Adipose tissue of overfed animals during and after overfeeding

ABSTRACT: We report the transcriptomes (polyadenylated mRNAs over 200bp) of animals overfed through intragastric tubes either in the overfed state (OIO) or 24 hours after cessation of overfeeding (OIOneg). We also report transcriptomes of weight-matched mice either fed (ALO, or fasted 24 hours (ALOneg).

Protection against overfeeding-induced weight gain in mice is preserved in obesity but does not require FGF21 or MC4R

Project description:Experimental overfeeding triggers homeostatic compensatory mechanisms that counteract weight gain. Here, we utilized intragastric overfeeding in mice to investigate the physiological and molecular responses to forced weight gain. Both lean and diet-induced obese (DIO) mice exhibited a potent and prolonged lowering of voluntary food intake following overfeeding-induced weight gain. Although overfeeding resulted in a marked increase in circulating fibroblast growth factor 21 (FGF21), experiments with FGF21 knockout (KO) mice demonstrated that FGF21 is dispensable for the homeostatic defense against experimental weight gain. Targeted proteomics unveiled novel circulating factors linked to overfeeding, including the protease legumain (LGMN). Notably, administration of recombinant LGMN lowered body weight and food intake in DIO mice. The protection against weight gain was also associated with reduced vascularization in the hypothalamus and sustained reductions in transcript levels of the orexigenic neuropeptides, Npy and AgRP, suggesting a role of hypothalamic signaling in the homeostatic recovery from overfeeding. Overfeeding of melanocortin 4 receptor (MC4R) KO mice showed that these mice can suppress voluntary food intake and counteract the enforced weight gain, although their rate of weight recovery is impaired. Collectively, these findings demonstrate that the defense against overfeeding-induced weight gain remains intact in obesity and involves mechanisms independent of both FGF21 and MC4R.

Juvenile postnatally overfed mice heart microarray anaylsis

Project description:C57BL/6 mice underwent early postnatal overfeeding by litter size reduction (3 pups/dam, instead of 10 pups/dam, in postatally normally fed mice). These overfeeding conditions lead to early and permanent modifications of body weight and to an increase in body fat mass in the adulthood. When these postnatally overfed mice become adult, they develop several cardio-metabolic alterations (mild hypertension, impairment of heart contractility, remodeling, fibrosis, hyperinsulinemia and hyperleptinemia) but also increases in markers of cardiac and plasma oxidative stress. Moreover, we observed a higher sensitivity of the heart to injuries induced by ischemia reperfusion, such as an increase in necrosis. We now aim to establish if these late-coming modifications might be induced in a very early state of postnatal development, such as a postnatal programming situation. To achieve this objective, we are now comparing heart gene expression between 24 days old postnatally overfed and normally fed mice. Epigenetic analysis of healthy young men following a control and high-fat overfeeding diet

Project description:Genome-wide DNA methylation profiling of healthy young men following a control and high-fat overfeeding diet using Illumina's Infinium 27k Human DNA methylation Beadchip v. 1.2. DNA methylation profiles were obtained for 27,578 CpG sites in human skeletal muscle. Randomized cross-over desgin, where all subjects receieved both treatments (control and high-fat overfeeding diet). Biopsies were obtained from 23 different individuals amounting to 22 samples following the control diet and 22 samples following the high-fat overfeeding diet (paired n=21). Bisulphite converted DNA from the 44 samples were hybridised to the Illumina Infinium 27k Human Methylation Beadchip.

Effect of Concentration and type of Dietary Fatty Acid on Development of Nonalcoholic Fatty Liver Disease

Project description:The current study was designed to determine if dietary fatty acid concentration and composition affects the development and progression of nonalcoholic fatty liver disease. Male SD rats were overfed diets low (5%) or high (70%) fat diets via total enteral nutrition where the fat source was olive oil (monounsaturated), or corn oil (polyunsaturated). Overfeeding 5% corn oil produced little steatosis relative to feeding 5% olive oil. This was associated with lower fatty acid synthesis and reduced SREBP-c signaling in the 5% corn oil group. Overfeeding 70% fat diets increased steatosis and lead to increased liver necrosis in the 70% corn oil but not olive oil group. Increased injury after feeding polyunsaturated fat diets was linked to peroxidizability of hepatic free fatty acids and triglycerides and appearance of peroxidaized lipid products HETES and HODES previously linked to clinical nonalcoholic steatohepatitis. Male SD rats were overfed diets low (5%) or high (70%) fat diets via total enteral nutrition where the fat source was olive oil (monounsaturated) or corn oil (polyunsaturated).
