Induced pluripotent stem cells of patients with Tetralogy of Fallot reveal transcriptional alterations in cardiomyocyte differentiation

ABSTRACT: We generated induced pluripotent stem cells (iPSCs) of two patients with Tetralogy of Fallot (TOF) and three healthy relatives of two unrelated familes. We furhter performed mRNA sequencing of iPSCs (day 0) and derived cardiomyocytes (CMs) at day 15 and 60 of patients and healthy relatives.

mRNA expression profiling in patients with Tetralogy of Fallot and healthy unaffected individuals

Project description:Right ventricular mRNA profiles from 22 patients with Tetralogy of Fallot (TOF) and mRNA-seq profiles from the left and right ventricle (LV and RV, respectively) of 4 healthy unaffected individuals (NH) were generated. The total RNA was isolated from the 30 human heart samples using TRIzol. mRNAs were isolated from total RNA and prepared for sequencing using Illumina Kit RS-100-0801 according to the manufacturer's protocol (Preparing Samples for Sequencing of mRNA Sept 2008). The sequencing libraries were generated using a non-strand-specific library construction method. The purified DNA fragments were used directly for cluster generation, and 36 cycles of single-end read sequencing were performed using the Illumina Genome Analyzer. Sequencing reads were extracted from the image files using the open source Firecrest and Bustard applications (Illumina Genome Analyzer pipeline 1.3.2, 1.4.0 and 1.5.0). Right ventricular mRNA-seq profiles from 22 patients with Tetralogy of Fallot and mRNA-seq profiles from the left and right ventricle of 4 healthy unaffected individuals. Sexual difference of small RNA expression in Tetralogy of Fallot

Project description:Small RNAs, especially the microRNAs, have been revealed to play great roles in heart development and congenital heart defects. Several studies have shown dysregulated miRNAs in ventricular tissues of Tetralogy of Fallot (TOF) patients. In the present study, we conducted high throughput sequencing to obtain the global profiling of small RNA transcriptome in heart right ventricular samples from 10 age -matched TOF patients. These samples showed dominant composition of miRNA and mitochondrial associated RNAs. By sRNA cluster identification and differential gene expression analysis, significant sexual difference was discovered for sRNA expression in TOF patients. miR-1/miR-133, the first identified miRNA cluster essential for cardiac development, account for the most variance of sRNA expression between sexes. Our results suggested divergent molecular basis between female and male TOF patients.

Analysis of Amniotic Fluid-Derived Exosomal miRNAs in Fetuses with Tetralogy of Fallot

Project description:Tetralogy of Fallot (TOF) is the most common cyanotic congenital heart disease (CHD). Amniotic fluid (AF) contains a higher abundance of biological compounds which could direct reflect the information of fetal health. AF-derived exosomal miRNAs have been investigated as etiological factors in the pathogenesis of TOF. The purpose of this study was to analyze the expression of AF-derived exosomal miRNAs and their roles in TOF development. In this study, we identified differentially expressed (DE)-miRNAs between TOF group and control group. A total of 257 miRNAs were differentially expressed (DE) between TOF and control groups. KEGG pathway enrichment demonstrated that these target genes of DE-miRNAs were involved in Wnt and Notch signaling pathway. Twenty five of 257 DE-miRNAs were overlapped with Notch-regulated and Wnt-regulated miRNAs simultaneously.
