Characterizing a role for HNRNPA2B1 during normal myogenesis

ABSTRACT: The RNA localization mechanisms in muscle are incompletely known however numerous RNA binding proteins and RNA aggregate into higher order amyloid-like assemblies in degenerative muscle disease. Here we assign a role for the RNA binding protein hnRNPA2B1, heterogeneous RNA binding protein A2/B1, in normal myogenesis. Loss of hnRNPA2/B1 impairs muscle differentiation in vitro. hnRNPA2B1 binds to the 3’UTR of select myogeneic transcripts distinct from other RNA-binding proteins during muscle differentiation. hnRNPA2B1 associated RNPs are capable of assembling into higher order microtubule associated RNP assemblies and redistribute from the nucleus into the cytoplasm during muscle differentiation both in vitro and in vivo. Together, our results contribute to a broader theme in RNA localization during myogenesis and may provide additional insights into the origins of pathological cytoplasmic RNP aggregation.
