Project description:The downregulated pathways in early pregnant CL unraveled in this study span a wide range of luteal functions: angiogenesis, apoptosis and survival, steroidogenesis, and ECM remodeling. Notably, selective DEGs in luteinized granulosa cells (LGCs) were modulated by IFNT in vitro in a similar manner to their regulation in the CL of P cows, suggesting that IFNT may contribute to the characteristics of early pregnant CL.

Project description:The study aimed to identify miRNAs expression profiles associated with growth and regression of dominant-size follicles in bovine. Follicles were collected from abattoir ovaries and their status (healthy/atretic) was assessed by measuring steroid levels and aromatase expression. Corpora lutea (CL) were collected from ovarian pairs correponding to days 1 to 4 of the oestrus cycle. Total RNA was isolated from whole follicles at different developmental stages and from CL. An heterologous microarray (Exiqon, Denmark) approach followed by RT-qPCR validation (Qiagen, UK) was used to identify and compare miRNA profiles between large healthy follicles (diameter, 13–16 mm, n=6) and each of small (4–8 mm, n=6 pools of follicles), large atretic folllicles (13-16 mm, n=6) and CL (n=6). RNA from the above groups was hybridized to the miRCURY LNA™ microRNA Hi-Power Labeling Kit,Hy3™/Hy5™ (Exiqon) and hybridized on the miRCURY LNA™ microRNA Array (6th gen). A total of 17 and 57 microRNAs were differentially expressed (> 2 fold, adj. P-value < 0.05) between Large Healthy and each of Small and Large Atretic follicles, respectively, a fraction of which corresponded to registered bovine miRNA sequences. A subset of 5 bovine miRNAs (miR-144, miR-202,vmiR-451, miR-652, miR-873) were confirmed by qPCR to be upregulated in Large Healthy follicles, were enriched in mural granulosa cells and their predicted targets mapped to genes involved in follicular cell proliferation and differentiation, suggesting an involvemet of this subset of microRNAs in ovarian follicle development. In addition, the miR-183-96-183 cluster was identified as upregulated in CL and was shown to be involved in luteal survival and progesterone production.

Project description:Next-generation sequencing (RNA-Seq) was performed on CL from the late luteal phase and compared with normally luteolyzing CL collected at the prepartum P4 decrease, and following antigestagen (Aglepristone)- treatment. The analysis of transcriptomes presented herein supports the hypothesis describing luteal regression in non-pregnant dogs as a degenerative process devoid of the acute luteolytic principle and without an acute involvement of the immune system observed prepartum. The contribution of the immune system seems, however, to be critical in the PGF2alpha-mediated active prepartum luteolysis, which appears to be an acute immune process. The antigestagen-medited effects point towards the withdrawal of the luteotropic function of P4 with lesser involvement of immune system than during natural luteolysis. In summary, a deeper insights have been obtained into possible endocrine, paracrine and autocrine mechanisms governing the luteal life span in the domestic dog during pregnancy and in non-pregnant cycles.

