RNA-seq profiling of FACS purified MPs from adult skeletal muscle after conditional knockout of Hic1 [MP RNA-seq]

ABSTRACT: To determine the effect of the deletion of the trancriptional repressor Hypermethylated in cancer 1 (Hic1) on quiescence, adult mesenchymal progenitors (MPs) were isolated from skeletal muscle of both wild type mice and those conditionally deleted for Hic1 and profiled by RNA sequencing. Comparison to MPs activated by using a muscle injury model of the tibialis anterior muscle revealed similar pathways affected under both conditions, highlighting the role for Hic1 in maintaining the quiescent state of MPs

Project description:To determine the effects of long term deletion of the transcriptional repressor Hypermethylated in cancer 1 (Hic1) on skeletal muscle, samples were collected of both wild type mice and those conditionally deleted for Hic1 and profiled by RNA sequencing. Aspects of muscle homeostasis were negatively impacted, highlighting cell and non cell autonomous roles for Hic1 in maintaining functional MPs and muscle tissue.

Gene expression in human prostate cancer cell lines regulated by HIC1 gene

Project description:This study aimed to further our understanding of the role that hypermethylatioted in cancer 1 (HIC1) plays in prostate cancer (PCa) development. Microarrays were searched for some genes that had correlated expression with HIC1 mRNA. Our data showed that HIC1 promoter hypermethylation was presented in cell lines, tissues and plasma of PCa patients. According to fold-change screening between restoring expression of HIC1 and its respective control cells, both up-regulated and down-regulated genes were commonly observed in PC3 and C4-2B cells. The restoring expression HIC1 in PCa lines were respectively noted as PC3-HIC1 and C4-2B-HIC1 cells, and the respective controls were noted as C4-2B-GFP and PC3-GFP cells.

Effect of HIC1 and miR-212/132 expression on MCF7 cells

Project description:To identify the target of miR-212, miR-132 and HIC1, we have employed whole genome microarray expression profiling on the human breast cancer MCF7 cells. To generate miR-212/132 or HIC1 inducible MCF7 cells, doxycycline-dependent miR-212/132 or HIC1 gene expression system was used. Either Tet-ON miR-212/132 MCF7 or Tet-ON HIC1 MCF7 were treated with 1μg/ml of Doxycycline for 36 hours with EMEM containing 0.01 mg/ml bovine insulin and 10% FCS. Two independent experiments were performed.

Microarray analysis of Normal and MPS VII mouse aorta

Project description:Mucopolysaccharidosis VII (MPS VII) is due to mutations within the gene encoding the lysosomal enzyme beta-glucuronidase, and results in the accumulation of glycosaminoglycans. MPS VII causes aortic dilatation and elastin fragmentation. In this study we performed microarray analysis of ascending aortas from normal and MPS VII mice, trying to find out possible genes responsible for the phenotype observed. In addition, during our breeding strategy, we noticed that some MPS VII mice had less dilated aortas, and we proposed that an yet-unidentified gene could be responsible for the difference observed. We therefore included in the analysis two MPS VII mice with aortas that were not dilated. Total RNA extracted from ascending aortas from 3 Normal mice, 3 MPS VII mice with dilated aortas and 2 MPS VII mice with aortas that were not dilated.

Project description:To explore the role of HIC1 silencing in regulating PCa EMT development, Human Gene Expression Microarrays were searched for altered genes upon silencing HIC1 expression in C4-2B and DU145 cells. According to fold-change screening betweenHIC1-silenced and its respective control cells, both up-regulated and down-regulated genes were shown. Human PCa cell lines C4-2B and DU145 were transfected with lenti-virus (shRNA targeting HIC1 and respective control). HIC1-silenced cells were respectively noted as C4-2B-shHIC1 and DU145-shHIC1. Their respective control cells were respectively noted as C4-2B-shctrl and DU145-shctrl.
