and matrisome.

Project description:The upper papillary and deeper reticular dermis differ structurally and functionally. Although the papillary and reticular fibroblasts produced distinct extracellular matrix (ECM), the matrisome of these two fibroblast subpopulations has not been defined. Therefore we performed a transcriptomic analysis of papillary and reticular fibroblasts freshly isolated from skin of young donors around 20, at a time they produced high level of ECM. Bioinformatics analysis delivered 230 upregulated and 139 downregulated transcripts in papillary vs reticular fibroblasts. Expression of various selected genes was validated by q-PCR. The papillary fibroblasts were characterized by a higher expression of genes involved in the defense function, in the regulation of cell motility and proliferation and in the MAPK cascade whereas reticular fibroblasts showed higher expression of genes related to the development of connective tissues. Papillary fibroblasts were characterized by the expression of a high number of matrisome-associated genes whereas reticular fibroblasts gene signature mainly related to the core matrisome and ECM regulators. The regulation of selected genes was validated at protein level attesting to the robustness of the transcriptome analysis. Altogether, our data brought new insights into an ECM signature that is coherent with the organization and function of the papillary and reticular dermis.

The lower, reticular dermis is composed of solid tissue made up of thicker highly dense fibers. Spatial and single-cell transcriptional profiling identifies functionally distinct human dermal fibroblast subpopulations

Project description:Fibroblasts synthesize the extracellular matrix of connective tissue and play an essential role in maintaining tissue integrity. We have previously shown that mouse skin connective tissue, the dermis, is comprised of functionally distinct fibroblast lineages. However, the extent of fibroblast heterogeneity in human skin is unknown. Here, using a combination of spatial transcriptional profiling of human and mouse dermis and single cell transcriptional profiling of human dermal fibroblasts, we show that there are at least four distinct fibroblast populations in adult human skin. We define markers permitting prospective isolation of these cells and show that although marker expression is rapidly lost in culture, different fibroblast subpopulations retain distinct functionality in terms of Wnt signalling, T cell communication and the ability to support human epidermal reconstitution in organotypic culture. Furthermore, while some fibroblast subpopulations are spatially segregated, others are not. These findings have profound implications for normal wound healing and diseases characterized by excessive fibrosis, and suggest that ex vivo expansion or in vivo ablation of specific fibroblast subpopulations may have therapeutic applications.

Prrx1 fibroblasts represent a pro-fibrotic lineage in the mouse ventral dermis

Project description:Skin scarring following dermal injury causes extreme pain and pschological trauma for patients. Currently, we do not have effective treatments to prevent or reverse skin scarring. Fibroblast heterogeneity has been shown within the unwounded mouse dorsal dermis, with fibroblast subpopulations being identified according to anatomical location and embryonic lineage. Using RNA-sequencing and single cell RNA sequencing, we demonstrate that Prrx1-expressing mouse fibroblasts are responsible for acute and chronic scaring in the ventral mouse dermis. In summary, we have identified and characterized a fibroblast subpopulation in the mouse ventral dermis with intrinsic scar-forming potential.

Project description:Skin fibrotic disease representsa major global healthcare burden, characterized by fibroblast hyperproliferation and excessive accumulation of extracellular matrix.Fibroblasts are found to be heterogeneous in multiple fibrotic diseases,but the fibroblast heterogeneity of skin fibrotic diseases remains unknown.In this study, we performed single-cell RNA-seq in keloid, a paradigm of skin fibrotic diseases, andnormal scardermis tissues.Our results indicate thatkeloid and normal scar fibroblasts could be divided into 4 subpopulations: secretory-papillary, secretory-reticular, mesenchymal and pro-inflammatory.The percentage of mesenchymal fibroblast subpopulationincreased significantly in keloid compared to normal scar. Interestingly, we also found increasing mesenchymal fibroblast subpopulation in scleroderma, another skin fibrotic disease.Function studies showed that the mesenchymal fibroblasts promoted collagen synthesis of the other fibroblasts in keloid partiallythrough secreting POSTN. These findings will help us understandskin fibroticpathogenesis in depth,and provided potential target cells for fibrotic diseases therapies.

Project description:The aim of our study was to determin if papillary and reticular fibroblasts cell sheets expressed distinct genes involved in angiogenesis regulation and extracellular matrix. Thanks to our RNAseq analyssis, we demonstrated that papillary and reticular fibroblasts express specific signature of genes related to secreted angiogenic regulators and matrisome genes, suggesting that each subtype of fibroblasts differently regulate the formation of capillary via secreted factors and the microenvironment they generate. We confirmed this hypothesis with functional angiogenesis model in vitro. Overall, we show that papillary fibroblasts have a greater angiogenic potential and support the formation of dense branched vascular netword. On the other hand, reticular fibroblasts allow the formation of fewer vessel of larger diameter. This resultats are coherent with native skin vasculature and suggest that each fibroblasts subtypes play a role in regulating skin vascularisation.

Cell-derived matrices from human dermal fibroblasts (HDF), skin melanoma associated fibroblasts (MAF) and two different lymph node fibroblast reticular cells (FRC) were denuded of cells and their composition was analyzed by mass spectrometry.

Parallel single cell multi-omics analysis of neonatal skin reveals transitional fibroblast states that restrict differentiation into distinct fates

Project description:One of the keys to achieving skin regeneration lies within understanding the heterogeneity of neonatal fibroblasts, which support skin regeneration. However, the molecular underpinnings regulating the cellular states and fates of these cells are not fully understood. To investigate this, we performed a parallel multi-omics analysis by processing neonatal murine skin for single-cell ATAC-sequencing (scATAC-seq) and single-cell RNA-sequencing (scRNA-seq) separately. Our approach revealed that fibroblast clusters could be sorted into papillary and reticular lineages based on transcriptome profiling, as previously published. However, scATAC-seq analysis of neonatal fibroblast lineage markers, such as, Dpp4/CD26, Corin, and Dlk1 along with markers of myofibroblasts, revealed accessible chromatin in all fibroblast populations despite their lineage specific transcriptome profiles. These results suggests that accessible chromatin does not always translate to gene expression and that many fibroblast lineage markers reflect a fibroblast state, which includes neonatal papillary, reticular, and myofibroblasts. This analysis also provides a possible explanation as to why these marker genes can be promiscuously expressed in different fibroblast populations under different conditions. Our scATAC-seq analysis also revealed that the functional lineage restriction between dermal papilla and adipocytes fates are regulated by distinct chromatin landscapes.