Project description:Corpus luteum (CL) is an ephemeral gland whose main function is to secrete progesterone required for the establishment and maintenance of pregnancy. In non-fertile cycles, primate CL has a finite life span of 14-16 days, following which it undergoes regression. Although it has been suggested long time back that PGF2alpha of intra-ovarian or intra-luteal origin acts as a physiological luteolysin in primates, the mechanisms by which PGF2alpha mediates its luteolytic actions are poorly understood. Earlier, we standardized an induced luteolysis model, where 3 injections of PGF2alpha to female bonnet monkeys on day 10 of luteal phase led to luteolysis. To delineate the mechanism of this PGF2alpha-induced luteolysis, we tried to study the global changes in gene expression following PGF2alpha treatment using Affymetrix microarray analysis of PGF2alpha and VEH treated CL and results suggested that PGF2alpha exerts its luteolytic effects by altering gene expression. Project description:The LH-like molecule chorionic gonadotropin (CG) is secreted by the macaque conceptus during and following implantation, M-bM-^@M-^\rescuingM-bM-^@M-^] the CL from impending regression and extending its functional lifespan in early pregnancy for approximately two weeks. CG binds to the same receptor as LH; i.e., LHCGR, and promotes production of steroids and other factors such as relaxin (RLN1). Our research group recently used AffymetrixM-bM-^DM-" rhesus macaque total genome arrays to compare the effects of CG on the luteal transcriptome from rhesus females during simulated early pregnancy (SEP) with changes during luteal regression in the non-fecund menstrual cycle. This analysis demonstrated that CG-rescue affected expression levels of 4,500 mRNA transcripts between days 10 and 15 of the luteal phase. Previous analyses indicated that a portion of the transcriptome in the macaque CL of the menstrual cycle was regulated indirectly by LH via the local actions of steroid hormones, including progesterone (P). Therefore, this study was designed to distinguish CG-regulated luteal genes that are dependent versus independent of local steroid (P) action. A protocol of increasing dosages of hCG (SEP) was begun on day 9 of the luteal phase in rhesus females combined with concurrent administration of the 3BHSD inhibitor trilostane (TRL) +/- the synthetic progestin (P) R5020. CL were collected on day 10 (no treatment) of the luteal phase to serve as a baseline comparison (n=8), 1 day of SEP (Day 10+hCG+/-TRL+/-R5020) and 6 days of SEP (Day 15+hCG+/-TRL+/-R5020); n=4/group. In the presence of CG, treatment with TRL reduced serum P levels to less than 1 ng/ ml after 1 day and all of the Day 15+h+TRL-treated females initiated menses before CL collection. The isolated CL were processed for total RNA and hybridized to microarrays. Compared to hCG treatment alone, 50 significantly altered mRNA transcripts were identified on day 10, rising to 95 on day 15 (P<0.05, M-bM-^IM-% 2-fold change in gene expression). The mRNA levels for several genes were validated in CL by real-time PCR. RNL1 levels increased with CG-treatment, but were not affected by steroid ablation, similar to previously reported relaxin protein expression. Steroid-sensitive genes included CDH11, IL1RN, INSL3, LDLR, OPA1, SERPINE1, SFRP4, and TNSF13B1; however differential sensitivity was observed and effects of steroid ablation and P replacement varied by day. Expression of some genes (e.g., 3BHSD2, ADAMTS1, ADAMTS5, MMP9, STAR, and VEGFA) previously identified as steroid regulated in the macaque CL during the menstrual cycle were not significantly altered by steroid ablation and P replacement during CG exposure in SEP. These data indicate that the majority of CG-regulated luteal transcripts are differentially expressed independently of local steroid actions. The proportion of steroid sensitive mRNA transcripts in the presence of CG is much smaller than in the presence of LH during the non-fecund cycle. Nevertheless, the steroid-regulated genes in the macaque CL may be essential during early pregnancy, based on the previous report that TRL treatment initiates premature structural regression of the CL during SEP. These data reinforce the concept that the structure, function, and regulation of the rescued CL in early pregnancy is different from the CL of the menstrual cycle. Project description:A large body of evidence suggests that the development and maintenance of corpus luteum (CL) in primates requires the action of LH. Earlier, using CET-induced luteolysis model, we demonstrated changes in luteal transcriptome suggesting nuclear actions of LH in the primate CL. To further demonstrate the role of LH in maintenance of primate CL, replacement studies were carried out and it was observed that administration of a single injection of rhLH was sufficient to restore the progesterone to pre-CET treatment levels and prevent the CET-induced luteolysis. To elucidate the molecular mechanisms underlying the rescue of CL function, we used LH-replacement model to study immediate early changes in gene expression at a global level (Affymetrix oligonucleotide microarray) following LH replacement in CET-treated monkeys and to evaluate if the changes in gene expression mediated by LH-withdrawal can be reversed by LH replacement. Results demonstrated up-and down-regulation of various genes following LH replacement and suggested that LH-withdrawal induced changes in gene expression are reversible at least for some genes. Project description:The success of the early reproductive events depends on an appropriate communication between gametes/embryos and the oviduct. Extracellular vesicles (EVs) contained in oviductal secretions have been suggested as new players in mediating this crucial cross-talk by transferring their cargo (e.g. proteins, mRNA and small ncRNA) from cell to cell. Therefore, the objective of the project was to analyze the protein content of EVs (exosomes and microvesicles) contained in the bovine oviductal fluid at different stages of the estrus cycle. For this purpose, pairs of oviducts with their attached ovaries were collected from cyclic cows at a local abattoir under different hormonal regulation along the estrous cycle. Oviducts were classified into one of the 4 stages of the estrous cycle according to the morphology of the corpus lutea(CL) and follicle populations: Stage 1 (S1): showed recently ovulated follicle (d 1 to 4 of estrous cycle); Stage 2 (S2): early luteal development with medium or large or both present (d 5 to 11); Stage 3 (S3): fully functional CL yellow or orange in color (d 11 to 17); or Stage 4 (S4): regressing CL with little vasculature and a large preovulatory follicle present (d 18 to 20). EVs were collected by ultracentrifugation and prepared for mass spectrometry analysis.
