Project description:Changes of genome-wide mRNA transcription levels of human ciliary smooth muscle (hCSM) cells were determined by treating hCSM cells in culture with 200 nM of either an prostaglandin E2 receptor subtype EP2 or subtype EP4 selective agonist for 6 hours in comparison to untreated controls. This was followed by competitive hybridization of fluorescent Cy3 or Cy5 labeled cRNA probes derived from the treated versus untreated control total RNA samples onto an Agilent Human Whole Genome Expression oligonucleotide microarray. Log 2 (LN) of the intra-slide ratios (RATIO, PRE_VALUE) of treated versus untreated samples was reported as VALUE in the sample files. Global gene expression of human iPSC directed differentation towards smooth muscle cells using an Acta2eGFP human iPSC reporter line

Project description:Here, we generated and differentiated a human induced pluripotent stem cell line with an ACTA2eGFP reporter towards a smooth muscle-like cell that can be purified and expresses characteristic markers of smooth muscle cells. We performed microarray analysis of day 30 ACTA2eGFP+ and ACTA2eGFP- sorted populations, and the transcriptomic profile of the ACTA2eGFP+ cells is reminiscent of an immature or a synthetic smooth muscle cell phenotype.

Project description:Human coronary smooth muscle cells were treated with two different ß-blocker (metoprolol and nebivolol). RNA from three replicates of each, treated and the untreated control group, were isolated and the expression profiles were determined using Affymetrix Human Genechip U133A arrays. Comparisons between the sample groups allow the identification of genes with different expression patterns between the treated and untreated control cells. Transcriptomic effects of estradiol treatment on cultured human uterine smooth muscle cells

Project description:The myometrium is an important reproductive tissue composed primarily of smooth muscle cells. Contractility of the myometrial smooth muscle cells during the estrous cycle and pregnancy is modulated by estrogen. Despite much research, little is known about the molecular mechanism by which estrogen regulates myometrial contractility. This study investigates global gene expression profile of cultured human uterine smooth muscle cells (hUtSMCs) following 17M-NM-2-estradiol (E2) treatment using cDNA microarray technology. In response to E2 treatment 540 genes were identified as significantly differentially expressed. Gene ontology analysis identified several significant biological processes for these genes including muscle contraction, gland development, cell migration, cell adhesion, apoptosis, response to external stimulus and phosphorylation. In this study evidence is presented that in human smooth muscle cells 17M-NM-2-estradiol, contrary to expectations, altered expression of several genes that may favor myometrial relaxation. Genes related to focal adhesion function were downregulated suggesting a loosening of SMC connections to the ECM while upregulation of MMP9 is also associated with weakened ECM interactions. The hUtSMC system is an additional useful model to investigate steroid effects on smooth muscle cells in isolation from other myometrial cell types. In this study human myometrial smooth muscle cells (hUtSMC) purchased from Lonza were cultured and treated with 17beta estradiol. RNA was isolated after 6h, 24h and 72h from both treated and untreated control cells. The experiment was repeated at least 3 times.

JNK-induced genes in rat vascular smooth muscle cells are defined as those induced by adenoviruses encoding constitutively active MKK7 (caMKK7) + wild type JNK1 (wtJNK1) and suppressed by adenovirus encoding dominant negative JNK1 (APF). JNK-suppressed genes in rat vascular smooth muscle cells are defined as those suppressed by adenoviruses encoding constitutively active MKK7 (caMKK7) + wild type JNK1 (wtJNK1) and induced by adenovirus encoding dominant negative JNK1 (APF). Adenovirus encoding nuclear localizing GFP was used as a control.

Project description:We developed a new method for the isolation of bronchial smooth muscle cells. This method is built upon a double fluorescent mouse SMA-GFP;NG2-DsRed and cell sorting. Using this method, we isolated bronchial smooth muscle cells from control and asthmatic C57BL/6J mice for gene profiling. Double fluorescent mice were induced to develop asthma using OVA. Bronchial smooth muscle cells were purified from asthmatic and control mice for mRNA array.

Global gene expression of mouse iPSC directed differentation towards smooth muscle cells using an Acta2hrGFP mouse iPSC reporter line

Project description:Here, we generated and differentiated a mouse induced pluripotent stem cell line with an Acta2hrGFP reporter towards a smooth muscle-like cell that can be purified and expresses characteristic markers of smooth muscle cells. We performed microarray analysis of 3 timepoints in our smooth muscle directed differentiation protocol and compared it to primary adult aortic smooth muscle cells. The profiles indicated that the day 13 Acta2hrGFP+ and Acta2hrGFP- populations are fairly similar, and that the day13 Acta2hrGFP+ population's transcriptomic profile is reminiscent of an immature or a synthetic smooth muscle cell phenotype. Using a mouse iPSC line with a transgenic GFP reporter for Acta2, we characterized the global transcriptomic proifle of cells during different developmental time points in our in vitro differentiation (undifferentiated iPSC, sorted Kdr+ mesodermal progenitors, and sorted Acta2hrGFP+ and Acta2hrGFP- candidate SMCS). We also included freshly isolated primary aortic Acta2hrGFP+ smooth muscle cells as a control.
