Identification of transcriptional targets of FGF20.

ABSTRACT: Fgf20 is expressed from the hair follicle placode and is required for development of the dermal condensate. Here we used RNAseq to identify the immediate transcriptional targets of FGF20 in Fgf20-/- dermis at E13.5, a timepoint immediately prior to dermal condensate initiation.

SKPs derive from hair follicle precursors and exhibit properties of adult dermal stem cells

Project description:Global expression analysis of neural crest-like skin-derived precursors (SKPs) and Sox2-positive follicle dermal cells that SKPs originate from. In spite of the remarkable regenerative capacity of mammalian skin, an adult dermal stem cell has not yet been identified. Here, we provide evidence that SKPs, multipotent neural crest-like skin-derived precursors, represent an adult dermal stem cell. When transplanted into adult skin, SKPs can reconstitute the adult dermis, contribute to dermal wound-healing, home to a hair follicle niche, and instruct epidermal cells to make hair follicles. Hair follicle-derived SKPs self-renew, maintain their multipotency, and serially reconstitute hair follicles. The endogenous origin of SKPs are Sox2-positive follicle dermal cells that share a similar global gene expression profile with SKPs. These endogenous cells home back to their follicle niche, induce hair follicle morphogenesis, and differentiate into neural and dermal progeny. Hair follicle-associated dermal cells will move out of their niche to contribute to dermal maintenance and wound-healing. These studies therefore identify a dermal stem cell, and provide a biological rationale for the presence of a multipotent precursor within adult dermis, findings with important therapeutic implications.

Single-cell RNA-seq of the developing C57/BL6 mouse back skin at E13.5, E16.5 and P0

Project description:We performed single-cell RNA seq on C57/BL6 mouse back skin at E13.5, E16.5, and P0 to study embryonic hair follicle development. We analyzed 15,086 single cell transcriptome profiles from E13.5, E16.5 and newborn mice (postnatal day 0, P0) dorsal skin cells across hair follicle induction, organogenesis, cytodifferentiation stage. Based on t-distributed Stochastic Neighbor Embedding (tSNE) clustering, we identified 14 cell clusters from skin cells and delineated their cell identity gene expression profile. By using Monocle pseudotime ordering analysis, we constructed epithelium/dermal cell lineage differentiation trajectory and revealed sequential activation of key regulons involved during embryonic hair follicle morphogenesis. Our findings here provide molecular landscape during hair follicle epithelium/dermal cell lineage fate decisions.

Obtaining DPPCs with trichogenic ability is critical for human HF regeneration because dermal papilla cells (DPCs) rapidly lose their trichogenic ability in culture. Here, we generated trichogenic DPPCs from human induced pluripotent stem cells (iPSCs) via neural crest stem cells (NCSCs), based on the developmental evidence at the hair placode stage. SDC1+CD133− cells showed signature DP gene expression, spontaneous sphere formation and represented intermediate population in the differentiation way from NCSCs to DPCs. hiPSC-derived DPPCs generated HF equivalents in vitro and reconstituted de novo human HFs in vivo combined with hiPSC-derived epithelial stem cells. Remarkably, trichogenic ability of DPPC was only proven in the specific time window, providing insights into the loss of trichogenicity in cultured DPC. Thus, this study provides an in vitro model for studying DPC development and biology.

We used microarrays to analyze differences in gene expression between alpha-6 integrin+CD133+ basal cells and other human fetal epidermal populations to provide candidates defining this unique hair follicle placode subpopulation.

The authors confirm similar age-related decline in function is present in adult human DPCs. RNA-Seq is utilized to identify further explanations for this functional decline in aged hDPCs.

Transcription profiling of mouse dermis from epithelial activated beta-catenin mutant embryo

Project description:Î²-catenin signaling is required for hair follicle development, but it is unknown whether it is sufficient to activate expression of hair follicle genes in embryonic skin. To address this we profiled gene expression in dermis from E15.5 KRT14-Cre Ctnnb1(Ex3)fl/+ embryos carrying an activating mutation in epithelial beta-catenin, and control littermate embryos. Experiment Overall Design: Total dermal RNA from two KRT14-Cre Ctnnb1(Ex3)fl/+ and two control littermate E15.5 embryos was hybridized to Affymetrix GeneChip Mouse Genome MOE430 2.0 oligonucleotide microarrays. Experiment Overall Design: Appended below is Table S2: Full list of differentially expressed genes in KRT14-Cre Ctnnb1(Ex3)fl/+ mutant compared with control littermate dermis at E15.5, including normalization and filter parameters. Fold change, listed in the second column, gives the ratio of normalized mutant : control transcript levels.
