Next Generation Sequencing of exosomal microRNA derived from MCU downregulated and control MDA-MB-231 cells.

ABSTRACT: To explore the role of the mitochondrial Ca2+ uniporter (MCU) in exosomal microRNA(miRNA) sorting in MDA-MB-231 cells, we isolated miRNA from the exosomes derived from MCU-downregulated and control MDA-MB-231 cells. We then performed Next Generation Sequencing (NGS) and real-time quantitative polymerase chain reaction (qRT-PCR) to find and validate differetial expressed exosomal miRNAs that regulated by MCU.

Project description:Mitochondrial calcium is an important second-messenger controlling fight-or-flight responses in the heart. The molecular identity of MCU (Mitochondrial Calcium Uniporter) was recently discovered allowing us to test this hypothesis in vivo by expressing a myocardial delimited dominant negative form of MCU. We queried mRNA expression changes induced by dominant negative MCU expression in the heart. Neonatal (< 24 h) and 6 week old hearts were harvested from mice expressing the dominant negative MCU construct or wild-type littermates.

Deep sequencing of exosomal and cellular miRNAs in MDA-MB-231 and MCF-10A cells

Project description:To identify the miRNAs that are differentially expressed and secreted between the MDA-MB-231 metastatic breast cancer cells and the MCF-10A non-cancerous human mammary epithelial cells, we profiled the cellular and exosomal small RNAs (between 17 and 52 nt) isolated from these two cell lines by Solexa deep sequencing. MiRNAs that are significantly different between the two cell lines are identified. RNA was extracted from cultured MDA-MB-231 and MCF-10A cells or purified exosomes secreted by these cells, and subjected to library construction and Solexa deep sequencing.
