Project description:Hematopoietic stem cells (HSCs) in the bone marrow are able to differentiate into all types of blood cells and supply the organism each day with billions of fresh cells. They are applied to cure hematological diseases such as leukemia. The clinical need for HSCs is high and there is a demand for being able to control and multiply HSCs in vitro. The hematopoietic system is highly proliferative and thus sensitive to anti-proliferative drugs such as chemotherapeutics. For many of these drugs suppression of the hematopoietic system is the dose-limiting toxicity. Therefore, biomimetic 3D models of the HSC niche that allow to control HSC behavior in vitro and to test drugs in a human setting are relevant for the clinics and pharmacology. Here, we describe a perfused 3D bone marrow analog that allows mimicking the HSC niche under steady-state and activated conditions that favor either HSC maintenance or differentiation, respectively, and allows for drug testing.

Inflammation of the Aged Bone Marrow Niche Drives Blood Aging

Project description:Hematopoietic aging is defined by a loss of regenerative capacity and skewed differentiation from hematopoietic stem cells (HSC) leading to dysfunctional blood production. Signals from the bone marrow (BM) microenvironment dynamically tailor hematopoiesis, but the effect of aging on the niche and the contribution of the aging niche to blood aging still remains unclear. Here, we show the development of an inflammatory milieu in the aged marrow cavity, which drives both niche and hematopoietic system remodeling. We find decreased numbers and functionality of osteogenic endosteal mesenchymal stromal cells (MSC), expansion of pro-inflammatory perisinusoidal MSCs, and deterioration of the central marrow sinusoidal endothelium, which together create a self-reinforcing inflamed BM milieu. Single cell molecular mapping of old niche cells further confirms disruption of cell identities and enrichment of inflammatory response genes. Inflammation, in turn, drives chronic activation of emergency myelopoiesis pathways in old HSCs and multipotent progenitors, which promotes myeloid differentiation at the expense of lymphoid and erythroid commitment, and hinders hematopoietic regeneration. Remarkably, both defective hematopoietic regeneration, niche deterioration and HSC aging can be improved by blocking inflammatory IL-1 signaling. Our results indicate that targeting the pro-inflammatory niche milieu can be instrumental in restoring blood production during aging.

Kinetic barriers under steady-state conditions.

Project description:It is shown by analysis of a numerical example that kinetic barrier diagrams [Südi (1991) Biochem. J. 276; 265-268] are also useful for displaying the phenomenological resistance of an enzymic turnover to net chemical fluxes observed under steady-state conditions. Most importantly, a new additivity rule is revealed by net flux profiles, which refers to limiting ('ideal') conditions. The rule states that the net fluxes that one obtains under initial steady-state conditions are strictly identical with the overall flux in the corresponding equilibrium system. This finding amounts to defining the relation between unidirectional fluxes and net fluxes, and explains why the total 'one-way flux resistance' to the overall reaction at chemical equilibrium on the one hand, and the 'net flux resistance' to both initial steady-state reactions on the other hand, have to be equal. The conclusions are claimed to be generally valid for consecutive chemical reactions.
