Transcriptional profiling of HT-29 cells with different treatments

ABSTRACT: Through RNA-seq, we investigated the transcriptional changes of necroptotic cell induced by TNF-α, SM-164 and zVAD. We report here that necroptosis leads to transcriptional activation of a large set of genes including pro-inflammatory cytokines, which is much stronger than that triggered by TNF-α alone. Furthermore, the robust induction of cytokine expression could be blocked by necroptotic inhibitor Nec-1s. Thus, we show that necroptosis exerts a profound effect on transcription, and activate the expression of pro-inflammatory genes, providing insights into the mechanism by which necroptosis promotes inflammation in human diseases.
