An investigation of the effect of anti-apoptosis mutations on human pluripotent stem cells transcriptome when apoptosis stress was applied

ABSTRACT: We performed RNA-sequencing on hPSCs treated with mitosis poison nocodazol (NOC) or MCL1 inhibitor A1210477 (A121). Our data revealed that NOC and A121 treatment lead to significant upregulation of apoptosis related genes as well as NIK/NF kappa B pathway gene. To investigate effects of anti-apoptotic mutations on hPSCs under stress, we also preformed RNA-seq on BCL-XL overexpression cells and NOXA-/- cells with NOC or A121 treatment. Results showed that anti-apoptotic mutations can promote hPSCs survival by inhibiting both apoptosis pathway and NIK/NF kappa B pathway. Furthermore, SNP analysis showed that cells with anti-apoptotic mutation gained more SNP under NOC or A121 treatment.
